This has been achieved in the main by the invention and development of new methodology, parti- larly in that area generally referred to as "genetic en- neering". While this revolution has been taking place in the field of nucleic acids research, the protein chemist has at the same time developed fresh methodology to keep pace with the requirements of present day molecular bi- ogy. Today's molecular biologist can no longer be content with being an expert in one particular area alone. He/she needs to be equally competent in the laboratory at h- dling DNA, RNA, and proteins, moving from one area to another as required by the problem he/she is trying to solve. Although many of the new techniques in molecular biology are relatively easy to master, it is often difficult for a researcher to obtain all the relevant information nec- sary for setting up and successfully applying a new te- nique. Information is of course available in the research l- erature, but this often lacks the depth of description that the new user requires. This requirement for in-depth pr- tical details has become apparent by the considerable - mand for places on our Molecular Biology Workshops held at Hatfield each summer.

Book excerpt: Peptide nucleic acids (PNAs) have now existed for slightly more than ten years, with the interest in and applications of this pseudopeptide DNA mimic steadily increasing during the entire period. PNAs have rapidly attracted the attention of scientists from a diversity of fields ranging from (bio)organic and biophysical chemistry to prebiotic evolution, and from molecular biology to genetic diagnostics and drug development. Many of the applications take advantage of the unique properties of PNA—an uncharged pseudopeptide—that distinguish this DNA mimic from more traditional DNA analogs. Rather than trying to create a comprehensive collection of all published methods and protocols involving PNA—many of which have not yet been validated— I have decided to concentrate on select protocols that are either very well established by several groups around the world, such as PCR-clamping and in situ hybridization, or on new methods that may have broader future impact. Basic methods for PNA oligomer synthesis and analyses have also been included. In the five years which have elapsed since the publication of the classical monograph of Konev, a wealth of new information has ap peared on the emission properties of proteins and nucleic acids. Indeed, since new publications in this area appear to be proliferating in a geometric ratio, this may be the last opportunity to provide a comprehensive summary of the field in a book which is not of prohibitive length. This is what we have attempted to do here. While the orientation of each chapter naturally reflects the interests and point of view of the author, there has been a general effort to present .a critical assessment of existing results and interpretations, rather than a compendium of data with minimal comment. Finally, it should be stressed that the rapid evolution of the subject at the time of writing makes it inevitable that the book will age to some degree over the next few years, although this will occur at differing rates for the various chapters. We can only hope that most of the material in this interim summing-up will prove resistant to the erosion of time and provide a solid foundation for further progress.
