This book was released on 1986 with total page 4 pages. Available in PDF, EPUB and Kindle. Book excerpt: The adaptation of modern chromatographic techniques, especially capillary gas chromatography and high performance liquid chromatography (HPLC), to seawater analysis has lead to new insights into chemical and biological oceanographic processes. In this project the main long-range objective is to extend the role of HPLC by exploring the applicability of microbore HPLC to trace organic analysis of seawater. This technique offers substantially greater sensitivity and separation efficiency than conventional HPLC and, furthermore, the entire effluent of microbore columns may be fed into selective detectors, such as mass spectrometers and flame-base detectors. Initially, the microbore HPLC system will be used for the analylsis of carboxylic acids as part of our ONR-funded project. Other potential marine applications, such as the analysis of amino acids, pigments, photosensitizers, thiols, macromolecules and metal-organic complexes, will also be investigated as opportunities arise. In order to obtain information on the nature of the substances eluting off the microbore column the effluent will be analyzed spectroscopically with an on-the-fly UV-VIS diode array detector. Finally, we will explore potential marine application of laser excitation fluorescence detection in conjunction with microbore HPLC.

Available in PDF, EPUB and Kindle. Book excerpt: In the early 1980s capillary liquid chromatography was being established; it was a period in which only a few research groups published a relatively small number of papers on the subject. In terest has since taken off, and a period of intense development, to which no end is yet in sight, is now upon us. More investiga tors and instrument-making firms are now entering the field. This greater interest has resulted in the rapid appearance of two collec tions [1, 2] and a series of topical reviews [3-6]. However, it could hardly be said that all the problems in this area have been formulated, let alone solved. The preparation of very efficient - open tubular or packed - microbore columns, for example, remains more an art than a science, while the relation ship between radial and longitudinal mass transfer, and the effect of transcolumn velocity profiles on chromatographic efficiency, have been very poorly studied. Indeed, recent publications on these subjects have sometimes, far from clarifying matters, only muddied them further. Many instrument-making firms are trying to unify their equip ment so that it is suitable for microbore, conventional (analytical), and preparative liquid chromatography. This approach has not real ized the full potential of capillary chromatography, and there also remains room for improving the performance of capillary columns.

Scott, Chemistry Department, Birkbeck College, University of London, UK and Chemistry Department, Georgetown University, Washington DC, USA Analytical techniques based on separation processes, such as chromatography and electrophoresis, are finding a growing range of applications in chemical, biochemical and clinical laboratories. The aim of the series is to provide the analyst in these laboratories with well-focused books covering individual techniques and important aspects of the techniques, so that they can be applied more efficiently and effectively to contemporary analytical problems. This book is designed to cover the important subject of liquid chromatography column theory. It provides a lucid account of the principles involved in the separation process which will allow the analyst to understand the function of the column, how to design the optimum column for a specific application and how to use it in the most efficient manner. This reference work will be of value to a broad spectrum of scientists since chromatography is now one of the more popular methods of analysis and is used in such diverse fields as biotechnology, environmental science, forensic science and in pharmaceutical product control.
