Mesoporous silica nanoparticles (MSNs) and mesoporous silica films have been of increasing interest among the scientific community for its use in cargo delivery. Silica provides ease of functionalization, a robust support and biocompatibility. Several methods have been used in order to give the mesoporous silica nanomaterials different qualities that render them a useful material with different characteristics. Among these methods is surface modification by taking advantage of the OH groups on the surface. When a molecule attached to the surface can act as a molecular machine it transforms the nanomaterial to act as delivery system that can be activated upon command. The work covered in this thesis focuses on the development and synthesis of different mesoporous silica materials for the purpose of trapping and releasing cargo molecules. Chapter 2 focuses in the photoactivation of "snap-top" stoppers over the pore openings of mesoporous silica nanoparticles that releases intact cargo molecules from the pores. The on-command release can be stimulated by either one UV photon or two coherent near-IR photons. Two-photon activation is particularly desirable for use in biological systems because it enables good tissue penetration and precise spatial control. Chapter 3 focuses on the design and synthesis of a nano-container consisting of mesoporous silica nanoparticles with the pore openings covered by "snap-top" caps that are opened by near-IR light. A photo transducer molecule that is a reducing agent in an excited electronic state is covalently attached to the system. Near IR two-photon excitation causes intermolecular electron transfer that reduces a disulfide bond holding the cap in place, thus allowing the cargo molecules to escape. The operation of the "snap-top" release mechanism by both one- and two photon is described. This system presents a proof of concept of a near-IR photoredox-induced nanoparticle delivery system that may lead to a new type of photodynamic drug release therapy. Chapter 4 focuses on the attachment of a photoacid molecule on the surface of silica nanoparticles. Upon light irradiation the pKa of the photoacid molecules decreases causing the dissociation of the proton and the acidification of the nanoparticle surface. The local nanoparticle surface acidification was probed using a pH sensitive nanovalve that was attached to MSNs next to a photoacid. The particles were loaded with a fluorescent dye that was contained by the naovalve and released upon acidification of the surrounding environment. The amount of the dye release was measure continuously by detecting its fluorescence. Chapter 5 focuses on the synthesis of materials that utilize the micropatterned structure of a mesoporous silica film to successfully load and release cargo using a thermal sensitive polymer. Films with pore sizes of ~2 and ~5 nm aligned in the pulling direction were synthesized using evaporation induced self-assembly techniques. The pores are exposed using a new method of stamping micropatterns without the use hydrofluoric acid. A well-studied temperature dependent polymer [poly(N-isopropylacrylamide-co-acrylamide)] was grafted onto the surface of these films to act as a temperature activated gatekeeper. Below the lower critical solution temperature (LCST) the polymer is erect and can block the pore openings, trapping cargo inside the pores. When the temperature is above the LCST the polymer collapses and unblocks the pores, allowing cargo to escape. The loading capacities as well as the reusability of these films were studied.

The third project discussed in this thesis utilizes hollow mesoporous silica nanoparticles to crystalize a perchlorate sensor inside the core to be used as "smart sand" for perchlorate detection. The last project focuses on periodic mesoporous organosilica (PMO) nanoparticles and their ability to trap and release cargo molecules. This research demonstrates the versatility of mesoporous silica materials and its potential in both microscopic and macroscopic applications.

The six chapters of this dissertation are presented as follows: 1) the design, synthesis and modification of silica nanoparticles for their use in biology, 2) a light activated, reversible nanovalve assembled on mesoporous silica nanoparticles to achieve a size-selective cargo delivery, 3) biological applications and the delivery of anti-cancer drugs using a pseudorotaxane-based light activated nanovalve, 4) a nanogate machine that is capable of the storage and delivery of both small metal ions and useful organic cargo molecules, 5) biological applications of the nanogate machine in order to deliver calcium ions to cancerous cells to induce cell apoptosis, and 6) thin wax coated microporous silica nanoparticles that are capable of delivering small ions including oxidizers.

It is shown that a high release capacity is necessary to reach a high efficacy ratio, compared with free drug. Chapter 5 discusses the uptake and release capacities of Fe3O4@SiO2 core/shell nanoparticles when modified with a pH-sensitive nanovalves, and its thermally cargo release behavior when surrounding temperature increases or an oscillating magnetic field is applied. In Chapter 6, successful distribution of Fe3O4@SiO2 core/shell nanoparticles in biofilms and on-command release of cargo inside biofilms are shown. Overall, these chapters demonstrate the ability of modifying both the outer surface and interior of MSNs, and their capability to act as a biocompatible controlled release platform that is more effective than equivalent amount of free drug.

Book excerpt: This dissertation makes contributions to the fields of functionalized mesoporous silica nanoparticles (MSNs) designed for magnetic resonance imaging (MRI) contrast enhancement and high-intensity focused ultrasound (HIFU) triggered cargo release. MRI-guided HIFU (MRIgHIFU) has been applied as a therapeutic tool in the clinic, and enhanced MRI contrast will improve the precision and applicability of HIFU therapy. In Chapter 2, the ultrasound responsive MSNs carriers are reviewed. In Chapter 3, the proof-of-concept spotlight technique based on MRIgHIFU and MSNs is introduced. With periodic HIFU modulation, the functionalized MSNs generated reversible MRI T1 relaxivity changes at the 1.5 mm3 focal point. Fourier analysis was used to extract signal changes at the modulation frequency, and a modulation enhancement map spotlights the precise region of interest by increasing contrast almost 100-fold. In Chapter 4, another functionalized MSNs were designed for the spotlight technique, and the spectral analysis was further studied to maximize the contrast enhancement. In particular, a framework was presented to analyze the trade-offs between different parameter choices for the signal processing method. In Chapter 5, the application of the spotlight technique was demonstrated at human body temperature. The MSNs were functionalized with the tailored poly(2-oxazoline)s with a lower critical solution temperature (LCST) at 40 °C, so that it could generate reversible MRI contrast change at human body temperature. The temperature oscillation range was small (3 °C), and the data acquisition was relatively fast (in 100 s), which made it a step closer to the biomedical applications in the clinic. The HIFU-responsive cargo delivery was studied in Chapters 6 and 7. The large pore MSNs (LPMSNs) were studied as the nanocarriers to load a large drug molecule, Docetaxel. With the loading procedure and HIFU parameters tuned, the HIFU-responsive release was demonstrated in a biologically relevant solvent. HIFU-responsive MRI contrast agent delivery was studied using MSNs capped with thermo-responsive polymer. The cargo loading and release conditions were optimized, and the HIFU-responsive cargo release can be monitored by MRI contrast change. The spotlight technique in this dissertation is promising for biomedical applications of precision therapy. The framework on parameter choice and demonstrated MRI contrast enhancement at human body temperature provides the foundation for further clinical translation.
