Although man biochemical changes in brain have been reported to occur as a result of oxygen exposure, these changes are not necessarily the cause of oxygen-induced convulsions. From in vivo studies in mice, inhibition of brain energy metabolism was not found. Furthermore, although oxygen caused an increased oxidation of pyridine nucleotides, the decrease in both NADPH and NADH in cerebral cortex was not-related to the susceptibility fo mice to oxygen convulsions. GABA in brain cortex was decreased by oxygen, however, this decrease did not influence the susceptibility of mice to oxygen convulsions. The most effective CNS protectants seem to be those agents containing a disulfide bridge in their molecular structure, and which can be reduced to thiols. Since thiols are good free radical scavengers, free radicals may be the initial event in provoking the insult to produce oxygen convulsions.

Book excerpt: Significant progress occurred in four areas: (1) Effect of hyperoxia on reverse glycolysis: We concluded that glucose protects Escherichia coli against hyperoxia better than other intermediates later in glycolysis and in the Krebs cycle but not because of a specific inhibition of reverse glycolysis. There was no significant impairment of the three enzymes of reverse glycolysis: fructose-1,6-diphosphatase, phosphoenolpyruvate synthase, or phosphoenolpyruvate carboxykinase. (2) Effect of hyperoxia on synthesis of RNA, DNA and pyridine nucleotide coenzymes: Evidence was obtained for direct inhibition of RNA synthesis in cells poisoned by hyperoxia, while protein synthesis was not directly affected. DNA synthesis is impaired but it is not proven whether this is direct or indirect. NAD and NADH were severly decreased in cells during poisoning by hyperoxia. This decrease was prevented by niacin, but not quinolinate which is consistent with poisoning of quinolinate phosphoribosyltransferase (QPT), as previously reported. PRPP does not protect against oxygen poisoning, PRPP synthetase is not affected by hyperoxia, but PRPP accumulates in oxygen poisoned cells which is in agreement with inhibiton of QPT. (3) Relationship of thiamine to oxygen toxicity.

Formaldehyde is used to produce a wide array of products, particularly building materials; it is emitted from many sources, including power plants, cars, gas and wood stoves, and cigarettes; it is a natural product in come foods; and it is naturally present in the human body as a metabolic intermediate. Much research has been conducted on the health effects of exposure to formaldehyde, including effects on the upper airway, where formaldehyde is deposited when inhaled, and effects on tissues distant from the site of initial contact. The U.S. Environmental Protection Agency (EPA) released noncancer and cancer assessments of formaldehyde for its Intergated Risk Information System (IRIS) in 1990 and 1991, respectively. The agency began reassessing formaldehyde in 1998 and released a draft IRIS assessment in June 2010. Given the complexity of the issues and the knowledge that the assessment will be used as the basis of regulatory decisions, EPA asked the National Research Council (NRC) to conduct an independent scientific review of the draft IRIS assessment. In this report, the Committee to Review EPA's Draft IRIS Assessment of Formaldehyde first addresses some general issues associated with the draft IRIS assessment. The committee next focuses on questions concerning specific aspects of the draft assessment, including derivation of the reference concentrations and the cancer unit risk estimates for formaldehyde.
