Available in PDF, EPUB and Kindle. Book excerpt: This book is a comprehensive review of the detailed molecular mechanisms of and functional crosstalk among the replication, recombination, and repair of DNA (collectively called the "3Rs") and the related processes, with special consciousness of their biological and clinical consequences. The 3Rs are fundamental molecular mechanisms for organisms to maintain and sometimes intentionally alter genetic information. DNA replication, recombination, and repair, individually, have been important subjects of molecular biology since its emergence, but we have recently become aware that the 3Rs are actually much more intimately related to one another than we used to realize. Furthermore, the 3R research fields have been growing even more interdisciplinary, with better understanding of molecular mechanisms underlying other important processes, such as chromosome structures and functions, cell cycle and checkpoints, transcriptional and epigenetic regulation, and so on. This book comprises 7 parts and 21 chapters: Part 1 (Chapters 1–3), DNA Replication; Part 2 (Chapters 4–6), DNA Recombination; Part 3 (Chapters 7–9), DNA Repair; Part 4 (Chapters 10–13), Genome Instability and Mutagenesis; Part 5 (Chapters 14–15), Chromosome Dynamics and Functions; Part 6 (Chapters 16–18), Cell Cycle and Checkpoints; Part 7 (Chapters 19–21), Interplay with Transcription and Epigenetic Regulation. This volume should attract the great interest of graduate students, postdoctoral fellows, and senior scientists in broad research fields of basic molecular biology, not only the core 3Rs, but also the various related fields (chromosome, cell cycle, transcription, epigenetics, and similar areas). Additionally, researchers in neurological sciences, developmental biology, immunology, evolutionary biology, and many other fields will find this book valuable.

Book excerpt: Mechanistic Studies of DNA Replication and Genetic Recombination emerged from a symposium on DNA replication and genetic recombination held from March 16-21, 1980 in Keystone, Colorado. The event featured 30 plenary session talks, 13 workshop discussion groups, and the 210 poster sessions. The studies described in this book are paving the way for the elucidation of other basic genetic mechanisms, including ""new"" areas in molecular genetics such as those of eukaryotic gene expression and the transposition of mobile genetic elements. This book is divided into 10 parts: summaries of workshop discussion groups (Part I); studies on eukaryotic model systems for DNA replication (Part II); studies on bacterial replication origins (Part III); studies on replication origins of bacterial phages and plasmids (Part IV); studies on eukaryotic replication origins (Part V); studies on prokaryotic replication enzymology (Part VI); studies on eukaryotic replication enzymology (Part VII); studies on the fidelity of DNA replication (Part VIII); studies on DNA topoisomerases (Part IX); and studies of genetic recombination mechanisms (Part X).

Available in PDF, EPUB and Kindle. Book excerpt: This book contains the papers presented at the Twenty-Seventh Annual Biology Division Research Conference which was held April 1-4, 1974 in Gatlinburg, Tennessee. The topic of the symposium was Mechanisms in Recombination and it follows by exactly twenty years the previous Gatlinburg Symposium on Genetic Recombination. During this interval, and the preceding years as well, the process of recombination has remained a central and tantalizing problem for geneticists. The subject assumes added significance with the recent appeal by a committee of leading scientists for a moratorium on the construction of certain types of recombinant molecules. That autonomously replicating molecules linking portions of pro karyotic and eukaryotic DNA can now be produced in vitro attests to the technical advances that have taken place in this field. Nevertheless, the details underlying the process in vivo continue to be elusive. This symposium brought together individuals studying recombi nation in organisms as widely separated as bacteriophage and mammals and using disciplinary approaches of comparable diversity. Conse quently the present volume summarizes much of current strategies and concepts concerning the subject. The meeting was sponsored by the Biology Division of the Oak Ridge National Laboratory (operated by the Union Carbide Corporation for the U. S. Atomic Energy Commission) with the support and encour agement of its director, H. I. Adler. The organizing committee was chaired by J. K. Setlow and included R. F. Grell, R. D. Hotchkiss and E. Volkin. Special thanks are due to the speakers, to I. R.

Book excerpt: "DNA to DNA transitions are spectacular events involving phenomenal biochemical and topological complexity and astounding requisites for precision and control. Molecular Mechanisms in DNA Replication and Recombination offers a detailed understanding of the molecular mechanisms of DNA replication and recombination and their regulation. The book represents a thorough picture of the various topological forms that the DNA double helix can assume, and the way in which these forms can recognize and interact with their cognate bindings proteins and enzymes. This volume features the work of X-ray crystallographers, structural chemists, and nucleic acid enzymologists to promote the cross-fertilization of ideas, experimental approaches, and techniques. It assesses major advances in the field such as: the ways in which replication od duplex DNA genomes, both prokaryotic and eukaryotic are initiated, the replication potential of SV40 (the state of phosphorylation of large T antigen and presumably its host cell analogue) and ori C (the functional interaction of the DNA protein with phospholipids, and presumably the cell membrane), the structure and dynamics of DNA polymerase action, and the molecular mechanism of site specific and homologous recombination. Molecular Mechanisms in DNA Replication and Recombination is of importance to scientists involved in nucleic acid research, molecular biology, enzymology, and celluar biochemistry." --

Book excerpt: Replicating and Repairing the Genome provides a concise overview of the fields of DNA replication and repair. The book is particularly appropriate for graduate students and advanced undergraduates, and scientists entering the field or working in related fields. The breadth of information regarding DNA replication and repair is vast and often difficult to absorb, with terminology that differs between experimental systems and with complex interconnections of these processes with other cellular pathways. This book provides simple conceptual descriptions of replication and repair pathways using mostly generic protein names, laying out the logic for how the pathways function and highlighting fascinating aspects of the underlying biochemical mechanisms and biology. The book incorporates extensive and informative diagrams and figures, as well as descriptions of a number of carefully chosen experiments that had major influences in the field. The process of DNA replication is explained progressively by starting with the system of a simple bacterial virus that uses only a few proteins, followed by the well-understood bacterial (E coli) system, and then culminating with the more complex eukaryotic systems. In the second half of the book, individual chapters cover key areas of DNA repair — postreplication repair of mismatches and incorporated ribonucleotides, direct damage reversal, excision repair, and DNA break repair, as well as the related areas of DNA damage tolerance (including translesion DNA polymerases) and DNA damage responses. The book closes with chapters that describe the huge impact of DNA replication and repair on aspects of human health and on modern biotechnology.

Book excerpt: This book reviews the latest trends and future directions of DNA replication research. The contents reflect upon the principles that have been established through the genetic and enzymatic studies of bacterial, viral, and cellular replication during the past decades. The book begins with a historical overview of the studies on eukaryotic DNA replication by Professor Thomas Kelly, a pioneer of the field. The following chapters include genome-wide studies of replication origins and initiation factor binding, as well as the timing of DNA replications, mechanisms of initiation, DNA chain elongation and termination of DNA replication, the structural basis of functions of protein complexes responsible for execution of DNA replication, cell cycle-dependent regulation of DNA replication, the nature of replication stress and cells’ strategy to deal with the stress, and finally how all these phenomena are interconnected to genome instability and development of various diseases. By reviewing the existing concepts ranging from the old principles to the newest ideas, the book gives readers an opportunity to learn how the classical replication principles are now being modified and new concepts are being generated to explain how genome DNA replication is achieved with such high adaptability and plasticity. With the development of new methods including cryoelectron microscopy analyses of huge protein complexes, single molecular analyses of initiation and elongation of DNA replication, and total reconstitution of eukaryotic DNA replication with purified factors, the field is enjoying one of its most exciting moments, and this highly timely book conveys that excitement to all interested readers.

Book excerpt: Bioinformatics, which can be defined as the application of computer science and information technology to the field of biology and medicine, has been rapidly developing over the past few decades. It generates new knowledge as well as the computational tools to create that knowledge. Understanding the basic processes in living organisms is therefore indispensable for bioinformaticians. This book addresses beginners in molecular biology, especially computer scientists who would like to work as bioinformaticians. It presents basic processes in living organisms in a condensed manner. Additionally, principles of several high-throughput technologies in molecular biology, which need the assistance of bioinformaticians, are explained from a biological point of view. It is structured in the following 9 chapters: cells and viruses; protein structure and function; nucleic acids; DNA replication, mutations, and repair; transcription and posttranscriptional processes; synthesis and posttranslational modifications of proteins; cell division; cell signaling pathways; and high-throughput technologies in molecular biology.

Book excerpt: DNA Repair Mechanisms is an account of the proceedings at a major international conference on DNA Repair Mechanisms held at Keystone, Colorado on February 1978. The conference discusses through plenary sessions the overall standpoint of DNA repair. The papers presented and other important documents, such as short summaries by the workshop session conveners, comprise this book. The compilation describes the opposing views, those that agree and dispute about certain topic areas. This book, divided into 15 parts, is arranged according to the proceedings in the conference. The plenary sessions are grouped with the related workshop and poster manuscripts. The first two parts generally tackle repair in terms of its identification and quantification, as well as the models, systems, and perspectives it utilizes. The following parts discuss the various types of repair including base excision, nucleotide excision repair in bacteria, excision repair in mammalian cells, inducible/error-prone repair in prokaryotes, and strand break repair in mammalian cells among others. This reference material looks into the replicative bypass mechanisms in mammalian cells, viral probes, and hereditary repair defects. It explains repair deficiency and human disease, as well as mutagenesis and carcinogenesis. The last part of this book deals with the consequences and effects of DNA repair. This volume is a helpful source of reference for students, teachers, scientists, and researchers in the different fields of genetics, radiology, biochemistry, and environmental biology.
