Book excerpt: Sheep, much like humans, have a propensity to overeat if presented with ad libitum feed, thus allowing researchers an opportunity to study the effects of maternal obesity on fetal outcomes. In this study, the concentrations of fatty acids from maternal and fetal tissues taken at day 135 of gestation were analyzed and compared using Gas Liquid Chromatography (GLC). It was hypothesized that maternal diet would affect fetal tissue fatty acid concentrations. In maternal liver, there is a greater concentration of FA fatty acids in maternal liver when compared to fetal liver and this finding could suggest that there iss less fetal liver membrane development in maternal liver than in fetal liver; FA are key components of cell membranes. It is possible that placental transfer of maternal FA could be regulating the availability of maternal FA fatty acids for fetal liver development. For cardiac tissue, maternal diet had no effect on fetal fatty acid concentrations. However, 18:2 seems to have been preferentially stored in maternal cardiac tissues and not transported across the placenta in appreciable amounts. Maternal plasma is known to contain high levels of 18:2 while fetal plasma contains a low proportion of this FA (Nobel, et al. 1982); thus, suggesting that 18:2 does not cross the placenta in appreciable amounts. Skeletal muscle (Longissimus dorsi) was only obtained from fetuses and not dams and the results for this tissue indicate that maternal diet did not affect fatty acids in this tissue at day 135 of gestation. Book excerpt: The developing fetus requires substantial amounts of fatty acids to support rapid cellular growth and activity, and among these the omega-3 (n-3) polyunsaturated fatty acids (PUFAs) are crucial. Maternal dietary supplementation with n-3 PUFAs during pregnancy has been shown to increase gestation length and reduce the risk of pregnancy complications, although the precise mechanisms governing these effects remain uncertain. Omega-3 PUFAs are involved in several physiological pathways which could account for these effects, including anti-oxidative, anti-inflammatory and pro-resolving pathways. Because several placental disorders are associated with heightened placental oxidative stress and inflammation, there is considerable interest in the therapeutic potential of dietary n-3 PUFAs. Therefore, the overall aim of this thesis was to investigate the impact of maternal dietary supplementation with n-3 PUFAs on placental growth and function, particularly in relation to oxidative and inflammatory status, in the rat. Placental oxidative stress plays a key role in the pathophysiology of several placental-related disorders, including intrauterine growth restriction and preeclampsia. Oxidative stress occurs when accumulation of reactive oxygen species (ROS) damages DNA, proteins and lipids, an outcome normally limited by antioxidant defences. Omega-3 PUFAs have been shown to exert anti-oxidative effects in several tissues. As such, the initial study presented in this thesis tested the hypothesis that n-3 PUFA dietary supplementation would reduce placental oxidative damage, and consequently enhance fetal and placental growth. In addition, whether diet-induced changes in maternal plasma fatty acid profiles were associated with comparable changes in placental and fetal tissues was investigated. Rats were fed either a standard (Std) or high n-3 PUFA (Hn3) semipure diet from day 1 of pregnancy, and tissues collected on days 17 or 22 (term = day 23).

Book excerpt: Dietary Risk Assessment in the WIC Program reviews methods used to determine dietary risk based on failure to meet Dietary Guidelines for applicants to the Special Supplemental Nutrition Program for Women, Infants, and Children (WIC). Applicants to the WIC program must be at nutritional risk to be eligible for program benefits. Although "dietary risk" is only one of five nutrition risk categories, it is the category most commonly reported among WIC applicants. This book documents that nearly all low-income women in the childbearing years and children 2 years and over are at risk because their diets fail to meet the recommended numbers of servings of the food guide pyramid. The committee recommends that all women and children (ages 2-4 years) who meet the eligibility requirements based on income, categorical and residency status also be presumed to meet the requirement of nutrition risk. By presuming that all who meet the categorical and income eligibility requirements are at dietary risk, WIC retains its potential for preventing and correcting nutrition-related problems while avoiding serious misclassification errors that could lead to denial of services for eligible individuals.
