Book excerpt: Scientific progress with respect to the important role of various lipids in myocardial function and the dearrangements of lipid metabolism underlying ischaemic heart disease(s) has been con siderable in recent years. In 1986 alone, an overall number ofl 044 full papers covering the topics "Lipids-Heart-Heart Disease", have been published in a variety of biochemistry, physiology, pharmacology and cardiology journals (source: Index Medicus). They have broadened our in sight into the molecular basis of myocardial lipidology and the lipido-chemical basis of car diology in health and disease, and have narrowed the gap between promising pharmacological intervention in experimental animals and clinical treatment of patients suffering from an ischaemic heart. Furthermore, they illustrate the fundamental significance of the product of the union between basic and clinical science. The rapid development of knowledge in the "Lipids-Heart-Heart Disease" triad prompted us to organize the International Symposium on Lipid Metabolism in the Normoxic and lschaemic Heart, held in Rotterdam (The Netherlands) on September 22 and 23, 1986. This meeting was a coproject of the Department of Biochemistry I (Medical Faculty, Erasmus University Rotter dam) and the Department of Physiology (Medical Faculty, University of Limburg) and was held under the auspices of the GerritJan Mulder Foundation (named after a famous Rotterdam physi cian and chemist who lived from 1802-1880) whom we thank for their co-operation. The present volume is a compendium of invited papers and contributions selected from the posters presented during the symposium.

Book excerpt: During recent decades, bewildering progress has occurred in the field of Molecular and Cellular Biochemistry. Progress has been extraordinarily rapid primarily because of the challenge for finding solutions to a wide variety of diseases and the availability of new techniques for monitoring biochemical processes. This has resulted in a voluminous and complex literature in the field of biochemical medicine so that there is a clear need for the synthesis and analysis of the continuing expansion of valuable data. It was thus considered appropriate to initiate a new series of monographs, each dedicated to a specialized area of investigation, encompassing molecular and cellular processes in health and disease. Most of the biochemical scientists have devoted their energies in understanding the fundamentals of biochemistry and indeed impressive advances have been made in the past. However, the full potential for explanation has been hampered by the concept of universality of biochemical reactions occurring in the cell. In view of the fact that each organ in the body performs a distinct function, it is now beginning to be realized that each cell type is unique in its need to survive and perform its specific function. Accordingly, the aspect of individualty is receiving increased attention for revealing new avenues in the study of pathophysiology of cellular abnormalities.

Book excerpt: Diabetes is an autoimmune, inflammatory disease affecting many different organ systems and exhibiting both primary and secondary defects. Because diabetes affects a wide range of cellular systems, a multidisciplinary effort has been mounted over the past several decades using a wide range of investigative techniques and methodologies in order to identify molecular mechanisms responsible for cellular dysfunction. Because primary defects at various levels of sub-cellular signaling, intracellular calcium handling, protein expression and energy regulation are often a primary consequence of diabetes. This volume is a compilation of new multidisciplinary research that will broaden our current understanding of diabetes and cardiovascular disease as well as provide the basis for the development of novel therapeutic interventions.

Available in PDF, EPUB and Kindle. Book excerpt: The heart has a very high energy demand but very little energy reserves. In order to sustain contractile function, the heart has to continually produce a large amount of ATP. The heart utilizes free fatty acids mainly and carbohydrates to some extent as substrates for making energy and any change in this energy supply can seriously compromise cardiac function. It has emerged that alterations in cardiac energy metabolism are a major contributor to the development of a number of different forms of heart disease. It is also now known that optimizing energy metabolism in the heart is a viable and important approach to treating various forms of heart disease. Cardiac Energy Metabolism in Health and Disease describes the research advances that have been made in understanding what controls cardiac energy metabolism at molecular, transcriptional and physiological levels. It also describes how alterations in energy metabolism contribute to the development of heart dysfunction and how optimization of energy metabolism can be used to treat heart disease. The topics covered include a discussion of the effects of myocardial ischemia, diabetes, obesity, hypertrophy, heart failure, and genetic disorders of mitochondrial oxidative metabolism on cardiac energetics. The treatment of heart disease by optimizing energy metabolism is also discussed, which includes increasing overall energy production as well as increasing the efficiency of energy production and switching energy substrate preference of the heart. This book will be a valuable source of information to graduate students, postdoctoral fellows, and investigators in the field of experimental cardiology as well as biochemists, physiologists, pharmacologists, cardiologists, cardiovascular surgeons and other health professionals.

Available in PDF, EPUB and Kindle. Book excerpt: This presentation describes various aspects of the regulation of tissue oxygenation, including the roles of the circulatory system, respiratory system, and blood, the carrier of oxygen within these components of the cardiorespiratory system. The respiratory system takes oxygen from the atmosphere and transports it by diffusion from the air in the alveoli to the blood flowing through the pulmonary capillaries. The cardiovascular system then moves the oxygenated blood from the heart to the microcirculation of the various organs by convection, where oxygen is released from hemoglobin in the red blood cells and moves to the parenchymal cells of each tissue by diffusion. Oxygen that has diffused into cells is then utilized in the mitochondria to produce adenosine triphosphate (ATP), the energy currency of all cells. The mitochondria are able to produce ATP until the oxygen tension or PO2 on the cell surface falls to a critical level of about 4–5 mm Hg. Thus, in order to meet the energetic needs of cells, it is important to maintain a continuous supply of oxygen to the mitochondria at or above the critical PO2 . In order to accomplish this desired outcome, the cardiorespiratory system, including the blood, must be capable of regulation to ensure survival of all tissues under a wide range of circumstances. The purpose of this presentation is to provide basic information about the operation and regulation of the cardiovascular and respiratory systems, as well as the properties of the blood and parenchymal cells, so that a fundamental understanding of the regulation of tissue oxygenation is achieved.

Available in PDF, EPUB and Kindle. Book excerpt: ATP plays a central role in the two leading causes of cardiac morbidity and mortality in the western world: ischemia and heart failure. We are in our infancy applying what is known about biology and chemistry of ATP toward developing effective therapies for these diseases. In this volume, the current understanding of the chemistry and biology of ATP specifically in the cardiomyocyte is presented. New insights into ATP have been gleaned using biophysical techniques allowing dynamic measurement of chemical events in the intact beating heart and using new animal models in which cardiac proteins are either over expressed, deleted or harbor specific mutations. This book provides a summary of the basic understanding and includes illustrations of why ATP and the Heart is important to both the clinician and scientist.

Book excerpt: The Advanced Study Institute on "Structure, Biogenesis and Dynamics of Biological Membranes, held in Cargese from June 14-26, 1993, has been dealing with four major topics in membrane biochemistry today: lipid dynamics and lipid-protein interactions, protein translocation and insertion, intracellular traffic aud protein structure and folding. The lecturers discussed these topics starting from several disciplines, including biochemistry, cell biology, genetics, and biophysics. This wayan interdisciplinary and very inte~sting view on biological membrane systems was obtained. At first an extensive overview of -mainly biophysical -techniques which can be used to study dynamic processes in membranes was presented. Sophisticated approaches such as ESR and NMR have been applied succesfully to unravel details of specific lipid-protein interactions. x ray analysis provides detailed structural information of several proteins and the possible implications for protein functions. Information obtained this way is complemented by studies on mechanisms and kinetics of protein folding. The latter information is indispensable when discussing protein translocation and insertion: proces:;es in which folding and unfolding play essential roles. Extensive insight was offered in the complicated machinery of phospholipid biosynthesis. In particular, the application of sophisticated genetic techniques has allowed a better understanding of the mechanisms regulating the synthetic machinery and detailed studies on a variety of mutants, lacking one or more of the essential enzymes, have resulted in the beginning of a bL!:

Book excerpt: Over the past three decades, impressive progress in the field of pathogenesis, prevention and therapy of ischemic heart disease has resulted in a marked decline in mortality in the Western World. However, the incidence of this devastating disease is on the rise in developing countries. The Ischemic Heart is based upon a recent symposium in Tokyo on the subject. This volume is organized into two sections: (i) Pathophysiologic Mechanisms of Ischemia-Reperfusion Injury and (ii) Preconditioning and Protection of Ischemia-Reperfusion Injury, and contains up-to-date information concerning the current concepts of ischemia-reperfusion injury, the sequence of events resulting in the loss of contractile dysfunction, and mechanisms of cardioprotection by several drugs as well as the role of ischemic preconditioning in attenuating problems associated with ischemia-reperfusion injury.
