Book excerpt: Scientific progress with respect to the important role of various lipids in myocardial function and the dearrangements of lipid metabolism underlying ischaemic heart disease(s) has been con siderable in recent years. In 1986 alone, an overall number ofl 044 full papers covering the topics "Lipids-Heart-Heart Disease", have been published in a variety of biochemistry, physiology, pharmacology and cardiology journals (source: Index Medicus). They have broadened our in sight into the molecular basis of myocardial lipidology and the lipido-chemical basis of car diology in health and disease, and have narrowed the gap between promising pharmacological intervention in experimental animals and clinical treatment of patients suffering from an ischaemic heart. Furthermore, they illustrate the fundamental significance of the product of the union between basic and clinical science. The rapid development of knowledge in the "Lipids-Heart-Heart Disease" triad prompted us to organize the International Symposium on Lipid Metabolism in the Normoxic and lschaemic Heart, held in Rotterdam (The Netherlands) on September 22 and 23, 1986. This meeting was a coproject of the Department of Biochemistry I (Medical Faculty, Erasmus University Rotter dam) and the Department of Physiology (Medical Faculty, University of Limburg) and was held under the auspices of the GerritJan Mulder Foundation (named after a famous Rotterdam physi cian and chemist who lived from 1802-1880) whom we thank for their co-operation. The present volume is a compendium of invited papers and contributions selected from the posters presented during the symposium.

In view of the fact that each organ in the body performs a distinct function, it is now beginning to be realized that each cell type is unique in its need to survive and perform its specific function. Accordingly, the aspect of individualty is receiving increased attention for revealing new avenues in the study of pathophysiology of cellular abnormalities.

Book excerpt: Over the past years, the thrust of research in cardiology has been toward an understanding of the engineering of the heart as a pump that transports blood to the various organs of the body. More recently, the fields of biochemistry and biophysics have come to influence heart research. The modern cardiologist can no longer pretend to understand, for example, what is happening to the patient with myocardial infarction or ischemia without understanding the principles of molecular biology. The structure and function of the heart are therefore central themes of cardiological research and practice, which incorporate knowledge and discoveries from diverse disciplines. The importance of lipid metabolism in the myocardium has become clearly understood. In the well-oxygenated heart, fatty acids are the preferred substrates. The fact that the heart derives most of its energy from the oxidation of lipids, which represent the larg est energy store of the body, is logical for an organ that must work throughout our lifetime. There are, however, several lines of evidence that during ischemia, lipids may. be harmful to the heart. High levels of free fatty acids in the serum have been suggested to play a role in causing life-threatening arryhthmias and damage in the ischemic heart. The molecular basis for these effects remains poorly understood, and several possible mechanisms for these harm ful effects have been suggested.

The lecturers discussed these topics starting from several disciplines, including biochemistry, cell biology, genetics, and biophysics. This wayan interdisciplinary and very inte~sting view on biological membrane systems was obtained. At first an extensive overview of -mainly biophysical -techniques which can be used to study dynamic processes in membranes was presented. Sophisticated approaches such as ESR and NMR have been applied succesfully to unravel details of specific lipid-protein interactions. x ray analysis provides detailed structural information of several proteins and the possible implications for protein functions. Information obtained this way is complemented by studies on mechanisms and kinetics of protein folding. The latter information is indispensable when discussing protein translocation and insertion: proces:;es in which folding and unfolding play essential roles. Extensive insight was offered in the complicated machinery of phospholipid biosynthesis. In particular, the application of sophisticated genetic techniques has allowed a better understanding of the mechanisms regulating the synthetic machinery and detailed studies on a variety of mutants, lacking one or more of the essential enzymes, have resulted in the beginning of a bL!:
