Book excerpt: vi as did non-appreciation that % values for bought-in solutions (notably ammonia) may be on a weight basis, not made evident by the manufacturer. Notwithstanding the shortcomings or lateness of some texts, authors are thanked for compiling them amidst other pressures. Elsevier and the American Chemical Society are also thanked, for Figures now reproduced with source acknowledgement. This Editor has generally respected authors' phrasing, whilst shuddering when the term 'incubate' is encountered in a 0° context. He remains a 'diehard' in certain respects, notably in favouring 'M' rather than 'mol/I', and a wt./ml basis for drug concentrations in test samples; he regards 'mmol/l' as a fatuous fashion. Concerning infelicitous abbreviations, a distinction is made between electron capture (detector context; 'ECD') and electrochemical ('EC', never 'ECD'); the hallowed GC term 'FID' means free induction decay to NMR practi tione:ts, who may pardon the term 'Fid' as introduced editorially. The convention for ,0C' throughout the book is '0'. Undefined but well-known abbreviations include GC, HPLC and TLC. MS (mass spectrometry), NPD (nitrogen-phosphorus detector), tr (retention time) and RIA (radioimmunoassay) are usually defined in the article concerned, as are the HPLC modes NP (normal-/straight phase) and RP (reversed-phase; C-lS and ODS are synonymous), and i.s.

Book excerpt: "With its high efficiency and reproducibility, open tubular capillary electrochromatography (OTCEC) is beginning to prove itself as a promising method for the separation of proteins, peptides and pharmaceuticals. OTCEC, when coupled to an absorbance-based detector, suffers from poor sensitivity and high concentration limits of detection. However, laser induced fluorescence coupled to CEC has shown picomolar limits of detections for some compounds.2 This thesis describes the coupling of OTCEC to a laser induced fluorescence (LIF) detector to optimize both the separation and detection aspects of protein and amino acid analysis. OTCEC capillaries were etched and silanized, and the 4,4' cyanopentoxy biphenyl phase was attached through a radical-initiated process. Such biphenyl phases have shown unique dependence on temperature and mobile phase composition and demonstrated higher selectivity for certain compounds3 than bare silica capillaries. Proteins and amino acids were modified using pre-column derivatization formats with two different fluorescein-based dyes (fluorescein isothiocyanate, FITC) and Atto-tag FQ (3-(2-furoyl)quinoline-2-carboxaldehyde). These conditions were varied to attempt consistent labeling conditions with varying protein size. Optimizations of the analyses were achieved by modifying conditions such as background electrolyte, pH, voltage, and percent organic modifiers. These modified conditions were used to achieve the highest resolution on protein and amino acid derivatives. Results determined that high sensitivity and efficiency are achieved when LIF detection is coupled to cyano pentoxy biphenyl coated capillaries using optimal conditions. These conditions include 1) the use of Atto-tag FQ derivatization and 2) an 80:20 acetonitrile: 50 mM phosphate buffer (pH 2.00) background electrolyte with a positive electrical current applied."--Abstract from author supplied metadata.

Available in PDF, EPUB and Kindle. Book excerpt: This report summarizes the progress and accomplishments of this research project from September 1, 1989 to February 28, 1993. During this period, we have accomplished all of the primary scientific objectives of the research proposal: (1) constructed and evaluated a laser-induced fluorescence detection system that allows direct examination of the chromatographic column, (2) examined nonequilibrium processes that occur upon solute injection and elution, (3) examined solute retention in liquid chromatography as a function of temperature and pressure, (4) examined solute zone dispersion in liquid chromatography as a function of temperature and pressure, and (5) developed appropriate theoretical models to describe these phenomena. In each of these studies, substantial knowledge has been gained of the fundamental processes that are responsible for chromatographic separations. In addition to these primary research objectives, we have made significant progress in three related areas: (1) examined pyrene as a fluorescent polarity probe insupercritical fluids and liquids as a function of temperature and pressure, (2) developed methods for the class-selective identification of polynuclear aromatic hydrocarbons in coal-derived fluids by microcolumn liquid chromatography with fluorescence quenching detection, and (3) developed methods for the determination of saturated and unsaturated (including omega-3) fatty acids in fish oil extracts by microcolumn liquid chromatography with laser-induced fluorescence detection. In these studies, the advanced separation and detection techniques developed in our laboratory are applied to practical problems of environmental and biomedical significance.
