The toxicity associated with the lipid soluble agents has been reduced and their effectiveness against radiation has been increased. The water soluble agents have been expanded to include new effective functional groups, thereby increasing the flexibility in this part of the program. For the first time in this laboratory, monkeys have been protected against fully lethal doses of radiation. The chronic studies preliminary to administering these compounds to man have been initiated. The progress of the last year is such that the staff is confident that with sufficient effort, these agents can be made effective without appreciable side effects.

Agents have been found which have an increased therapeutic index; for the first time since the inception of the program a com pound has been found which exhibits some protec tive activity at 1/8th of the maximum tolerated level. Continued success is to be found in another series of antiradiation agents which are effective in very small amounts. These compounds which formerly were not effective by mouth have now been modified so that they may be given orally. Histamine release which was an important toxic side effect has been greatly reduced. The continued success in drug development convinces us that an antiradiation pill offering a dose reduction of approximately two to one can be developed for man and will not possess appre ciable side effects. The symposium focussed on cytometric approaches to biological dosimetry, and was, to the best of our knowledge, the first meeting on this subject ever held. There was strong encouragement from the 75 attendees and from others to publish a proceedings of the symposium. Hence this book, containing 30 of the 36 presentations, has been assembled. Dosimetry, the accurate and systematic determination of doses, usually refers to grams of substance administered or rads of ionization or some such measure of exposure of a patient, a victim or an experimental system. The term also can be used to describe the quantity of an ultimate, active agent as delivered to the appropriate target material within a biological system. Thus, for mutagens, one can speak of DNA dosimetry, meaning the number of adducts produced in the DNA of target cells such as bone-mar row stem cells or spermatogonia.
