Book excerpt: The triplet state of chlorophyll has been utilized as a nondestructive probe into the structural and dynamical nature of the photosynthetic apparatus, both in vitro and in isolated in vivo subunits. During the past year, using zero-field triplet state optically detected magnetic resonance spectroscopy, the research program has examined triplet energy transfer between photosynthetic pigments in solution; has isolated and studied subchloroplast particles enriched with photo-system I and the light harvesting chlorophyll protein (antenna) system; and has examined model chlorophyll-protein systems, specifically chlorophyll protein 668 and the reconstituted myoglobins. The overall aim of the research program has been to detail the physical features and interactions of the photosynthetic pigment systems through examination of structural subunits of the photosynthetic apparatus and model systems which represent them.

Book excerpt: The photoexcited triplet state properties of chlorophyll dimer systems in vitro were investigated by zero-field optically detected magnetic resonance (ODMR) spectroscopy at 2°K. Measurements of the triplet state zero-field splittings, overall triplet lifetimes, and individual spin sublevel intersystem crossing rate constants were obtained for solutions of chlorophyll a and zinc-substituted chlorophyll a and for the covalently linked dimeric derivative of pyrochlorophyllide a. The triplet state properties found for these systems are interpreted within the framework of the triplet exciton model to assess the applicability of the exciton approach in determining the geometry of chlorophyll aggregate systems and to evaluate structural features of chlorophyll systems proposed as models for in vivo chlorophyll.

Book excerpt: Triplet state properties of solutions containing chlorophyll monomers and dimers have been determined by optically detected magnetic resonance (ODMR) spectroscopy. Using a simple exciton formalism, the triplet state zero-field splittings and intersystem crossing rate constants found for the chlorophyll dimers and for the isolated chlorophyll molecule are utilized to determine the orientation of the pigment molecules in the dimer. The triplet state results indicate that water-linked chlorophyll dimers in vitro form single plane-parallel structures. Similar calculations using monomeric measurements by ODMR on bacteriochlorophyll and zero-field splittings for in vivo preparations of photosynthetic bacteria indicate that the dimer structures in vivo may be more complex than those observed in solution.

Book excerpt: During the initial two years of our ERDA-supported research program we have attempted to define and understand the mechanisms active in the intersystem crossing processes of large polycyclic hydrocarbons and chlorophyll. To study these excited states energy conversion processes we have utilized optical detection of magnetic resonance spectroscopy, as developed in our laboratory, which is uniquely applicable to these molecular systems. Our results on polycyclic hydrocarbons have shown that an understanding of triplet state radiationless transitions in planar aromatic molecules can be obtained from a consideration of the symmetry-selected spin-orbit interactions and positions of available mixing states; this work provides a predictive model of triplet state nonradiative energy conversion with general applicability to aromatic systems. This treatment has been applied successfully to chlorophyll triplet state dynamics and provides a basis for the utilization of the triplet state of chlorophyll as a (nondestructive) spin probe into the makeup of more complex chlorophyll-containing systems in vitro and in vivo.

An understanding of the mechanisms involved in the coupling of these reactions in more complex systems to obtain a vectorial storage of energy across an insulating barrier (i.e. a lipid bilayer) is developing. The information must ultimately prove useful in the long-range goal of developing practical means of solar energy utilization. (ERB).
