Book excerpt: Brassica napus L. is an economically important oilseed species cultivated across Western Canada. Hybrid B. napus cultivars compose the majority of the market due to their seed yield and agronomic quality. It is important to attempt to predict high-yielding parental combinations in order to conserve resources during experimental hybrid evaluation. Genetic distance between parents has been implicated in producing high-yielding hybrids and is used as one criteria for determining parental combinations.In the current study, the genetic distance between high erucic acid rapeseed (HEAR) genotypes of B. napus was established using both phenotypic and genotypic criteria. Phenotypic criteria took the form of nine agronomic and seed quality traits gathered from 318 distinct B. napus genotypes over the 2013 and 2014 field seasons in Southern Manitoba. Genotypic criteria took the form of either 291,782 SNP markers identified in 231 distinct B. napus genotypes using genotyping-by-sequencing (GBS) or 230 polymorphic sequence-related amplified polymoprhism (SRAP) markers identified in 160 B. napus genotypes. The genetic distance between available pollinators and a single male-sterile female was established using each set of criteria in an attempt to correlate genetic distance with hybrid yield. Regression analysis was conducted with yield data from hybrid genotypes gathered from 37 field sites from 2011-2014. Using the phenotypic-derived genetic distance, a significant correlation between genetic distance and hybrid yield was uncovered explaining either 22 % or 42 % of the variation in hybrid yield depending upon whether hybrids were grown at three or more, or five or more sites in the analysis, respectively. No significant link was found between GBS or SRAP-derived genetic distance and hybrid yield. These results provide evidence that that phenotypic criteria can be used to establish genetic distance with utility in the selection of high-yielding hybrid genotypes.

However, parental combinations that exhibit high heterotic gains can be difficult to obtain if no prior genetic architecture is known. The current research focuses on heterotic gene pool development using genetic distance and multivariate cluster analysis. Seventy-nine inbred accessions were grouped using three different methods including; 1) phenotypic classification based on 20 qualitative and quantitative traits; 2) Sequence related amplified polymorphisms (SRAP) using 29 forward and reverse primer combinations; 3) genotyping-by-sequencing (GBS) using 80,005 single nucleotide polymorphisms. Both genotypic methods (SRAP and GBS) were compared to each other, and hierarchical clustering produced similar results with 68% homology between the two methods. Heterotic cluster accuracy was investigated between the two genotypic methods through hybrid performance. Forty-four hybrid combinations along with parental cultivars were grown at one location in Winnipeg, MB with three replicates in a randomized complete block design. Book excerpt: In oilseed rape (Brassica napus L.), the use of a pollination control system is essential for hybrid seed production. The Ogu-INRA CMS system is a cytological variant of the radish- (Raphanus sativus L.) derived Ogu CMS system introduced through interspecific introgression into B. napus. In this system, there are three different lines: A-lines are male sterile, B-lines are fertile maintainer lines for the respective A-lines and R-lines (restorer) are male fertility restorer lines. The nature of the introgression produces a variety of issues within the restorer genotypes. These genotypes tend to have poor agronomic performance and high levels of glucosinolates. Apart from the phenotypic caveats, there is limited genomic data regarding the specific size, location and sequence of the introgression. The work presented in this thesis addresses both the genotypic and phenotypic issues present in B. napus restorer genotypes. Homozygous restorer lines are required to produce a fertile F1 generation making a marker for the Rfo gene a requirement in any breeding program. In the first project, an allele-specific Rfo marker was developed using KASP assays. This was achieved for one restorer population and this marker development will aid in several screening processes for breeding programs. The second project focused on the development of high-density SNP-based linkage maps for the C9 chromosome in which the radish introgression had been previously mapped to. The developed maps provided a better understanding of the introgression in our improved restorer populations and will facilitate the development of further markers and guides for sequencing and editing that can result in improved agronomic performance of the restorers. In the third project, the number of intermating cycles required to obtain restorer lines with improved agronomic traits was evaluated using a recurrent selection protocol. Results showed that more than a third of the families had improved seeds per pod, pod number and yield when compared to the best parent after one (C1) and two cycles (C2) of random full-sib intermating in select crosses. This indicates that intermating restorer genotypes can produce improved restorer genotypes. All the findings of this thesis benefit the entire rapeseed industry and expand the knowledge available on the Ogu-INRA CMS restorers.

Since the construction of genetic maps using molecular markers requires the development of segregating doubled haploid populations in numerous crop species, we hope that this Manual will also help molecular biologists in establishing such mapping populations. For many years, both the Food and Agriculture Organization of the United Nations (FAO) and the International Atomic Energy Agency (IAEA) have supported and coordinated research that focuses on development of more efficient doubled haploid production methods and their applications in breeding of new varieties and basic research through their Plant Breeding and Genetics Section of the Joint F AO/IAEA Division of Nuclear Techniques in Food and Agriculture. The first F AO/IAEA scientific network (Coordinated Research Programme - CRP) dealing with doubled haploids was initiated by the Plant Breeding and Genetics Section in 1986.
