Available in PDF, EPUB and Kindle. Book excerpt: The endothelium, a monolayer of endothelial cells, constitutes the inner cellular lining of the blood vessels (arteries, veins and capillaries) and the lymphatic system, and therefore is in direct contact with the blood/lymph and the circulating cells. The endothelium is a major player in the control of blood fluidity, platelet aggregation and vascular tone, a major actor in the regulation of immunology, inflammation and angiogenesis, and an important metabolizing and an endocrine organ. Endothelial cells controls vascular tone, and thereby blood flow, by synthesizing and releasing relaxing and contracting factors such as nitric oxide, metabolites of arachidonic acid via the cyclooxygenases, lipoxygenases and cytochrome P450 pathways, various peptides (endothelin, urotensin, CNP, adrenomedullin, etc.), adenosine, purines, reactive oxygen species and so on. Additionally, endothelial ectoenzymes are required steps in the generation of vasoactive hormones such as angiotensin II. An endothelial dysfunction linked to an imbalance in the synthesis and/or the release of these various endothelial factors may explain the initiation of cardiovascular pathologies (from hypertension to atherosclerosis) or their development and perpetuation. Table of Contents: Introduction / Multiple Functions of the Endothelial Cells / Calcium Signaling in Vascular Cells and Cell-to-Cell Communications / Endothelium-Dependent Regulation of Vascular Tone / Conclusion / References

Available in PDF, EPUB and Kindle. Book excerpt: Intracellular Calcium Regulation Editor Felix Bronner A notable development in biological research is the recent widespread interest in the role played by calcium in cellular metabolism. Calcium homeostasis has been studied intensively for many years and mechanisms by which calcium absorption, excretion and bone deposition are effected and regulated are beginning to be understood in detail. It now appears there may exist a parallel between the maintenance and regulation of intracellular and extracellular calcium. This volume presents up-to-date information on intracellular calcium regulation and thus provides a foundation for studies that aim to explore the relationship between intracellular and extracellular calcium metabolism. Intracellular Calcium Regulation examines in detail what happens to calcium inside the cell and offers authoritative discussions on such topics as: calcium fluxes: defining the role played by the plasma membrane in regulating the cellular calcium level, the application of tracer techniques to an analysis of the cellular calcium pool, the compartments making up the pool and the rates of exchange between compartments processes of calcium entry into the cell: describing entry into cells that do not fire action potentials, as well as three types of voltage-sensitive calcium channels, including their distribution, pharmacology, molecular properties and modulation organelles, such as the endoplasmic reticulum, the Golgi complex, lysosomes, or mitochondria and their role in regulating intracellular calcium levels the new, exciting field of calcium-binding or calcium modulated proteins, such as troponin-C, parvalbumin, S-100 proteins, calbindins, and calmodulin, as well as some novel calcium-binding proteins like the 42C and 42A proteins, calcyclin and caltractin calcium efflux: analyzing the role and function of the plasma membrane calcium pump and the Na+/Ca2+ exchanger, including the kinetics of ATP hydrolysis, how calcium transport and ATP hydrolysis are linked, the kinetics and properties of the sodium-calcium exchange, its regulation and role in such specialized cells as retinal rods and cardiac myocytes calcium transport as carried out by resting, stimulated and excitable cells with emphasis on activated phospholipase C, involvement of the G proteins and protein kinase C calcium transport as carried out by the specialized transport cells of intestine and kidney, entry via calcium channels, intracellular diffusion as facilitated by the calcium-binding proteins, and extrusion via the Ca-ATPase. Intracellular Calcium Regulation will be of interest to researchers and graduate students in the areas of biochemistry, biophysics, cell biology, nutrition, gastroenterology, nephrology and anatomy.

Book excerpt: The crucial role played by calcium as a cellular messenger has become increasingly evident, as has the recognition that cells spend much energy in maintaining the cytosolic concentration of this cation both constant and low. It is thought they do this to avoid precipitating phosphate, needed as a source of bond energy and to modulate protein structure. Moreover, since calcium that does enter the cell must be disposed with, processes that utilize calcium have evolved, e.g. secretion, contraction, signaling, to name just some. New knowledge concerning the processes of cellular calcium entry, extrusion and the fate of intracellular calcium has accumulated in recent years. Much has also been learned about calcium transport by and across epithelial cells. It seems logical to think that the processes of calcium entry, extrusion and intracellular handling are similar in all cells. We have therefore assembled in one volume overviews and research reports of transport and cellular calcium regulation so as to explore similarities and differences between cells that utilize calcium for metabolic purposes and those whose primary function is transport.

Available in PDF, EPUB and Kindle. Book excerpt: Calcium Entry Channels in Non-Excitable Cells focuses on methods of investigating the structure and function of non-voltage gated calcium channels. Each chapter presents important discoveries in calcium entry pathways, specifically dealing with the molecular identification of store-operated calcium channels which were reviewed by earlier volumes in the Methods in Signal Transduction series. Crystallographic and pharmacological approaches to the study of calcium channels of epithelial cells are also discussed. Calcium ion is a messenger in most cell types. Whereas voltage gated calcium channels have been studied extensively, the non-voltage gated calcium entry channel genes have only been identified relatively recently. The book will fill this important niche.

Calcium, which is an important second messenger of signal transduction pathways, can also operate in the cell nucleus. Different calcium binding proteins, which are the targets of cellular calcium, have been identified in the nucleus of many different cell types. Prominent among these calcium binding proteins is calmodulin, which appears to be involved in the regulation of major nuclear functions such as gene expression and DNA replication.

Critical to our survival, it consists of a tantalizing array of interacting phenomena, from ionic transport, membrane channels and receptors through cellular metabolism, energy production to fiber mechanics, microcirculation, electrical activation to the global, clinically observed, function, which is measured by pressure, volume, coronary flow, heart rate, shape changes and responds to imposed loads and pharmaceutical challenges. It is a complex interdisciplinary system requiring the joint efforts of the life sciences, the exact sciences, engineering and technology to understand and control the pathologies involved. The Henry Goldberg Workshops were set up to address these multivariable, multidisciplinary challenges. Briefly, our goals are: To encourage international cooperation and foster interdisciplinary interaction between scientists from the different areas of cardiology; to relate microscale cellular phenomena to the global, clinically manifested cardiac function; to relate conceptual modeling and quantitative analysis to experimental and clinical data; to gain an integrated view of the various interacting parameters, identify missing links, catalyze new questions, and lead to better understanding of the cardiac system. The outstanding success of past workshops has encouraged their continuation. The first Henry Goldberg Workshop, held in Haifa in 1984, introduced the concept of interaction between mechanics, electrical activation, perfusion and metabolism, emphasizing imaging in the clinical environment. The second Workshop, in 1985, discussed the same parameters with a slant towards the control aspects.

Book excerpt: All of the parasitic organisms highlighted in this new book represent medically important human pathogens that contribute significantly to the global burden of disease. As such there is intense interest in understanding the molecular basis of infection by these pathogens—not only with regard to their clinical relevance but also the fascinating biology they reveal. For most of the parasites discussed here the ability to penetrate biological barriers and/or to establish intracellular residence is critical to survival of the pathogen in the mammalian hosts. For other parasites, a tissue invasive phenotype is a key virulence determinant. In the ensuing 18 chapters, select members of this diverse set of protozoan parasites, as well as some examples of the extremely reduced fungal parasites classified as Microsporidia, are discussed within the context of the fascinating molecular strategies employed by these organisms to migrate across biological barriers and to establish residence within target host cells.
