Available in PDF, EPUB and Kindle. Book excerpt: Today, the basic mood of researchers and clinical investigators, both at the center and on the periphery of interferon studies, is optimistic regarding the future of interferons as therapeutic substances. Many also feel these polypeptides will prove invaluable probes in unraveling certain fundamental biochemical processes which control the life cycle and developmental pattern of many human cells. In contrast, only a year or two ago, this optimism had given way to an attitude almost of disenchantment as public and scientific expectations were raised steeply, then rapidly waned, as it turns out, prematurely. Both the mUltiple actions of interferons (a virtual cascade of biochemical reactions may be induced, as documented herein) and the high visibility of interferon research provided by the millions of dollars invested both by national health agencies and by multinational pharmaceutical companies, contributed to an upsweep in public attention to drug development probably unprecedented in this century. Virtually every oncologist, it would seem, was plagued by requests for the experimental agent, although they already had therapies of more proven value. As recently as 1980, even though interferon had achieved success against certain cancers and certain viral diseases, the variability in clinical results was seemingly ever present and little evidence emerged to suggest interferons could cure advanced diseases. Why then the resurgence of an optimistic mood? There are almost always many elements which contribute to happiness, and this is certainly true of the broad frontier of interferon and its place in biochemical research and treatment.

Book excerpt: Interferons documents the proceedings of a major international conference on Chemistry and Biology of Interferons: Relationship to Therapeutics held at Squaw Valley, California, in March 1982. The meeting was one of the 1982 UCLA Symposia on Molecular and Cellular Biology organized through the Molecular Biology Institute of the University of California at Los Angeles. The book is organized into four parts. Part I presents papers on recombinant DNA production of interferon. It includes studies on the synthesis of human immune interferon in E. coli and monkey cells; the expression of human ß interferon gene in heterologous cells; and the molecular activities of recombinant and hybrid leukocyte interferons. Part II deals with the biology of interferons, covering topics such as interferon production control process; the effect of interferon on cellular enzymes; and human amniotic interferon. Part III examines the factors relevant to clinical application of interferons, including structure-function studies with human interferon-gamma and on human alpha interferon. Part IV presents clinical studies of interferon, such as interferon a trials in human tumor diseases and interferon in renal transplant recipients.

Book excerpt: Interferons: A Primer covers general information on interferon, including analysis, purification, production, properties, mechanism of action, and clinical uses. Organized into 10 chapters, the book starts with a short history of interferon’s discovery by Isaacs and Lindenmann, followed by topics on assays for interferon, such as factors affecting interferon assays and rapid biological assay. Chapters 3 to 6 discuss the purification, properties, production, antiviral action, and other functions of interferon. Chapters 7 and 8 examine the recovery from viral infection and clinical uses of interferon, with emphasis on treatment of human cancer. The concluding chapters focus on the application of interferon studies on two revolutionizing fields of biology, namely, the cloning of animal genes in microorganisms and the production of monoclonal antibodies. This book is intended for students, scientists, physicians, or educated laypersons who wish to know something about interferons, but do not plan carrying out research in this area.

Available in PDF, EPUB and Kindle. Book excerpt: Interferons were among the first cytokines to be studied clinically in recombinant form in the 1980s, beginning a new era of pharmacotherapy in internal medicine. Interferons were soon employed in the treatment of tumors and it was shown that those hematopoietic neoplasias which had been difficult to treat with chemotherapy responded well to interferon treatment. In the present volume, renowned international authors update the importance of interferons in various fields of internal medicine, dermatology, and urology. The book also contains a general introduction into the classification, structure, and mode of action of interferons. Further chapters discuss the range of side effects of these substances and point out possible focuses of further clinical interferon research.

The book starts with an analysis of the profound effect that viral epidemics had on world history resulting in demographic upheavals by destroying total populations. It also provides a brief history of virology and immunology. Furthermore, the use of viruses for the treatment of cancer (viral oncolysis or virotherapy) and bacterial diseases (phage therapy) and as vectors in gene therapy is discussed in detail. Several chapters focus on viral diseases such as smallpox, influenza, polio, hepatitis and their control, as well as on HIV and AIDS and on some emerging viruses with an interesting story attached to their discovery or vaccine development. The book closes with a chapter on biological weapons. It will serve as an invaluable source of information for beginners in the field of virology as well as for experienced virologists, other academics, students, and readers without prior knowledge of virology or molecular biology.
