Book excerpt: "The onset of acute graft-versus host disease (aGVHD) is accompanied by macrophage priming and the presence of bacteria derived lipopolysaccharide (LPS) in the sera and organs of transplanted animals. Priming of macrophages occurs despite suppression of T cell function. We have investigated whether interleukin 12 (IL-12) mediates the continued production of interferon gamma (IFN-$ gamma$) during the state of T cell immunosuppression accompanying aGVHD. AGVHD was induced in non-irradiated AxC57BL/6 mice by an injection of C57BL/6 lymphoid cells. Despite T cell immunosuppression, macrophages remained primed as shown by their expression of inducible nitric oxide synthase (iNOS) mRNA and production of nitric oxide (NO) in response to exogenous LPS. Continued exposure to IFN-$ gamma$ was found to be required in order to maintain the primed state of macrophages during aGVHD; IFN-$ gamma$ mRNA within target organs of aGVHD including thymus, salivary gland, and lung was increased between day 7 and 14 after transplantation and was accompanied by the induction of the p40 peptide of IL-12 and iNOS mRNA. p40 peptide mRNA was also increased in macrophages purified on day 14 of aGVHD. These results provide evidence for localized production of IFN-$ gamma$ in aGVHD target organs and suggest that it is mediated by LPS induced IL-12 production by macrophages." --

Book excerpt: There are few effective treatments for sclerotic skin disorders including systemic scleroderma, chronic graft-versus-host disease (GVHD) and morphea. Because interferon (IFN)-u03b3 directly inhibits the proliferation of skin fibroblasts and the production of collagen in vitro, IFN-u03b3 is known as an antifibrotic factor. However, administration of recombinant IFN-u03b3 could not inhibit fibrosis in patients with systemic scleroderma. To elucidate the role of IFN-u03b3 on skin fibrosis, we established a new murine model of scleroderma. We found that chicken ovalbumin (OVA)-specific CD8 T cell (OT-I cell)-transferred transgenic mice that express membrane-bound OVA under the control of a keratin 14 promoter (K14-mOVA Tg mice), which develop acute GVHD-like mucocutaneous disease 14 days after the transfer, present chronic GVHD-like scleroderma 28 days after the transfer. While GFP+IFN-u03b3-/-OT-I cells-transferred K14-mOVA Tg mice developed acute GVHD-like mucocutaneous disease as severe as GFP+OT-I cell-transferred mice 14 days after the transfer, GFP+IFN-u03b3-/-OT-I cells-transferred K14-mOVA Tg mice developed significantly milder scleroderma than GFP+OT-I cell-transferred mice 28 days after the transfer, which were assessed by dermal thickness (the averages: 93.1 v.s. 118.6u03bcm), the numbers of infiltrating u03b1SMA+ fibroblasts (126 v.s. 291/HPF), and hydroxyproline contents (5.9 v.s. 10.9u03bcg/mg). In addition, the levels of mRNA of Col1a2 (30.5 v.s. 90.0), u03b1SMA (0.6 v.s. 1.3) 28 days after the transfer, TGFu03b2 (0.7 v.s. 1.3) 14 days after the transfer were significantly lower in the skins of GFP+IFN-u03b3-/-OT-I cell-transferred K14-mOVA Tg mice than in those of GFP+OT-I cell-transferred mice by quantitative real-time PCR. Moreover, immunostaining with anti-GFP antibodies detected more GFP+OT-I cells infiltrating in the skins of K14-mOVA Tg mice than GFP+IFN-u03b3-/-OT-I cells 21 days after the transfer, which correlated to the levels of mRNA of IFN-u03b3-inducible chemokines (CXCL9, CXCL10 and CXCL11). Collectively, IFN-u03b3 may play a crucial role as a trigger in the pathogenesis of skin fibrosis.
