Available in PDF, EPUB and Kindle. Book excerpt: This volume deals specifically with those antituberculosis drugs which passed the preclinical phase and have been or are used in the treatment of tuberculosis and other mycobacterial diseases (except leprosy) in at least some parts of the world. Despite this restriction, there are 14 such drugs, and as a result this volume has reached rather large proportions. To prevent it from becoming even larger and more unwidely, most derivatives of antituberculotics have been omitted, especially where it is claimed that they provide only better bioavailibility or tolerability. Only in the chapter on the chemotherapy of diseases due to so-called atypical mycobacteria is the clinical use of the drugs described to a certain extent. In addition to antituberculotics, also discussed are antimicrobials which have been found to be effective against these mycobacteria. The sequence in which the drugs are described is historical, reflecting not the time of discovery but rather the first clinical application. This order was selected for reasons which are now no longer relevant. In this volume less emphasis is placed on detection, biological or synthetic production of antituberculotics, and structure-activity relationships. In contrast, emphasis is put on the degree, type, and mechanism of antimyco bacterial activity, pharmacokinetics, and biotransformation in animals and man, on experimental pharmacodynamics, and on the toxicity of antituberculotics used therapeutically.

Available in PDF, EPUB and Kindle. Book excerpt: The "functional" in the title of this book not only reflects my personal bias about neuroanatomy in brain research, it is also the gist of many chapters which describe sophisticated ways to resolve structures and interpret them as dynamic entities. Examples are: the visualization of functionally identified brain areas or neurons by activity staining or intracellular dye-iontophoresis; the resolution of synaptic connections between physiologically identified nerve cells; and the biochemical identification of specific neurons (their peptides and transmitters) by histo- and immunocytochemistry. I personally view the nervous system as an organ whose parts, continuously exchanging messages, arrive at their decisions by the cooperative phenome non of consensus and debate. This view is, admittedly, based on my own ex perience of looking at myriads of nerve cells and their connections rather than studying animal behaviour or theorizing. Numerous structural studies have demonstrated that interneurons in the brain must receive hundreds of thousands of synapses. Many neurons receive inputs from several different sensory areas: each input conveys a message about the external world and possibly also about past events which are stored within the central nervous system. Whether an interneuron responds to a certain combination of inputs may be, literally, a matter of debate whose outcome is decided at the post synaptic membrane. A nerve cell responding to an overriding command is possibly a rare event.

Available in PDF, EPUB and Kindle. Book excerpt: From a mathematical point of view, physiologically structured population models are an underdeveloped branch of the theory of infinite dimensional dynamical systems. We have called attention to four aspects: (i) A choice has to be made about the kind of equations one extracts from the predominantly verbal arguments about the basic assumptions, and subsequently uses as a starting point for a rigorous mathematical analysis. Though differential equations are easy to formulate (different mechanisms don't interact in infinites imal time intervals and so end up as separate terms in the equations) they may be hard to interpret rigorously as infinitesimal generators. Integral equations constitute an attractive alternative. (ii) The ability of physiologically structured population models to increase our un derstanding of the relation between mechanisms at the i-level and phenomena at the p-level will depend strongly on the development of dynamical systems lab facilities which are applicable to this class of models. (iii) Physiologically structured population models are ideally suited for the for mulation of evolutionary questions. Apart from the special case of age (see Charlesworth 1980, Yodzis 1989, Caswell 1989, and the references given there) hardly any theory exists at the moment. This will, hopefully, change rapidly in the coming years. Again the development of appropriate software may turn out to be crucial.

Book excerpt: General aspects of nucleic acid uptake by mammalian cells have been the subject of several reviews during the last few years (PAGANO, 1970; BHARGAVA and SHANMUGAM, 1971; DUBES, 1971; RYSER, 1967). These reviews covered methods used for the infection of cells by viral nucleic acids as well as interaction of mammalian cells with non-viral nucleic acids. This article is restricted to a discussion of experiments with poliovirus RNA and focuses special attention on the steps following the uptake of RNA into a cell, aspects that were not discussed in earlier review articles. The fate of input RNA once inside the cell is determined by the host cell but experimental conditions can be chosen to favor the survival of input RNA and the induction of a virus growth cycle by interfering with host-cell meta bolism through events that, in the case of infection with intact virus, might be controlled by viral proteins.

Available in PDF, EPUB and Kindle. Book excerpt: The International Society of Chemotherapy meets every two years to review progress in chemotherapy of infections and of malignant disease. Each meeting gets larger to encompass the extension of chemotherapy into new areas. In some instances, expansion has been rapid, for example in cephalosporins, pen icillins and combination chemotherapy of cancer - in others slow, as in the field of parasitology. New problems of resistance and untoward effects arise; reduction of host toxicity without loss of antitumour activity by new substances occupies wide attention. The improved results with cancer chemotherapy, es pecially in leukaemias, are leading to a greater prevalence of severe infection in patients so treated, pharmacokinetics of drugs in normal and diseased subjects is receiving increasing attention along with related problems of bioavailability and interactions between drugs. Meanwhile the attack on some of the major bacterial infections, such as gonorrhoea and tubercu losis, which were among the first infections to feel the impact of chemotherapy, still continue to be major world problems and are now under attack with new agents and new methods. From this wide field and the 1,000 papers read at the Congress we have produced Proceedings which reflect the variety and vigour of research in this important field of medicine. It was not possible to include all of the papers presented at the Congress but we have attempted to include most aspects of cur rent progress in chemotherapy.
