Book excerpt: One of the central questions in immunology is the understanding in molecular terms of antigen-antibody interactions and of the cellu lar recognition of antigens. It is hoped that this understanding will extend eventually to the immunobiological basis of host defense to infectious agents and of tissue damage or deranged cell functions which stem from these reactions. A variety of natural and artificial substances have been used as models for these studies. Emphasis was placed upon substances of known and relatively uncomplicated chemical structures. These included polysaccharides, amino acid polymers, nu cleic acids and haptens. On the other hand. until recently there has been very little information on protein antigens. The complexity of these molecules posed an immense chemical obstacle to precise immuno chemical analysis. Indeed, it is this difficulty with proteins that spurred the synthesis and immunological studies of amino acid poly mers. The control and normal regulation of the immune system at the cellular-molecular interface and the great majority of antigens asso ciated with immune disorders are attributed to protein molecules. In the last few years great advances have been made in the analysis and synthesis of the antigenic sites of some proteins. The entire antigenic structures of myoglobin and lysozyme and the partial anti genic structures of several other proteins have been determined. Moreover, in the past seven years several biological responses resulting from the reactions of proteins and their peptides with cells of the immune system were described.

The articles represent papers by invited speakers as well as papers selected by the Scientific Council, from among those submitted by the participants, on the basis of quality and timeliness. This symposium series was established in 1976 for the purpose of bringing together, once every two or three years, active investigators in the forefront of contemporary immunology, to present their findings, discuss their significance in the light of current concepts and identify important new directions of investigation. The founding of the symposium was stimulated by the achievement of major breakthroughs in the understanding of the immune recognition of proteins and peptides. We believed that these breakthroughs would lead to the creation of a new generation of peptide reagents, which could have enormous potential in biological, therapeutic, and basic applications. This anticipated explosion has since occurred and many applications ofthese peptides are now being realized. The eighth symposium focused on the manipulation or modulation of the immune response. This volume broadly covers the areas of adjuvants, cytokines, vaccines, and the use of intravenous immunoglobulins for disease management. There is a clear need to identify methods for improving vaccine efficacy and guiding the host to respond with a particular type of immune response.

Immunobiology still shows little sign of decreasing its active growth rate, but rather is continuing to broaden its range of interests and applications, particularly as new techniques and methods are adapted from other fields of medical research. This present volume reflects both several of the more classical areas of immunology now addressed in the light of contemporary immunology, and several newer directions that have been taken in other fields. The general subject of T-cell heterogeneity and functions of T-cell subpop ulations is addressed in Chapters 1 and 2. The potential role of genes of the major histocompatibility complex in controlling the immune functions of T lymphocytes still remains a major unresolved issue in immunogenetics, and the current status of this problem is excellently reviewed by J. F. A. P. Miller. The further elucidation of functional subpopulations of human T lymphocytes has been particularly hampered by the lack of available markers for character izing and isolating such sUbpopulations. A major step in this direction has been made by L. Moretta, M. Ferrarini, and M. D. Cooper, who review their ex perience with Fc-receptor-bearing human T-Iymphocyte populations.

With well over a thousand biomedical journals producing new issues each month, and with approximately five hundred new technical books in biomedicine being 1 published each year, not to mention the proliferation of information-ex change meetings, it is all too clear that we are in danger of being inundated by a flood of tables, figures, and hypotheses. The problem is particularly acute in immunology, as the rate of information production is increasing geometrically, and immunological approaches have been extended into other biological and medical fields to further diversify the research over a vast literature. Abstracting and information-retrieval services do much to improve the investigator's lot, but do not offer solutions for one particularly distressing aspect of the crisis. In the midst of our informational overabundance, one often finds that interrelationships between an investigator's collective findings are becoming blurred, or that the relation of his total work to the field are not clear. Although review articles are indispensable in fixing the status of a given problem, they do not provide the detailed attention to a single author's work that is needed.

With respect to the public health and economic problems, malaria, for example, remains a major health problem with approximately 200 million people being infected yearly and, on the basis of World Health Organiza tion estimates, more than 1 million children die each year of malaria infections (Chapter 4). This volume addresses state-of-the-art immunologic approaches to the development of vaccines for parasitic diseases (Chapter 9) and analyses of studies bearing on the antigenic characterization of protozoan and metazoan parasites (Chapters 4, 5, and 7), on investigations of the role of precise mecha nisms underlying natural resistance or non permissiveness of the host to parasitic infections (Chapters 1, 2, and 12), on induced mechanisms including the genera tion of parasite-specific T-cell lines and clones (Chapter 6), and on the generation of monoclonal antibodies (Chapters 4 and 5) to parasite antigens of distinct de velopmental stages. Great progress has been made in characterizing parasite antigens capable of inducing a protective response in the vaccinated host; further progress in this area strongly depends on biochemistry and molecular biology with the long-term goal of synthesizing such antigens chemically or producing them by means of recombinant DNA technology (Chapter 4).

Book excerpt: This book contains the proceedings of a conference devoted to the study of the structure and function of Terminal deoxynucleotidyl Transferase (TdT) and its utilization as biochemical marker in immuno biology and leukemia, held in Elba, Italy on May 28-31, 1981. The en zyme has been known to nucleic acid biochemists for more than 20 years and has proved to be an excellent tool for making deoxypolymers, label ing DNA fragments, and adding homopolymer tails to restriction endonu clease fragments from DNA. Since the discovery of its peculiar tissue distribution, normally restricted to the thymus and bone marrow, and of its abnormal occur rence in human leukemic lymphoblasts, TdT has become one of the most widely used markers in the study of lymphocyte differentiation and in the classification of hematopoietic neoplasia. The subject seemed most appropriate for a meeting where molecular and cellular biologists, immunologists and hematologists could convene for the first time to discuss both basic research and the clinical aspects of the problem. Among the goals achieved by this workshop was the sharing of in formation about the enzymology of TdT, biochemical and immunological methodology and the correlation of TdT with other markers in the diag nosis of leukemia. The remarkable accordance of results, presented here by five independent hematological institutions, from analysis of TdT in several thousands of leukemic patients, marks the importance of this enzyme as a diagnostic and prognostic tool in these diseases.
