The book is organized into seven parts that correspond to the seven sessions held during the workshop. The first session focused on the definition of the I-E and I-C subregions, and the mapping and cellular expression of complementing Ir genes. The second session included papers on the expression of gene products of the I region. The third session covered two major areas: (1) the biochemical characterization and primary structure of la antigens; and (2) the possible carbohydrate nature of la antigenic determinant. The studies presented in the fourth session focused on the role of I-region gene products in cellular interactions. The fifth session discussed the "recognition" functions controlled by the I region of the major histocompatibility complex (MHC). The sixth session featured papers on la-containing T cell factors. Finally, the seventh session examined the properties of T-cell antigen receptors.

Book excerpt: The Major Histocompatibility Complex (MHC) was discovered as a con sequence of the chronic problem encountered by cancer biologists in the early years of this century: the failure to maintain tumor lines by serial passage in outbred mice. A number of observations pointed to genetic similarity being a prerequisite for successful transplantation and they were incorporated into a genetic theory of transplantation by C.C. Little. This prompted scientists like Little to initiate breeding experiments designed to test his hypothesis and produce genetically identical mice which would permit the growth of trans planted tumors. Most inbred strains of mice commonly used in immunology derive from those efforts. Transplantation of normal tissues obeyed the same rules found for malignant tissues and rejection was shown to be an immunological phenomenon. G.D. Snell showed that a single genetic locus determined rapid rejection of skin grafts. This was initially called the Major Histocompatibility Locus but was subsequently shown to include many functionally related genes and renamed the Major Histocompatibility Complex (MHC). Trans plantation antigens were discovered and characterized by Gorer about 50 years ago, and the biological basis for the unequalled complexity of their variability between individuals within a species, in spite of extreme conservation between species, was the subject of intense research and discussion for many years. During the days of belief in "immune surveillance" against spontaneously developing tumors, it was suggested that histoincompatibility between members of one species would prevent cancer from being a contagious disease and thus a threat to the species. Immunologists involved in human transplantation had to learn and care about the complexity, especially after 1967, when it was found that HLA antigens were the products of the human MHC. Rejection of HLA-identical sib kidney grafts was so rare, even in those days, that cases of rejection were described in scientific papers.
