Book excerpt: Hormonal Steroids: Biochemistry, Pharmacology, and Therapeutics, Volume I focuses on various research on steroids and their biological and medical involvements. Comprised of 60 chapters, the book presents the literature of various authors who have conducted research on the relationship between hormonal steroids and biochemistry, pharmacology, and therapeutics. The discussions start with the identification of steroids with hormone-like activities. This discussion includes the nature, compositions, properties, possible uses, and reactions of these hormones when exposed to different conditions and controlled environments. The book then proceeds with discussions on synthesis and metabolism of hormonal steroids. These discussions are supported by graphical representations, reviews, recommendations, and methodologies. The book then explains the control of synthesis and release of steroid hormones. This part notes the relationship of renal and adrenal hormones; the control and production of corticosterone; neurosecretion and control of the pituitary gland; and release of ovulating hormones. The book also highlights the mechanism of steroid action, noting the probable interrelationships of steroids, nonsteroids, intermediary metabolism, and inflammation. Various research are presented on the possible clinical applications of steroids. The text is a vital reference for readers who are interested in the study of hormones.

Available in PDF, EPUB and Kindle. Book excerpt: The many new and fascinating data which have been gathered in the period 1985-1990 have made molecular endocrinology into a mature discipline stimulating creative chemists to design innovative drugs. Within a few years, the primary structures of all the steroid receptor proteins and most of the steroid-converting enzymes have been determined and detailed studies of their mechanism of action at a molecular level have become possible. Medicinal chemists have developed steroidal affinity probes to examine the binding sites of enzymes and receptors, and these investigations were supplemented with studies of modified enzymes and receptors prepared with the aid of site-directed mutagenesis. In view of these rapid scientific developments, the author deemed it appropriate to review the present knowledge of the medicinal chemistry of the steroids. In this book, the discussion is supported by the addition of some chapters which summarize the available data on the biological counterparts of the steroids, the steroid-converting enzymes and the steroid receptors.

Available in PDF, EPUB and Kindle. Book excerpt: Steroids regulate differentiation, growth and functioning of many tissues via a nuclear receptor gene activation mechanism. Molecular biology has opened the way for more detailed investigation of the complex set of intracellular interactions contributing to organ-selective actions of steroids and steroid-like compounds such as vitamin D. In this volume leading experts address basic molecular mechanisms of tissue specificity and related issue, with the goal of attaining a better understanding of the underlying mechanisms of organ selective actions of steroids. Discussed is the extent to which tissue specificity might selectively be modified by chemical modification of the hormones and how this may be applied towards improved therapies by taking advantage of tissue selectivity.

This book was released on 2012-12-05 with total page 310 pages. Available in PDF, EPUB and Kindle. Book excerpt: The discovery of mammalian nitric oxide (NO-) synthesis from I-ar ginine has led to profound increases in our understanding of basic physiological and pathophysiological processes. This understanding has been achieved by major advances in several areas. Three major ni tric oxide synthase (NOS) isoforms have been identified at the protein and gene level and their specific tissue localization has been linked to processes subject to regulation by NO-. The molecular basis for the di verse effects of NO- on cell function has been shown to derive from its chemical reactivity with oxygen-containing species and both heme and non-heme iron-dependent enzymes. The potential regulation of heme dependent enzymes such as cytochromes P450, guanylate cyclase, cy clooxygenase, and mitochondrial cytochrome oxidase continues to emerge as a key area in NO- research. In addition, it has become clear that the expression of NOS isoforms and NO- release from cells is subject to regulation by sexual steroids and that, in tum, NO- has the potential to regulate steroid biosynthesis via inhibition of cytochromes P450 involved in steroidogenesis. These recent observations on interactions between the NO-/NOS and cytochrome P450/sexual steroid pathways have important implica tions for understanding fundamental mechanisms involved in endocri nological processes. They are also likely to lead to novel insights and novel therapeutic approaches for the management of pathophysiologi cal conditions associated with alterations in sexual steroid hormones.

Available in PDF, EPUB and Kindle. Book excerpt: For many years, Springer has been publishing an impressive series of textbooks of pharmacology which have set standards in medical science. Surprisingly, an extensive overview of the current state of the art in research on estrogens and anti estrogens was still lacking. The present two volumes on estrogens and antiestrogens provide a comprehensive review of a field of research in which remarkable progress has been made over the past few years. New insights into the mechanisms of steroid hormone action resulted in a tremendous number of publications from which new principles of preventive and therapeutic appli cations of estrogens and antiestrogens emerged. Although various electronic data bases provide easy access to this copious information, there was a clear necessity for a monograph-style textbook which assesses and summarizes current knowledge in this rapidly expanding field of research. It should be noted, however, that, due to this dynamic development, it is barely possible to comprehensively update every aspect of basic and clinical knowledge on estro gens and antiestrogens. Thus, the intention of the editors was to provide the reader with an overview of the "classic" and most recently explored areas of research and stimulate future interests in basic and applied endocrinology. Estrogens were among the first steroid hormones described in the scientific literature. Since they were first isolated, since the chemical, synthesic and pharmacological characterization of naturally occurring estrogens and, later on, of orally active derivatives, estrogen research has produced continu ously hallmark results in reproductive endocrinology worldwide.
