Available in PDF, EPUB and Kindle. Book excerpt: High-temperature liquid chromatography has attracted much interest in recent years but has not yet recognized its full potential in the chromatographic community. There is a widespread reluctance in industry to use temperature to speed up the separation process, influence the selectivity of a separation or implement novel detection techniques. However, the technology has now matured and could revolutionize chromatography as we see it today. Better equipment, such as heating systems able to generate faster heating rates, is becoming more readily available. Also, columns based on silica gel, which can withstand higher temperatures for an extended period, are now being introduced. Nevertheless, further technological and methodical efforts are needed to establish the method in a regulated environment like the pharmaceutical industry. This is the only text to cover all the practical aspects, as well as the underlying theoretical principles, of setting up an HPLC system for high-temperature operation. It is not intended solely for academics but will also benefit the researcher interested in more practical considerations. The author is a recognized expert and has conducted several studies with partners from industry to validate the method. Many real examples from these studies have been included in the book. The aim is to support practitioners in the creation of their own protocols without the need to rely solely on trial and error. The book starts with a brief definition of high-temperature liquid chromatography before going on to cover: system set up; the heating system; mobile phase considerations; suitable stationary phases; method development using temperature programming; analyte stability, and special hyphenation techniques using superheated water as a mobile phase. In each chapter, experimental data is used to illustrate the main statements and the advantages over conventional HPLC are evaluated. The book concludes with a critical outlook on further developments and applications underlining the necessary advances needed to make high-temperature HPLC more robust.

Available in PDF, EPUB and Kindle. Book excerpt: Although size exclusion chromatography (SEC) is perhaps the most popular and widely used technique for determining the molecular weight distribution of polymeric materials, there have been very few texts written on this topic. During the past decade, SEC has experienced a considerable amount of growth in regard to column and detector technology and new applications. With these advances, SEC can now be used for determining absolute molecular weight, polymer chain conformation and size, and branching, as well as polymer solution properties. This book introduces the reader to the fundamentals of SEC with emphasis on practical aspects of the technique, such as column and mobile selection, calibration, new detector capabilities and guidelines for performing SEC on most types of polymers, especially those of industrial importance. This book is intended for either those new to the field of SEC, or for those research workers who require a more comprehensive background.

Book excerpt: There is a growing need for high-throughput separations in food and environmental research that are able to cope with the analysis of a large number of compounds in very complex matrices. Whereas the most common approach for solving many analytical problems has often been high-performance liquid chromatography (HPLC), the recent use of fast or ultra-fast chromatographic methods for environmental and food analysis has increased the overall sample throughput and laboratory efficiency without loss (and even with an improvement) in the resolution obtained by conventional HPLC systems. This book brings together researchers at the top of their field from across the world to discuss and analyze recent advances in fast liquid chromatography–mass spectrometry (LC–MS) methods in food and environmental analysis. First, the most novel approaches to achieve fast and ultra-fast methods as well as the use of alternative and complementary stationary phases are described. Then, recent advances in fast LC–MS methods are addressed, focusing on novel treatment procedures coupled with LC–MS, new ionization sources, high-resolution mass spectrometry, and the problematic confirmation and quantification aspects in mass spectrometry. Finally, relevant LC–MS applications in food and environmental analysis such as the analysis of pesticides, mycotoxins, food packaging contaminants, perfluorinated compounds and polyphenolic compounds are described. The scope of the book is intentionally broad and is aimed at worldwide analytical laboratories working in food and environmental applications as well as researchers in universities worldwide. Available in PDF, EPUB and Kindle. Book excerpt: This book elucidates the peculiar phenomenon of entropy/enthalpy compensation that takes place in high performance liquid chromatography (HPLC) of polymers. Numerous publications, including some books, are devoted to molecular characterization of synthetic polymers, materials presently produced in large and steadily growing quantities, applying methods of HPLC. A knowledge of the molecular characteristics of polymers is indispensable, not only for their proper applications but also for their recycling and remediation. Polymer scientists generally focus on synthesis and potential applications of polymers while not giving due attention to an important central link, their comprehensive characterization in context of development of structure-property correlations. To fill this gap is one of the aims of the present book. The process of entropy/enthalpy compensation plays a decisive role in the advanced method of polymer characterization such as liquid chromatography at critical conditions, eluent gradient interaction chromatography, and temperature gradient interaction chromatography. All chemists working on any aspect of polymer science will find this book a valuable resource for the development of structure-property correlations.

Available in PDF, EPUB and Kindle. Book excerpt: Two-dimensional liquid chromatography (2D-LC) is finding increasingly wide application principally due to the analysis of mixtures of moderate to high complexity. Many industries are developing increasingly complex products that are challenging the separation capabilities of state-of-the-art 1D-LC and need new analytical methodologies with substantially more resolving power, and 2D-LC meets that need. This text, organized by two leaders in the field, establishes a sound fundamental basis for the principles of the technique, followed by a discussion of important practical considerations. The book begins with an introduction to multi-dimensional separations and a discussion of the history and development of the technique over the past 40 years, followed by several chapters that provide a theoretical basis for development of 2D-LC methods, including foundational concepts regarding separation complementarity, under-sampling, and dynamics of liquid chromatography separations. Instrumentation for 2D-LC is discussed extensively, including practical aspects such as interface selection and setup. Building on this foundation, two separate chapters are focused on method development for non-comprehensive and comprehensive separations, followed by a chapter dedicated to data analysis. Finally, applications of 2D-LC in several fields ranging from pharmaceutical analysis to polymer science are summarized. The book is an important resource for both students and practitioners who are already using 2D-LC or are interested in getting started in the field. Key Features: Demonstrates the conditions under which a 2D-LC method should be considered as an alternative to a 1D-LC method. Establishes a sound fundamental basis of the principles of the technique, followed by guidelines for method optimization. Provides a single source for technical knowledge advances and practical guidance described in recent literature. Assists with the initial decision to develop a 2D-LC method. Guides the reader in developing a high-quality method that meets the needs of their application.

Book excerpt: Since the first edition of this book the major advances have been in column packings, where over ninety per cent of separations are now performed using chemically bonded microparticulate packings, and in instrumentation. The use of microprocessor control has brought about a rationalization of mobile phase delivery systems and in detectors, the introduction of electrochemical and spectrophotometric detection other than in the ultra-violet region, has widened the field of applications and the sensitivity of the technique. The use of ion pair chromatography has increased at the expense of ion-exchange and this together with the improvements in detectors has greatly increased the appli cation of the technique in the biomedical field. These advances are described together with the established methods to enable the beginner to carry out a satisfactory separation and to gain the experience necessary for the full exploitation of the technique. R. J. Hamilton P. A. Sewell Liverpool,1981 1 Introduction to high performance liquid chromatography 1. 1 Introduction Chromatography in its many forms is widely used as a separative and an analytical technique. Gas chromatography since its introduction by James and Martin [1] has been pre-eminent in the field. Uquid chromatography in the of paper, thin-layer, ion-exchange, and exclusion (gel permeation and gel form filtration) chromatography had not been able to achieve the same success, mainly because of the poor efficiences and the long analysis times arising from the low mobile phase flow rates.
