Available in PDF, EPUB and Kindle. Book excerpt: Microfabrication technology offers great potential for the integration of all steps of forensic DNA typing onto a single microdevice. This integration should enable rapid, low-cost and reliable short tandem repeat (STR) analysis not only in forensic laboratories but also at crime scenes or other relevant point-of-analysis. As a first step towards making on-site STR typing possible, I developed a microdevice consisting of a 160-nL polymerase chain reaction (PCR) chamber and a 7-cm capillary electrophoresis (CE) channel and a portable instrument for its operation. A four-plex mini Y STR typing system was constructed for testing the capability of this microsystem for forensic STR typing. The successful analyses of casework and mixture samples validate the concept of forensic STR typing on a portable microfluidic system. To critically evaluate the capabilities of this portable system and the feasibility of DNA typing at a crime scene, real-time DNA analyses using a 9-plex autosomal STR typing system on a modified PCR-CE microdevice containing a co-injection structure for fragment sizing calculation were carried out at a mock crime scene. Blood stain collection, DNA extraction, STR analysis on the PCR-CE microsystem, and a DNA profile search against a mock CODIS database were successfully conducted within 6 hours of crime scene arrival. This demonstration establishes the feasibility of real-time DNA typing at a crime scene or other point-of-care situations. Finally, to achieve a total integrated analysis system for real-time STR typing, an up-front sequence-specific DNA extraction and concentration method using magnetic beads was developed and incorporated into the PCR-CE microdevice. Fragmented genomic DNA was hybridized with capture probes and immobilized onto magnetic beads via streptavidin-biotin binding in microchannels. The bead-DNA conjugates were then transported to a PCR reactor for amplification followed by inline injection using a novel capture concentration method for CE separation. This fully integrated system significantly advances the forensic DNA typing by providing a high-performance platform with sample-in-answer-out capability for real-time human identification.

Book excerpt: Since the independent invention of DNA sequencing by Sanger and by Gilbert 30 years ago, it has grown from a small scale technique capable of reading several kilobase-pair of sequence per day into today's multibillion dollar industry. This growth has spurred the development of new sequencing technologies that do not involve either electrophoresis or Sanger sequencing chemistries. Sequencing by Synthesis (SBS) involves multiple parallel micro-sequencing addition events occurring on a surface, where data from each round is detected by imaging. New High Throughput Technologies for DNA Sequencing and Genomics is the second volume in the Perspectives in Bioanalysis series, which looks at the electroanalytical chemistry of nucleic acids and proteins, development of electrochemical sensors and their application in biomedicine and in the new fields of genomics and proteomics. The authors have expertly formatted the information for a wide variety of readers, including new developments that will inspire students and young scientists to create new tools for science and medicine in the 21st century.Reviews of complementary developments in Sanger and SBS sequencing chemistries, capillary electrophoresis and microdevice integration, MS sequencing and applications set the framework for the book.* 'Hot Topic' with DNA sequencing continuing as a major research activity in many areas of life science and medicine.* Bringing together new developments in DNA sequencing technology* Reviewing issues relevant to the new applications used

Book excerpt: As rapid technological developments occur in electronics, photonics, mechanics, chemistry, and biology, the demand for portable, lightweight integrated microsystems is relentless. These devices are getting exponentially smaller, increasingly used in everything from video games, hearing aids, and pacemakers to more intricate biomedical engineering and military applications. Edited by Kris Iniewski, a revolutionary in the field of advanced semiconductor materials, Integrated Microsystems: Electronics, Photonics, and Biotechnology focuses on techniques for optimized design and fabrication of these intelligent miniaturized devices and systems. Composed of contributions from experts in academia and industry around the world, this reference covers processes compatible with CMOS integrated circuits, which combine computation, communications, sensing, and actuation capabilities. Light on math and physics, with a greater emphasis on microsystem design and configuration and electrical engineering, this book is organized in three sections—Microelectronics and Biosystems, Photonics and Imaging, and Biotechnology and MEMs. It addresses key topics, including physical and chemical sensing, imaging, smart actuation, and data fusion and management. Using tables, figures, and equations to help illustrate concepts, contributors examine and explain the potential of emerging applications for areas including biology, nanotechnology, micro-electromechanical systems (MEMS), microfluidics, and photonics.

The understanding of molecular informa tion handling in nature is essential for the molecular optimization in chem istry, molecular biology, molecular pharmacology and therefore - as an ex ample - for the development of specifically acting drugs. The famous recent method of technical information management is digital electronics. Over the past few years, evidence has arisen that computerized and molecular information managements have many similar and overlapping aspects. For example, both technology and nature use digitized information and both use small structures for the efficient handling of information. Furthermore, they optimize their processes in order to gain a maximum of information with a minimum of invested energy. During the last two decades, novel experimental techniques in biomolec ular sciences have paved the way for artificial biomolecular optimization. In the same time interval, the progress of micro system technology has been extended from the field of digital electronics and sensing to micro liquid hand ling, and the field of chip-supported substance handling began. It appears that the "marriage" of physical micro technology and molecular processing will be consummated soon. The contact of both fields has been realized in for ex ample DNA chips. Such connections will also become relevant in additional fields in the near future. Biomolecular investigations are the first to profit from these fast growing scientific and technical connections between micro systems and molecular sciences.

Book excerpt: This thesis reports on the development of a fully integrated and automated microsystem consisting of low-cost, disposable plastic chips for DNA extraction and PCR amplification, combined with a reusable glass capillary array electrophoresis chip, which can be employed in a modular-based format for genetic analysis. In the thesis, DNA extraction is performed by adopting a filter paper-based method, followed by an “in-situ” PCR carried out directly in the same reaction chamber of the chip without elution. PCR products are then co-injected with sizing standards into separation channels for detection using a novel injection electrode. The entire process is automatically carried out by a custom-made compact control and detection instrument. The author thoroughly tests the system’s performance and reliability by conducting rapid genetic screening of mutations on congenital hearing loss and pharmacogenetic typing of multiple warfarin-related single-nucleotide polymorphisms. The successful development and operation of this microsystem establishes the feasibility of rapid “sample-in-answer-out” testing in routine clinical practice.

Available in PDF, EPUB and Kindle. Book excerpt: Micro-TAS '98 is the third of a series of symposia initiated by MBSA (University of Twente) in 1994, on the subject of miniaturizing, and integrating within a monolithic structure, the chemical, biochemical and biological procedures commonly used for analysis and synthesis. The primary tool used to develop micro-total analysis systems (mu- TAS) has been micro-photolithographic patterning and micromachining. These powerful tools of Micro System Technology (MST or MEMS) have been applied in highly imaginative ways to develop microchip chemical arrays, fully integrated pump and fluid manifolds, and electrokinetically driven micro-channel systems to be used for genetic analysis, clinical diagnostics and environmental monitoring, and to integrate reactions as diverse as the polymerase chain reaction (PCR) and the large volume, partial oxidation of ammonia. This text illustrates the rapid expansion of the field, the extensive industrial involvement, the increasing number of participating researchers, the expanding range of concepts and applications that utilize MST and microfluidic devices, and new MST-compatible plastic micro-machining to meet the needs of the life science community. This volume contains the proceedings of the Third International Symposium on Micro-Total Analysis Systems, mu-TAS '98, held on October 13-16 in Banff, Alberta, Canada. State-of-the-art invited and contributed papers presented by the world's leading mu- TAS research groups provide a highly informative picture of the growth since 1994 and of the promising future of this exciting and rapidly growing field.

Book excerpt: To produce a microsystem with favorable performance characteristics for genetic-based analyses, several key operational elements must be strategically chosen, including device substrate material, temperature control, fluidic control and reaction product readout. The advantages afforded by fully integrated microfluidic systems to enable challenging applications, such as single-copy DNA sequencing, single-cell gene expression analysis, pathogen detection, and forensic DNA analysis in formats that provide high throughput and point-of-analysis capabilities must be addressed by the researcher. This book focus on discussion on microfluidic systems that are composed of two or more microdevices directed toward DNA analyses. The discussions primarily focus on the integration of various processing steps with u-capillary electrophoresis (uCE) with discoveries that have led to the development of fully autonomous and functional integrated systems for genome processing that can supply "sample in / answer out" capabilities."

Book excerpt: The Fifth International Conference on Micro Total Analysis Systems, also known as JlTAS 2001, will highlight the latest exciting events in the world ofminiaturized devices and systems for performing chemical and biochemical experimentation This conference has become mandatory for those of us working in this field as it is indeed helping to define our discipline. We are grateful to the people of the MESA Research Institute of the University of Twente, particularly Piet Bergveld and Albert van den Berg, for starting this meeting in 1994. Their original intention was for the JlTAS meeting to be a small informal workshop. This workshop flavor was sustained through the second meeting held in Basel in 1996, but already in 1998 at the third meeting in Banff it was clear that the "workshop" had become a conference with 420 attendees. It was due to this clearly growing interest in microchemical systems that it was decided we should consider gradually moving toward an annual format and prepare for the possibility that the meeting would increase in popularity. Albert van den Berg was still yearning for a workshop at the JlTAS 2000 meeting and planned a single session format. Again there was a large increase in submitted abstracts (more than 230 total) and a further increase in attendance. The JlTAS steering committee again agreed that we would have to prepare to address the demand the meeting was receiving.
