Book excerpt: This contribution also includes the implementation of cocrystallization for various categories of compounds which includes nutraceuticals, pharmaceuticals and ionic salts for the design and synthesis of molecular and ionic cocrystals. Chapter 1 highlights how supramolecular synthon approach can be used to design and synthesize multi-component crystals, namely, cocrystals. The role of the CSD and its importance in crystal engineering has also been discussed. Chapters 2 and 3 focus on new persistent supramolecular synthons in the context of nutraceuticals. The cocrystals isolated in the study are also compared with the existing cocrystals in the CSD supramolecularly in terms of synthon formation. These persistent supramolecular synthons are helpful in developing hierarchies which could be utilized and applied to similar and analogous compounds. The main feature of Chapter 4 is expanding the field of cocrystallization by studying the properties of cocrystals. Some of the properties which have been examined here include effects of cocrystallization on solubility and correlations between the solubility of cocrystal with cocrystal former (CCF) and melting point of the cocrystal. The extension of cocrystals to the active pharmaceutical ingredients (APIs) has been explored in the context of pharmaceutical cocrystals by selecting a BCS class IV drug, hydrochlorothiazide in Chapter 5. Chapter 6 highlights the hybridization of organic and inorganic components for the synthesis of ionic cocrystals and is exemplified by considering lithium salts with achiral and homochiral amino acids for the generation of 1:1 and 1:2 cocrystals.

Book excerpt: The work presented in this thesis has demonstrated that supramolecular synthons can be used to make multicomponent crystals, and various synthons can be combined to make supermolecules. The synthons can also be used to construct nanoscale assemblies. Molecules containing single and multiple hydrogen-bond (HB) and halogen-bond (XB) acceptor sites have been synthesized in an effort to carry out supramolecular synthesis in order to establish a reliable hierarchy for intermolecular interactions. Pyrazole-based molecules have been made, combined with various carboxylic acids, and characterized using infrared (IR) spectroscopy to give a success rate of 55-70%. Reactions that gave a positive result were converted to solution experiments, and crystals were grown and characterized using single-crystal X-ray diffraction (XRD). The co-crystals display infinite 1-D chains with the intended stoichiometry and structural landscape on 6/6 occasions. The salts, on the other hand, display unpredictable stoichiometry and structural landscape on 5/5 occasions. Furthermore, the electrostatic charge on the primary hydrogen-bond acceptor, N(pyz), can be altered by adding a nitro, R-NO2, covalent handle to the backbone of the pyrazole molecule. Addition of a strongly electron withdrawing group significantly lowered the charge on the pyrazole nitrogen atom and, in turn, lowered the supramolecular yield to 10%. Ditopic molecules containing pyrazole and pyridine on the same molecular backbone were synthesized and characterized using 1H NMR. The molecules were co-crystallized with carboxylic acids, and the resulting solids were characterized using IR spectroscopy. The solids could then be classified as co-crystal or salt using specific markers in the IR spectrum. Single-crystal XRD was used to observe the intermolecular interactions in the co-crystals and salts, and the co-crystals were assigned to two groups: Group 1 (2) and Group 2 (2). The salts (4) show more unpredictability with stoichiometry and structural landscape. A library of ditopic molecules containing triazole and pyridine acceptor sites were synthesized and characterized using 1H and 13C NMR. The molecules were co-crystallized with carboxylic acids and the resulting solids were characterized using IR spectroscopy which demonstrated a 100% supramolecular yield whenever a pyridine moiety was present, consistent with results from Chapter 3. Single-crystal XRD was used to identify the intermolecular interactions in the co-crystals (2) and salt (1), and the results show that triazole can compete with pyridine for hydrogen bond donors. A library of ditopic molecules was also used for halogen-bonding (XB) studies with a series of activated iodine and bromine-based donors. The results show that iodine donors have a higher success rate range (12.5-75%) compared to bromine donors (16.7-50%) based on results obtained from IR spectra. Furthermore, the results from the XRD show that pyrazole nitrogen atoms can compete with pyridine for forming XB, and two groups of supramolecular synthons were observed. Finally, relatively weak non-covalent interactions, HB and XB, can influence the assembly of nanoparticles based on IR spectroscopy and TEM images. The assembly of the particles is influenced by specific capping ligands, which were synthesized and characterized using 1H, 13C and 19F NMR. The results demonstrate that relatively weak non-covalent interactions based on HB and XB interactions can influence nanoparticle assembly.

It is an attempted response to the question: how can additional crystal-engineering guidelines regarding supramolecular structures in molecular crystals be derived in research, and what is the relevance of supramolecular interactions to synthesise novel organic materials for paediatric drug formulations? The literature reports the importance of supramolecular interactions, in particular supramolecular synthons, to design functional organic materials using the synthon-based approach. However, the introduction of this thesis, presents an overview of the major guidelines applied at crystal design, and underlines the difficulty encountered in rationally making organic crystals despite our tremendous knowledge. Subsequently, the need for extensive research to derive additional guidelines is required. This thesis begins with the study of supramolecular interactions in organic crystals with five systematic studies dealing with structural equivalences. The results lead to a dataset composed of 73 crystal structures allowing the extraction of additional guidelines/trends that help building our knowledge in crystal design. Secondly, the synthon-based approach and the well-established guidelines are used to design, synthesise and characterise 25 new multicomponent solids, of which 14 potentially mask the bitter taste of four pharmaceutically-relevant molecules. The 1:1 caffeine:benzoic acid cocrystal has been further involved in taste studies to show the potential of organic crystals in taste-masking. The aim of this doctoral research is to present several case studies engaging experimental and theoretical readily-available tools to rationalise experimental observations and guide the experimental research.

Book excerpt: In order to study the as of yet unexplored competition between halogen bonds (XB) and chalcogen bonds (ChB) attached to the same molecular skeleton, a series of 4,7-bis(haloethynyl)benzo-1,2,5-chalcogenadiazoles capable of forming competing ChB and XB dimers were designed, synthesized and analyzed crystallogrpahically. Quantum mechanical calculations were carried out on all targets to provide further insight, and to try to predict the experimental outcome of these competing interactions. The computational results aligned perfectly with crystallographic outcomes, predicting a XB dimer to exist in only one out of twenty-four targets, and correctly calculating halogen bond lengths, angles and intramolecular bending angles. The ChB synthon was further employed in providing a template towards the design of multicomponent crystals. The 4 and 7 position substituted haloethynyl species were replaced with different acceptor molecules, which could then bind to bond donors on co-formers towards the formation of co-crystals. Liquid assisted grinding with fifteen co-formers showed excellent success rates towards the formation of co-crystals, and two crystal structures obtained showed that using carboxylic acid co-formers retains the ChB dimer as intended. Stronger halogen bonds lead to better synthon robustness, which in turn contribute to improved supramolecular synthesis reliability. With this in mind, a triple activation strategy was explored to design a library of triply activated ketones with among the highest reported [sigma]-hole potentials, which are used as a yardstick for the halogen bond donor ability of the iodine atom. Computational calculations were carried out to rank the molecules relative to each other and to benchmark them with literature, which confirmed they outperform previously reported molecules. The targets were subsequently synthesized and crystallized, and the single crystal structures of these targets confirmed that they can indeed form strong halogen bonds as the primary structure-directing motif. The ketones were then used as building blocks for supramolecular co-crystal synthesis. A co-crystal screening with thirty-five co-formers revealed an overall 64% success rate in the formation of co-crystals. A total of nine co-crystal structures were obtained, and these further confirmed the strength and structure directing influence exerted by these highly activated halogen-bond donors. In order to further test the robustness of the triple activation design strategy, a library of triply activated ester targets was assessed computationally and experimentally. Computational calculations showed that they slightly outperform the triply activated ketones in terms of [sigma]-hole potentials and are superior to previously reported molecules. After they were synthesized and crystallized, the crystal structures once again confirmed their ability in forming strong structure directing halogen bonds. To confirm that this new library too can be employed towards the formation of co-crystals, a co-crystal screening with thirty-three co-formers revealed an overall success rate of 76%, even higher than the 64% success rate of the triply activated ketones. A total of eight co-crystal structures were obtained, once again including for all targets with phenazine. Computational calculations were carried out to rationalize the 1:1 vs. 1:2 stoichiometric halogen bonds formed by different targets with phenazine. Results revealed that only very high [sigma]-hole potentials can lead to the formation of a 1:2 stoichiometric halogen bonding synthon, further articulating the importance of the strength of the interaction and its resultant ability to direct complex supramolecular assemblies.
