Book excerpt: Consumption of dry beans is low in the U.S. and the genetic improvement of end-use quality traits of dry beans and the development of new uses for beans offer potential opportunities to increase consumption. This research aimed to 1) characterize a yellow bean diversity panel for seed coat color and cooking time, 2) investigate the GxE and inheritance of seed coat color and cooking time of the diversity panel via genome-wide association (GWA), 3) evaluate the suitability of MI-bred bean varieties for sweet paste qualities, and 4) inform bean breeders of the food industry's needs for bean flour. A Yellow Bean Collection with 295 lines grown in MI and NE were evaluated for seed coat color, post-harvest darkening, and cooking time. Machine-learning was used to develop a procedure to automatically exclude hilum and corona areas in bean images and extract L*a*b* color values from seed coat. Color values, post-harvest darkening, hilum, and corona colors mapped to the ground factor P gene. Cooking time evaluation identified 20 lines that cooked within 20 min in MI and five lines that cooked within 31 min in higher-altitude NE. GWA identified a polygalacturonase gene as a candidate gene for cooking time. GxE was significant for seed coat color and cooking time, and MI-grown seeds were generally darker and faster-cooking than NE-grown. Six white bean varieties and one cranberry genotype were evaluated for sweet bean paste qualities: paste yield, color, stickiness, and flavor. Large-seeded white varieties performed similar or higher for many of the quality traits than Hime, an industry standard, showing their potential as a novel food application in the U.S. An electronic survey was conducted targeting U.S. food industry professionals who use pulse and/or wheat flour. Seventy-five valid responses were collected, and flavor and functionality rather than lectins or gluten contamination were the most important challenges in utilizing bean flour. These studies will be useful in selecting germplasm for favorable qualities to consumers such as bright yellow color, non-darkening after storage, fast cooking time, bean paste qualities, and bean flour attributes.

Book excerpt: Dry bean (Phaseolus vulgaris L.) is the most important legumes for human consumption worldwide and is an important source of protein, vitamins, and micronutrients in the human diet. This research aimed to i) uncover the genetic architecture of yield, Fe bioavailability and seed micronutrient concentration, ii) characterize the genetic control of canning quality traits, and ii) assess the accuracy of genomic prediction models for yield and end-use quality traits. The genetic architecture of yield and seed micronutrient concentration was assessed through a combination of meta-QTL analyses integrating published studies over the last two decades in dry bean. A Gaussian mixture model was used to determine the number of distinct QTL in the meta-QTL analyses. Consistent meta-QTL over different genetic backgrounds and environments were identified, reducing the confidence interval compared with initial QTL. Furthermore, a genome-wide association (GWA) study with 295 lines of the yellow bean collection and 82 yellow recombinant inbred lines identified a major QTL for Fe bioavailability related to the ground factor P gene. A black breeding panel with 415 lines was evaluated for yield and canning quality traits in two growing seasons. Consistent associations for color retention, appearance and texture of canned beans were identified across years. Genomic prediction models provided moderate to high accuracy for end-use quality traits on the yellow and black populations. The genomic prediction accuracy was related to the heritability of each trait, and improvement of accuracy was observed for complex traits when secondary traits were included in the model, while for traits with major QTL, the use of associated markers as fixed effects increased prediction ability. The use of meta-QTL analyses and GWA in this study lays a foundation of the genetic control of yield and end-use quality traits and reveals the potential of genomic prediction for these traits in dry beans.

Book excerpt: The consumer demand for organically produced dry beans (Phaseolus vulgaris L.) continues to increase in the U.S. Organic farmers are faced with production and harvest challenges that impact the processing and end-use quality of dry beans. One major challenge is seed coat mechanical damage induced during harvest, which is a concern for both organic and conventional growers, but the latter has more tools to manage harvest to reduce the impact of the damage. The goal of this research is to improve organic dry bean end-use quality by exploring the genetic variability for mechanically induced seed coat damage, developing enhanced end-use product processing methods, and investigating the variations in seed quality related characteristics. Kidney and black bean market classes are the focus of this work due to their importance in organic production and challenges with end-use quality, especially seed coat splitting and canning quality. Genetic variability for seed coat splitting was evaluated in a collection of 61 kidney beans from three market classes including dark red kidney (DRK), light red kidney (LRK), and white kidney (WK) beans. The beans were harvested under two conditions to induce mild and severe seed coat damage as well as the downstream processing quality of the beans. LRK was found to be more susceptible to seed coat damage than the other two market classes with the highest average seed coat check (SCC) severity score. Genotypes less susceptible to seed coat damage than others were identified within each market class. Seed coat thickness was found not to be a good indicator for SCC severity. SCC severity scores and canning quality appearance scores were found to be negatively correlated. SCC severity score of combine threshed seeds can be used as a selection factor in variety improvement for resilience to mechanical damage. Canning and pouch processing methods were further explored as means to add value to organically produced beans. Pilot-scale pouch and canning processing protocols were developed and updated for use in dry bean breeding programs. The protocols included innovative virtual quality evaluation methods and methods to improve the processing quality of organic dry beans. Both methods were utilized with dry beans from different market classes to test the methods and identify the expected variations in processing quality. A harvest survey was conducted with participation from Michigan bean growers. Nine out of 22 black bean samples had a higher than 20% SCC percentage which is considered unacceptable in the industry standard. In general, variations in mechanical seed coat damage is a result of a series of factors including the genetics of the seeds, the seed moisture content, the environmental condition, and℗ the operation of harvest method. The end-use quality of organic dry beans can be enhanced using the identified genetic variability and improved processing methods.℗
