The use of PH in laxatives has decreased since 1997 when the FDA proposed to withdraw its classification as an OTC drug. PH has been previously evaluated in carcinogenecity studies in 1996. The major route of human exposure to PH is via ingestion, dermal contact, & inhalation of contaminated air originating from process units manufacturing the compound. In this study, the carcinogenic effects of PH were studied in the haploinsufficient p16Ink4a/p19Arf mouse model as an ongoing goal to seek model systems for toxicology & carcinogenesis studies, especially those that can provide mechanistic info. relative to understanding an agent¿s model of action. Glycidol was selected for study in the haploinsufficient p16Ink4a/p19Arf mouse because it was found to be carcinogenic in rats and mice in conventional 2-year rodent studies (NTP, 1990), but was negative in a study in p53+/- mice (Tennant et al., 1999). Book excerpt: BACKGROUND: Benzene is a widely used solvent and is used in the production of many chemicals and gasoline. Benzene is known to cause cancer in laboratory animals and leukemia in humans. We tested benzene in a genetically modified mouse strain that lacks two tumor suppressor genes as part of a study to determine if this mouse model could detect cancer-causing chemicals more rapidly than the standard 2-year rodent bioassay. METHODS: We exposed groups of male or female haploinsufficient p16Ink4a/p19Arf mice by depositing solutions of benzene in corn oil directly into the animals' stomachs through a tube five times per week for 27 weeks. The daily doses were 25, 50, 100, or 200 milligrams of benzene per kilogram of body weight; other animals receiving only corn oil served as the control groups. Tissues from 22 organs were examined for every animal. RESULTS: Exposure to benzene caused malignant lymphomas in male haploinsufficient p16Ink4a/p19Arf mice receiving the greatest dose of benzene. Other effects seen in male mice included bone marrow atrophy, hematopoietic cell proliferation in the spleen, and atrophy of the thymus and lymph nodes. No increased incidences of cancer were seen in female haploinsufficient p16Ink4a/p19Arf mice, though atrophy of the mesenteric lymph nodes was observed. CONCLUSIONS: We conclude that benzene caused malignant lymphoma in male haploinsufficient p16Ink4a/p19Arf mice but not in females.

Book excerpt: BACKGROUND: 3′Azido-3′-deoxythymidine (AZT) is the most widely used chemotherapeutic agent for the treatment of people with acquired immune deficiency syndrome (AIDS) or positive for human immunodeficiency virus (HIV). AZT treatment is also given to prevent transmission of HIV from pregnant mothers to children before or during birth. We tested the effects of AZT on the offspring of female mice (genetically modified to be sensitive to cancer induction) where the mothers were given the drug during pregnancy and the pups were given the drug following birth. METHOD: We exposed groups of haploinsufficient C3B6.129F1-Trp53tm1Brd N12 mice by depositing solutions of AZT in a methylcellulose solvent directly into the animals' stomachs through a tube five times per week for 30 or 45 weeks following birth; in addition, their mothers were exposed to the drug for seven days during pregnancy. Other sets of mothers and pups received only the methylcellulose solvent and served as the control groups. Tissues from 34 organs were examined for every animal. RESULTS: Exposure to AZT caused increases in the rates of liver cancer in the male pups after 45 weeks. In addition, there were occurrences of a few malignant lymphomas in both male and female pups exposed to AZT in the 30-week studies. CONCLUSIONS: We conclude that AZT caused liver cancers in male pups exposed to AZT before and following birth. Malignant lymphomas in male and female pups may have been related to AZT exposure before and following birth.
