Book excerpt: The eastern subterranean termite, Reticulitermes flavipes, is the most widely dispersed termite in North America. The genus Reticulitermes spp. is responsible for 80% of total termite damage caused to urban structures each year. Little is known about the genetic structure of termites, particularly at the colony level. Evidence for what genetically defines a termite colony is a hotly debated topic in current literature due to the implications such findings would have regarding current lawsuits against pest control operations. Information on termite genetic structure is sparse. In this study, the genetic variation and gene flow among Texas populations of R. flavipes at the statewide level and city level was examined. A 324-337 base pairs segment of the mtDNA, AT-rich region was a polymerase chain reaction amplified from 104 different termite specimens from 12 Texas cities. The DNA extracts were then subjected to PCR amplification using specific primers and it was then sequenced. Using the sequence data and appropriate statistical measures it was found that, at the statewide level, nucleotide and haplotypic diversity is low. Gene flow was found to be low on a statewide basis. At the city level nucleotide and haplotypic diversity was high. The findings of this study provide insights into termite genetic structure.

Book excerpt: Subterranean termite species in the genus Reticulitermes are ecologically and economically important in the United States. At least six species of the genus Reticulitermes are native from North American forests where they feed on cellulose materials. In urban environments they feed upon dead wood, and therefore infest and destroy man-made wooden structures. Missouri is considered to have a moderate risk for termite infestations when compared with the states of the southeast part of the country. Despite their ecological and economic importance, Missouri's subterranean termite faunal composition and geographic distribution is not well known. Diversity, geographic distribution, and genetic variability of Reticulitermes species within Missouri were studied from approximately 600 samples of termite colonies collected during 2004 and 2005 in nine conservation areas, nine cities located near these conservation areas, and from home infestations occurring at many different locations within the state. Reticulitermes flavipes (Kollar), Reticulitermes virginicus (Banks), Reticulitermes tibialis Banks and Reticulitermes hageni Banks were found occurring in Missouri. Reticulitermes flavipes and R. hageni were the most abundant species. Reticulitermes flavipes was found to be more abundant in urban environments and home infestations than in woodlands. Reticulitermes hageni also occurs throughout the state but was found to be more abundant in woodlands than in urban environments. This study updated the inventory of Reticulitermes species occurring within the state, provided new records of occurrence of the species in geographical locations not known before, and provided a comprehensive morphometric study of soldiers of Reticulitermes species occurring in both urban and woodlands of Missouri. Ecological studies as well as integrated management may benefit from the information provided in this study.

Book excerpt: Abstract: Reticulitermes flavipes laboratory colonies survived orphaning by producing sexually functional neotenic reproductives. The [female] : [male] neotenic ratio in orphaned colonies was 2:1 after 10-11 months (N =22 colonies) and 6:1 after 16-17 months (N = 21 colonies). The majority of neotenics in orphaned colonies were of the ergatoid form, although a much greater percentage of nymphoid reproductives were found at the 16-17 month re-census than the 10-11 month re-census. Mortality was high for both orphaned and control colonies (in which the primary reproductive pair was retained) at both re-census times, but all surviving colonies showed at least a 400% increase in total colony size (excluding eggs and reproductives) when compared to initial colony size. Egg development (oogenesis) of all Reticulitermes flavipes female reproductives in laboratory colonies was assessed by means of trypan blue dye uptake in the middle growth region of ovarioles (evidence of vitellogenesis) and/or the production of terminal oocytes >̲ 0.2 mm in length. Oogenic activity was evident in 94% of the neotenic females (N(ergatoid) = 77, N(nymphoid) = 1) found in colonies 10-11 months after orphaning and in 99% of the neotenic females (N(ergatoid) = 102, N(nymphoid) = 18) found in colonies 16-17 months after orphaning. In control colonies of both time treatments, 100% of the primary reproductives (N(10-11 month) = 5, N (16-17 month) = 9) displayed evidence of egg development.. Results obtained from the laboratory colonies were compared with R. flavipes neotenic females found in a population occupying a severed tree stump. This wild population contained more male neotenics (N = 430) than female neotenics (N =316). A total of 33 nymphoid females was dissected, and all of these neotenics were undergoing oogenesis. It can be assumed with 95% confidence that at least 288 (91%) of the 316 female neotenics were also undergoing oogenesis.
