Book excerpt: "Genetic instability leads to the accumulation of many changes necessary to transform the stable genome of a normal cell into an unstable genome. Such a transformation can be the cause of radiation mutagenesis, carcinogenesis and aging - the main long-term effects of ionizing radiation, UV light, and other harmful environmental factors. A new quantitative assessment of the genetic instability was suggested, taking into account the delay in the formation of colonies by cells surviving after exposure to harmful agents. It was demonstrated that the genetic instability of yeast cells is mainly determined by cell ploidy, but not by the sigmoid shape of the survival curve or the cell's ability of post-irradiation recovery. New experimental data confirm the participation of recovery processes in the manifestation of the relative biological effectiveness of densely ionizing radiation both for cell inactivation and for radiation-induced genetic instability. A number of universal patterns of synergic interactions are demonstrated, which did not depend on the used biological test and objects nor the interacting physical and chemical factors. Potential significance of synergy at low intensities of physical and chemical environmental factors is proved. Original data on the influence of cyclic heating and cooling on cell survival are obtained. It is concluded that osmotic homeostasis of cells, along with DNA, is an important target responsible for cell viability under the damaging action of various agents. Data on the favorable effect of the increased natural background of ionizing radiation are discussed. The manifestations of radiation hormesis were revealed for various radiation accidents, professional and medical exposure. The results obtained demonstrate that the action of high-energy ionizing radiation is accompanied by irradiation with UV light that occurs when charged particles move at a speed exceeding the speed of light in a given medium"--

The results of the NCI investigation were used a primary resource for communicating with the public about the cancer risks near the nuclear facilities. However, this study is now over 20 years old. The U.S. Nuclear Regulatory Commission requested that the National Academy of Sciences provide an updated assessment of cancer risks in populations near USNRC-licensed nuclear facilities that utilize or process uranium for the production of electricity. Analysis of Cancer Risks in Populations near Nuclear Facilities: Phase 1 focuses on identifying scientifically sound approaches for carrying out an assessment of cancer risks associated with living near a nuclear facility, judgments about the strengths and weaknesses of various statistical power, ability to assess potential confounding factors, possible biases, and required effort. The results from this Phase 1 study will be used to inform the design of cancer risk assessment, which will be carried out in Phase 2. This report is beneficial for the general public, communities near nuclear facilities, stakeholders, healthcare providers, policy makers, state and local officials, community leaders, and the media.

However, cure of cancer is still hampered by the inherited capacity of tumors to become resistant to standard therapies, to metastasize from their initial location and to proliferate in other tissue compartments. Radiotherapy is one of the main treatment modalities to achieve locoregional tumor control. However, the treatment of distant metastases further remains to be a challenge. In this special topic we are interested to elucidate immunological aspects which are initiated and affected by radiotherapy. We also aim to describe the development of innovative immunological strategies from a preclinical stage to clinical application which could be combined with standard radiotherapeutic approaches. A special interest will also deal with the effects of radiotherapy on tumor initiating cells as well as on the tumor microenvironment. Last but not least the effects of different irradiation sources and qualities such as photones, protones and heavy ions will be analyzed with respect to immunological outcome.
