Book excerpt: Significant recent advances in cell culture technology now permit a detailed biochemical and molecular analysis of differentiation in both normal and tumor cells. These studies are important in attempting to understand the complex factors involved in normal growth and development, as well as the abnormalities associated with carcinogenesis. Mechanisms of Differentiation, Volumes I and II, is comprised of review chapters addressing various topics of current interest in this important area of research. Topics discussed include genes controlling differentiation, changes in gene expression during differentiation, induction of differentiation, induction of differentiation as a mode of action of chemotherapeutic agents, and the effect of cell shape, growth factors and differentiation modulating agents on the differentiated cell phenotype. Mechanisms of Differentiation is valuable to researchers involved in differentiation and development, carcinogenesis, cell biology, chemotherapy, and immunology.

Book excerpt: This series was established to create comprehensive treatises on specific topics in developmental biology. Such volumes serve a useful role in developmental biology, which is a very diverse field that receives contributions from a wide variety of disciplines. This series is a meeting ground for the various practi tioners of this science, facilitating an integration of heterogeneous information on specific topics. Each volume is comprised of chapters selected to provide the conceptual basis for a comprehensive understanding of its topic as well as an analysis of the key experiments upon which that understanding is based. The specialist in any aspect of developmental biology should understand the experimental back ground of the specialty and be able to place that body of information in context, in order to ascertain where additional research would be fruitful. The creative process then generates new experiments. This series is intended to be a vital link in that ongoing process of learning and discovery.

Available in PDF, EPUB and Kindle. Book excerpt: Epigenetic Gene Expression and Regulation reviews current knowledge on the heritable molecular mechanisms that regulate gene expression, contribute to disease susceptibility, and point to potential treatment in future therapies. The book shows how these heritable mechanisms allow individual cells to establish stable and unique patterns of gene expression that can be passed through cell divisions without DNA mutations, thereby establishing how different heritable patterns of gene regulation control cell differentiation and organogenesis, resulting in a distinct human organism with a variety of differing cellular functions and tissues. The work begins with basic biology, encompasses methods, cellular and tissue organization, topical issues in epigenetic evolution and environmental epigenesis, and lastly clinical disease discovery and treatment. Each highly illustrated chapter is organized to briefly summarize current research, provide appropriate pedagogical guidance, pertinent methods, relevant model organisms, and clinical examples. Reviews current knowledge on the heritable molecular mechanisms that regulate gene expression, contribute to disease susceptibility, and point to potential treatment in future therapies Helps readers understand how epigenetic marks are targeted, and to what extent transgenerational epigenetic changes are instilled and possibly passed onto offspring Chapters are replete with clinical examples to empower the basic biology with translational significance Offers more than 100 illustrations to distill key concepts and decipher complex science

Available in PDF, EPUB and Kindle. Book excerpt: Many inheritable changes in gene function are not explained by changes in the DNA sequence. Such epigenetic mechanisms are known to influence gene function in most complex organisms and include effects such as transposon function, chromosome imprinting, yeast mating type switching and telomeric silencing. In recent years, epigenetic effects have become a major focus of research activity. This monograph, edited by three well-known biologists from different specialties, is the first to review and synthesize what is known about these effects across all species, particularly from a molecular perspective, and will be of interest to everyone in the fields of molecular biology and genetics.

Book excerpt: The neural crest is a remarkable embryonic population of cells found only in vertebrates and has the potential to give rise to many different cell types contributing throughout the body. These derivatives range from the mesenchymal bone and cartilage comprising the facial skeleton, to neuronal derivatives of the peripheral sensory and autonomic nervous systems, to melanocytes throughout the body, and to smooth muscle of the great arteries of the heart. For these cells to correctly progress from an unspecifi ed, nonmigratory population to a wide array of dynamic, differentiated cell types-some of which retain stem cell characteristics presumably to replenish these derivatives-requires a complex network of molecular switches to control the gene programs giving these cells their defi ning structural, enzymatic, migratory, and signaling capacities. This review will bring together current knowledge of neural crest-specifi c transcription factors governing these progressions throughout the course of development. A more thorough understanding of the mechanisms of transcriptional control in differentiation will aid in strategies designed to push undifferentiated cells toward a particular lineage, and unraveling these processes will help toward reprogramming cells from a differentiated to a more naive state. Book excerpt: The fate of a cell is largely determined by the unique patterns of gene expression found within it. Complex biological machinery exists within each cell to manipulate chromatin state, and ultimately control gene expression. Developmental processes such as cellular differentiation require very specific chemical signals and environmental conditions. These serve as triggers to put the chromatin modification schemes that produce the resultant patterns of differential gene expression into action, leading to the formation of the cell type of interest. My thesis work is an in depth study of the link between chromatin modification, gene expression, and the unique genetic signatures that characterize distinct cells on unicellular and multi-cellular levels. On the multi-cellular level, I have examined histone modification patterns for their effects on gene activation and repression during human embryonic stem cell differentiation. On the unicellular level, I have worked with a variety of cell types to ascertain the degree of individuality that exists between single members of relatively homogenous cell groups while simultaneously looking for housekeeping gene expression signatures that can be used to classify each cell type into a unique group. To further elucidate the patterns of gene expression found within cell groups and the single cells that comprise them, I have worked to develop new computational methods that produce visual aids to elucidate gene expression signatures of single cells and cell groups.

Available in PDF, EPUB and Kindle. Book excerpt: Genetic Expression in the Cell Cycle provides an understanding of the molecular mechanisms that govern the expression of genetic information during the cell cycle. The initial five chapters describe the intimate relationships between the supramolecular complexes that form the basic structure of chromatin. Emphasis is placed on the dynamics of cycle-dependent changes in the structural organization of some of these components. Subsequent chapters demonstrate that small nuclear RNAs (SnRNA) are actively involved in gene regulation in eukaryotic cells; discuss the relationship between cell cycle regulation in the yeast Saccharomyces cerevisiae and transcription of ribosomal RNA genes; and describe the use of conditional lethal mutants to study the regulation of the cell cycle of eukaryotic cells. The remaining chapters discuss the concepts and methodologies employed to isolate and study specific cell cycle mutants of S. cerevisiae; the antiproliferative effect of interferon on cultured human fibroblasts; and the role of cell membrane and related subcellular elements in the control of proliferation, differentiation, and cell cycle kinetics.
