In recent years, the development of multifunctional nanomaterials has provided a new diagnosis and treatment platform to combat cancer. Polymer-inorganic nanomaterials with novel structures such as bowl-shaped/Janus/core-shell have drawn much attention owing to their diversity in composition or asymmetry in structure. More importantly, imparting unique optical, electrical, and magnetic properties to these nanocomposites can further extend their function repertoire. However, to fulfill this vision, fundamental understandings regarding strategies of precise synthesis, mechanisms of structure formation, in vivo synergistic effects in bioapplications, and biosafety of these materials are needed. Besides, nanomaterials with novel structures are well positioned for imaging-guided cancer theranostic. On one hand, nanomaterials themselves are suitable for imaging because of their intrinsic properties such as fluorescent or magnetic properties. On the other hand, nanomaterials can serve as functional platforms that integrate various therapeutic modalities including photothermal therapy, chemodynamic/ion-interference therapy, photodynamic therapy, and cuproptosis to efficiently kill cancer cells. This Research Topic aims at collecting works about synthesis, and biomedical applications of polymer/mesoporous inorganic nanomaterials, especially in the aspect of novel synthetic approaches for fabricating nanomaterials with unique structures. Additionally, we hope that in-depth research articles on this topic can provide insights into the mechanism of nanomaterials acting in cancer diagnosis and therapy. These include the mechanisms of customized drug load/release and synergistic effects in theranostics of these materials. Meanwhile, elucidations of key proteins’ roles in cancer development are also anticipated. Lastly, we hope that this topic can brew new ideas for the adaption of nanomaterials as platforms that allow for multimodal therapeutic modalities. The current Research Topic centers on the design, precise synthesis, and biomedical applications of nanomaterials. It aims to cover novel and promising research trends in nanomaterials with different morphology for cancer theranostics. Manuscripts from the following aspects, but not limited to, are welcomed: • Tailoring of asymmetrically structured (bowl-shaped, Janus, Yin Yang-like) polymer-inorganic nanomaterials; • Inorganic functional nanocrystals and functionalized mesoporous nanomaterials; • Design and synthesis of functional biomaterials, including lipids, polymers, and 2D materials • Non-viral DNA/mRNA delivery or drug/molecular inhibitor delivery; • Synthesizing biomaterials with novel nanostructures such as bowl-shaped, core-shell, spherical, Janus, and quantum dots; • Conquering drug resistance issues, tumor metastasis, and recurrence, as well as designing combination nanomedicines; • Dissecting the role of menin in prostate cancer and breast cancer: crosstalk between menin and AR signaling; • Multi-stimulus-responsive drug release and biological molecules.

Book excerpt: This dissertation describes the syntheses of functional polymeric biomaterials from natural monomeric building blocks for application specific to vectors for gene delivery. Chapter 1 of this dissertation introduces and discusses the various chemical and biological challenges of synthetic polymeric vectors for gene delivery. In particular, Chapter 1 examines various barriers at the cellular level for efficient gene transfer. In addition, chemical strategies to overcome such biological barriers are also discussed. Finally, a variety of synthetic vectors with various design and chemical diversities are presented. In particular, incorporation of an environmentally responsive bond into synthetic vectors is introduced as one of the main design characteristics necessary for overcoming multiple biological challenges. Overall, the introductory chapter attempts to illustrate the sensitive balances and complexities observed in the field of gene delivery. Chapter 2 of this dissertation describes the influence of structural modifications on saccharide-peptide hybrid copolymer (SPHC) as a synthetic vector for plasmid DNA and short interfering RNA delivery. SPHCs with various cationic charge densities, buffering capabilities, and cross-linking densities were prepared and consequences of these modifications on SPHCs are discussed. Chapter 3 of this dissertation describes the surfactant-free synthesis of bioreducible and biocompatible cationic nanogels intended for siRNA delivery. Nanogels were prepared by cross-linking the linear polymeric precursors in combination with polar and non-polar organic solvents without the addition of surfactants. The presence of amine side chains off the polymer backbone were determined to be crucial for efficient siRNA complexation. Our results (size, stability, environment responsiveness, and toxicity profile) clearly reveal the potential of this nanogel to be applied as a nano-colloidal drug carrier. Chapter 4 describes an efficient living ring-opening polymerization (ROP) of a permethoxylated-caprolactone [(OMe)CL] catalyzed by yttrium(III) isopropoxide for development of degradable protein-resistant polymers [P(OMe)CL]. The lactone monomer was efficiently prepared from a reduced sugar, D-dulcitol. A series of block copolymers consisting of permethoxylated lactone and [varepsilon]-caprolactone [P(OMe)CL-b-PCL] were synthesized and characterized. Finally, surface plasmon resonance (SPR) sensorgrams demonstrated that both P(OMe)CL and the P(OMe)CL-b-PCL block copolymers exhibit excellent resistance to protein adsorption.

Attempts to design ideal biomaterials for each specific application have led to an enormous increase in the number of polymeric biomaterials. Different biomedical applications require specific functionality of biomaterials. Unfortunately, the limited versatility of currently available biodegradable polymers fails to meet the wide range of requirement for biomedical applications. Therefore, a methodology of functional biomaterials development would be beneficial to the biomaterial field. It has been proved that citric acid can be utilized as a key element in the development of novel biodegradable polymers. This work presents the methodology of three citric acid based functional biomaterials design and application to meet the multifaceted needs of tissue engineering and theranostic therapy. Firstly, urethane doped biodegradable polyester with dual crosslinking mechanism is developed as scaffold materials to fulfill the wide range of mechanical properties of soft tissue engineering. We hypothesize that doping of urethane bond can greatly increase the mechanical strength and elasticity, and dual crosslinking mechanism confers parameters to manipulate mechanical properties over a wide range. Secondly, novel family of biodegradable photoluminescent polymers (BPLPs) is developed meet the increasing needs of marriage between biodegradable polymers and bioimaging. We hypothesize that the intrinsic strong fluorescence of BPLPs can be exploited as theranostic probes, and provide possibility of real-time monitoring for implants. Lastly, BPLPs are doped with urethane bond (UBPLPs) to achieve an ultra strong elastomer with strong fluorescence. We hypothesize that the greatly increased mechanical strength makes UBPLPs suitable for engineering cardiovascular tissue. It also can improve the stability of nanoparticles for theranostic application. To test these hypotheses, we first developed the methodology of all three functional biodegradable polymers. The synthesis, characterization, fabrication, and application of those polymers were discussed. The relationship between materials chemistry and phenomenon were devoted to the understanding of basic biomaterials science. The results presented in this work show that all three functional polymers succeed to fulfill the requirements of specific biomedical applications. The designing strategies can serve as a guide for developing of new citric acid based biodegradable polymers. These enabling new biodegradable polymers are able to address many of the existing challenges in tissue engineering, and theranostic applications.
