Among many types of biosensors available today, piezoelectric (PZT) class of materials are becoming very popular. In this thesis, Zinc Oxide nanowire PZT biosensor was fabricated and characterized to detect the presence of fungi which has some huge economic implications in US agriculture industry. Zinc Oxide nanowires were synthesized in a mass scale via wet solution method in a controlled temperature and growth environment. Different substrates including glass, indium tin oxide, and gold coated silicon substrates were utilized to grow the nanowires followed by layering with silane and subsequently etching them. The results show that the nanowires were grown homogenously on gold coated silicon wafers with cylindrical structures. The ideal morphology of the nanowires was found to be dependent on: incubation time, incubation temperature, and substrate material. Substrate catalyst was also varied from Au & Pd to pure Au which showed significant improvement in producing the nanowires. A systematic variation of hours was implemented from: 3, 5, 7, 9, 11, and 13 hours. Zinc Oxide nanowire features such as length, diameter, and aspect ratio were quantified through SEM micrographs. Linear increase in height, diameter, and aspect ratio was observed up to 13 hours along with density. The optimal condition for nanowire growth was determined at: 80 °C and 5 hours. Energy dispersive spectroscopy aided in generating presence of specific elements on the biosensor. Raman helps in verifying chemical composition of the device. Both Raman and EDS spectroscopy aided in enhancement and individualization of the biosensor at different proposed parameters. Keithley readings represented series of current-voltage (I-V) measurements under different forward biased voltages. The response of nanowires from these I-V measurements show a diode-like response. Next, nanowire displacement patterns of fungi, Fusarium proliferatum (F. proliferatum) were studied by I-V measurements. When I-V measurements were conducted on PZT nanowires in the presence of F. proliferatum a strong association from microbe attachment and growth was observed showing an increase in switch-on voltage with a 2V sweep. It is speculated that the observed high resistance is a result of mechanical movement of fungi on the piezoelectric device. Future studies will be designed to investigate this phenomenon. These results indicate that by simply reading the characteristic current-voltage measurement, one can better evaluate microbe pattern of displacement and maturation. Future application of this nanowire platform can characterize distinct displacement signature of disease carrying organism much more efficiently.

Abstract: Abstract of thesis entitled FABRICATION AND CHARACTERIZATION OF ZINC OXIDE (ZnO) NANOSTRUCTURES Submitted by Leung Yu Hang For the degree of Master of Philosophy at The University of Hong Kong in January 2006 Since the discovery of carbon nanotubes by Iijima in 1991, increasing research effort has been devoted to the synthesis and characterization of nanostructured materials. Materials at this scale exhibit novel properties which cannot be found in their bulk form. In silicon nanowires, for example, the bandgap of Si nanowires increases from 1.1 eV to 3.7 eV when the nanowire diameter decreases from 7 nm to 1.3 nm. In recent years, nanostructures of various semiconductors like Si, gallium arsenide, gallium nitride, and zinc oxide have been demonstrated. Among them, ZnO has been recognized as a promising material for a variety of applications in the field of photonics, optoelectronics, gas sensing, field emission, and piezoelectrics. To realize the incorporation of ZnO nanostructures into real life applications, lots of work need to be done, including synthesis of the nanostructures, and understanding of their properties. In this study, a number of ZnO nanostructures (tetrapods, nanorods, nanoribbons/combs, multipods) were fabricated by thermal evaporation and hydrothermal synthesis methods. The morphologies and, structural and optical properties of the resultant nanostructures were then characterized by various techniques, such as scanning electron microscopy, transmission electron microscopy, selected area electron diffraction, energy dispersive X-ray spectroscopy, X-ray diffractometry, photoluminescence and electron paramagnetic resonance. It was observed that fabrication conditions could significantly affect the properties of the nanostructures. All nanostructures exhibited one UV peak and one broad peak in the visible spectrum. The major difference is that the nanostructures fabricated by thermal evaporation exhibited green PL emission while the nanorods synthesized by hydrothermal method gave yellow emission. Green emission was observed even without the presence of the EPR signal at g 1.96 in some samples. This result contradicts the commonly cited oxygen vacancy hypothesis of green emission of ZnO. Obviously, that hypothesis does not apply to all ZnO samples. The nanorod sample with yellow emission also exhibited strong EPR signal similar to the green emitting sample. A possible explanation for the obtained results is that there are two different deep levels responsible for the green and yellow emission respectively. Stimulated UV emission of three different nanostructures (tetrapods, nanoribbons/combs, and nanorods) was studied by time-resolved photoluminescence (TRPL). It was observed that different nanostructures exhibited different lasing thresholds, emission delay times and decay times. The difference in the lasing behaviors of the nanostructures is likely due to the difference in morphology, mode of cavity, or even native defects present in the material. Book excerpt: In this thesis, a systematic study has been carried out on the synthesis, characterization and device fabrication of piezoelectric ZnO nanstructures. The achieved results are composed of the following four parts. Firstly, through a systematic investigation on the Sn-catalyzed ZnO nanostructure, an improved understanding of the chemical and physical process occurring during the growth of hierarchical nanostructures has been achieved. Decomposed Sn from SnO2 has been successfully demonstrated and proved to be an effective catalyst guiding the growth of not only aligned ZnO nanowires, but also the hierarchical nanowire-nanoribbon junction arrays and nanopropeller arrays. During the vapor-liquid-solid (VLS) catalyzing growth process at high temperature, Sn in the liquid state has been proved to be able to guide the growth of nanowires and nanoribbons in terms of growth directions, side facets, and crystallographic interfaces between Sn and ZnO nanostructures. Secondly, using pure ZnO as the only source material, by precisely tuning and controlling the growth kinetics, a variety of hierarchical polar surface dominated nanostructures have been achieved, such as single crystal nanorings, nanobows, nanosprings and superlattice nanohelices. High yield synthesis of ZnO nanosprings over 50% has been successfully obtained by mainly controlling the pre-pumping level associated with the partial pressure of residual oxygen during the vapor-solid growth process. The rigid superlattice nanohelices of ZnO have been discovered, which is a result of minimization of the electrostatic energy induced by polar surfaces. The formation process of the nanohelix has been systematically characterized. Thirdly, two new strategies have been successfully developed for fabricating ZnO quantum dots and synthesis of ZnO nanodiskettes and nanotubes. The formation process is based on a common concept of self-assembly. Finally, a series of devices and applications studies based on several piezoelectric ZnO nanostructures, such as nanobelts, nanopropellers and nanohelices, have been carried out utilizing the electro-mechanical resonance, bio-surface functionalization, devices fabrication and electrical characterization. Individual nanobelt and nanohelix based nanodevices have been successfully fabricated for applications in chemical and biological sensing. The study opens a few new areas in oxide nanostructures and applications.

AlGaN/GaN based HEMTs were used to detect kidney injury molecule-1 (KIM-1), an important biomarker for early kidney injury detection. The HEMT gate region was coated with KIM-1 antibodies and the HEMT source-drain current showed a clear dependence on the KIM-1 concentration in phosphate-buffered saline (PBS) solution. The limit of detection (LOD) was 1ng/ml using a 20 micron by 50 micron gate sensing area. This approach shows a potential for both preclinical and clinical disease diagnosis with accurate, rapid, noninvasive, and high throughput capabilities. The rest of this dissertation includes ZnO band edge electroluminescence from N- implanted ZnO bulk, and the investigation of cryogenic gold Schottky contact on GaAs for enhancing device thermal stability.

Book excerpt: Fabrication and characterization of a new kind of pressure sensor using self-assembly Zinc Oxide (ZnO) nanowires on top of the gate of a Metal-Oxide-Semiconductor Field-Effect Transistor (MOSFET) is presented. Self-assembly ZnO nanowires were fabricated with a diameter of 80 nm and 800 nm height (80:8 aspect ratio) on top of the gate of the MOSFET. The sensor showed a 110% response in the drain current due to pressure, even with the expected piezoresistive response of the silicon device removed from the measurement. The pressure sensor was fabricated through low temperature bottom up ultrahigh aspect ratio ZnO nanowire growth using anodic alumina oxide (AAO) templates. The pressure sensor has two main components: MOSFET and ZnO nanowires. Silicon Dioxide growth, photolithography, dopant diffusion, and aluminum metallization were used to fabricate a basic MOSFET. In the other hand, a combination of aluminum anodization, alumina barrier layer removal, ZnO atomic layer deposition (ALD), and wet etching for nanowire release were optimized to fabricate the sensor on a silicon wafer. The ZnO nanowire fabrication sequence presented is at low temperature making it compatible with CMOS technology.
