Available in PDF, EPUB and Kindle. Book excerpt: In many countries producers have been growing varieties of hard white winter (HWW) wheat since decades. The cause of concern is most varieties of HWW wheat are susceptible to pre-harvest sprouting (PHS) which affects grain quality. Environmental conditions like high humidity, precipitation, heavy dew and hormonal activity at physiological maturity stimulate PHS in HWW. To alleviate these conditions research was carried out at KSU. KS01HW163-4, a sprouting tolerance line was crossed with Heyne, a sprout susceptible cultivar. A total of 224 doubled haploid (DH) lines thus produced were phenotyped in the present study through experiments conducted in controlled environments. The objectives of this research were to (i) characterize and phenotype the doubled haploid lines for PHS in controlled environments; (ii) understand the impact of growth environment (high temperature and/or drought) and; (iii) impact of exogenous application of growth hormones on tolerance to pre-harvest sprouting in the parental lines of the doubled haploid population. The phenotypic data collected from this research will be ultimately combined with the genotypic data to identify DNA markers related to PHS tolerance and provide DNA markers for marker assisted selection. Based on my results of the germination percentages from the 224 DH lines, the population was distributed as susceptible, and tolerant to PHS showing a bimodal distribution and X[superscript]2 analysis indicating a complimentary gene action. From the study of the influence of environmental factors on PHS, my results confirmed a definite influence of stress on sprouting. Under optimum temperature (OT), KS01HS163-4 was tolerant to PHS, but at HT and/or drought it became susceptible to PHS. Growth under stressed conditions changed the tolerance levels to PHS. Seed dry-weight, and harvest index were also influenced negatively due to stress. Therefore multi-location tests must be conducted with variable environments to test the stability of a variety to PHS. From the study of the influence of phytohormones on PHS, the results suggest that tolerance to sprouting was seen in seeds from plants sprayed with abscisic acid (ABA) and paclobutrazol (GA-inhibitor) treatments where as those from gibberellic acid (GA) treatment showed susceptibility to sprouting.

This book was released on 2018 with total page 232 pages. Available in PDF, EPUB and Kindle. Book excerpt: Preharvest sprouting (PHS), germination of mature grain while still on the mother plant, occurs when conditions become cool and wet before harvest. The hydrolytic enzyme ?-amylase, induced during germination, mobilizes starch into simple carbohydrates to fuel seedling growth. Because this enzyme activity in flour causes poor end-use quality, sprouted grain sells at a severe discount. The falling number (FN) test measures ?-amylase activity in wheat meal or flour in the wheat industry. Seed dormancy, the inability to germinate even under favorable conditions, contributes about 60--80 % of genetic PHS tolerance. Red wheat varieties have higher seed dormancy and PHS tolerance than white. To improve white wheat, a genome-wide association study (GWAS) examined the genetic architecture of PHS tolerance in 469 soft white winter wheat accessions. Based on FN after natural or artificial rain, the GWAS identified 9 QTL (QFN.wsu), of which 4 co-localized with known PHS QTL and 3 with known FN/quality QTL. Based on visible sprout in spike-wetting tests, the GWAS identified 34 QTL (QPHS.wsu), of which 19 co-localized with known PHS loci and genes such as MOTHER OF FLOWERING TIME (TaMFT) and mitogen-activated protein kinase kinase 3 (TaMKK3-A). PHS tolerance in white wheat can result from higher sensitivity to the seed dormancy-inducing hormone abscisic acid (ABA). Enhanced Response to ABA (ERA8 ) is a semi-dominant ABA hypersensitive mutant, resulting in increased seed dormancy and PHS tolerance in the soft white spring wheat 'Zak'. The ERA8 locus was mapped to a large region of chromosome 4A relative to mutagen-induced SNPs in a Zak/ZakERA8 backcross population using bulk segregant analysis (BSA) of exome sequence from BC 3F2:3 wild-type and mutant DNA. Fine mapping using mutagen-induced SNPs in additional backcross lines localized ERA8 to a 4.5 Mb region containing 70 predicted genes. The only mutagen-induced coding region mutation strongly linked to ERA8 (LOD 16.51) resulted in a missense mutation in MKK3-A, a gene involved in Arabidopsis ABA signaling. Natural variation in wheat and barley MKK3 was previously shown to control seed dormancy and PHS tolerance.

A shift in markets to the Pacific Rim has resulted in the need for white-grained cultivars, but white wheat germplasm has traditionally had poor pre-harvest sprouting resistance, presenting a drawback for the potential development of white wheat cultivars adapted to Saskatchewan. Seed dormancy is the main factor responsible for conferring pre-harvest sprouting tolerance to wheat grain. This report describes a project to develop hard white bread wheat cultivars with high seed dormancy levels. Standard small-plot techniques were used to evaluate hard white wheat experimental lines. Results of breeding efforts that started with testing of 280 advanced lines are presented.

Book excerpt: In wheat (Triticum aestivum) global production, pre-harvest sprouting (PHS) plays an important role for all processes including end-use quality. Rain and high humidity encourages PHS in the field. The U.S. domestic and export markets presently have high demand for white wheats, which tend to be more susceptible to PHS than red wheats. To allow the U.S. to reliably export white wheats to world markets, PHS tolerance will be essential.
