Available in PDF, EPUB and Kindle. Book excerpt: Muscarinic acetylcholine receptors have played a key role in the advancement of knowledge of pharmacology and neurotransmission since the inception of studies in these fields, and the effects of naturally occurring drugs acting on muscarinic receptors were known and exploited for both therapeutic and non-therapeutic purposes for hundreds of years before the existence of the receptors themselves was recognized. This volume presents a broad yet detailed review of current knowledge of muscarinic receptors that will be valuable both to long-time muscarinic investigators and to those new to the field. It describes the detailed insights that have been obtained on the structure, function, and cell biology of muscarinic receptors. This volume also describes physiological analyses of muscarinic receptors and their roles in regulating the function of the brain and of a variety of peripheral tissues. This volume shows how the study of muscarinic receptors continues to provide new and surprising insights not just to the cholinergic system but to the broad areas of neurobiology, cell biology, pharmacology, and therapeutics.

Book excerpt: A Pharmacology Primer: Techniques for More Effective and Strategic Drug Discovery, Fifth Edition features the latest ideas and research regarding the application of pharmacology to the process of drug discovery. Written by well-respected pharmacologist, Terry P. Kenakin, this primer is an indispensable resource for all those involved in drug discovery. This updated edition has been thoroughly revised to include material on quantifying drug efficacy through bias and cluster analysis, the impact of molecular dynamics and protein structural analysis, the real time kinetic analysis of drug effect, virtual screening for new drug chemical scaffolds, and much more. With full color illustrations and new examples throughout, this book remains a top reference for all industry and academic scientists that is also ideal for students directly involved in drug discovery or pharmacologic research. Highlights changes surrounding strategies for drug discovery, providing a comprehensive reference and featuring advances in the methods involved Includes multiple new sections, such as development and utilization of models in pharmacology, de-orphanization of new drug targets, predicting impact of disease on drug pharmacokinetics, and the impact of enzyme kinetics on drug-drug interactions Illustrates the application of rapid inexpensive assays to predict activity in the therapeutic setting, showing data outcomes and the limitations inherent in interpreting this data

Book excerpt: Experimental Design and Statistical Analysis for Pharmacology and the Biomedical Sciences A practical guide to the use of basic principles of experimental design and statistical analysis in pharmacology Experimental Design and Statistical Analysis for Pharmacology and the Biomedical Sciences provides clear instructions on applying statistical analysis techniques to pharmacological data. Written by an experimental pharmacologist with decades of experience teaching statistics and designing preclinical experiments, this reader-friendly volume explains the variety of statistical tests that researchers require to analyze data and draw correct conclusions. Detailed, yet accessible, chapters explain how to determine the appropriate statistical tool for a particular type of data, run the statistical test, and analyze and interpret the results. By first introducing basic principles of experimental design and statistical analysis, the author then guides readers through descriptive and inferential statistics, analysis of variance, correlation and regression analysis, general linear modelling, and more. Lastly, throughout the textbook are numerous examples from molecular, cellular, in vitro, and in vivo pharmacology which highlight the importance of rigorous statistical analysis in real-world pharmacological and biomedical research. This textbook also: Describes the rigorous statistical approach needed for publication in scientific journals Covers a wide range of statistical concepts and methods, such as standard normal distribution, data confidence intervals, and post hoc and a priori analysis Discusses practical aspects of data collection, identification, and presentation Features images of the output from common statistical packages, including GraphPad Prism, Invivo Stat, MiniTab and SPSS Experimental Design and Statistical Analysis for Pharmacology and the Biomedical Sciences is an invaluable reference and guide for undergraduate and graduate students, post-doctoral researchers, and lecturers in pharmacology and allied subjects in the life sciences.

Available in PDF, EPUB and Kindle. Book excerpt: Some important constraints of anesthesia must be taken into consideration when the pharmacological properties of modern anesthetics are discussed. The most imp- tant of these could be that the target effect be achieved preferably within seconds, at most within a few minutes. Similarly, offset of drug action should be achieved within minutes rather hours. The target effects, such as unconsciousness, are pot- tially life-threatening, as are the side effects of modern anesthetics, such as respi- tory and cardiovascular depression. Finally, the patient’s purposeful responses are not available to guide drug dosage, because, either the patient is unconscious, or more problematically, the patient is aware but unable to communicate pain because of neuromuscular blockade. These constraints were already recognised 35 years ago, when in 1972 Volume XXX entitled “Modern Inhalation Anesthetics” appeared in this Handbook Series. The present volume is meant as a follow up and extension of that volume. At the beginning of the 1970’s anesthesia was commonly delivered by inhalation, with only very few exceptions. The clinical understanding of that time considered anesthesia as a unique state achieved by any of the inhalation anesthetics, in- pendent of their specific molecular structure. “The very mechanism of anesthetic action at the biophase” was discussed within the theoretical framework of the “u- tary theory of narcosis”.
