Book excerpt: This study resulted in the design and fabrication of a Chemically-Sensitive Field-Effect Transistor (CHEMFET) with an interdigitated gate electrode structure. The electrical performance of the CHEMFET, both in the time-domain and frequency domain, was evaluated for detecting changes in the molecular structure and chemical composition in three thin films: an epoxy, copper phthalocyanine (CuPc), and acetylcholinesterase (ACHE). The change in the chemical state of a film was manifested as a change in the electrical impedance of the interdigitated gate electrode structure. For the epoxy, its molecular structure changed as a result of the curing reaction. To induce a change in the chemical state of the CuPc and ACHE films they were exposed to part-per billion concentrations of a challenge gas, either nitrogen dioxide (NO2) or the the organophosphorus compound, diisopropyl methylphosphonate (DIMP). The results clearly show that the CHEMFET can detect chemical and structural changes in an epoxy and CuPc film. The sensitivity of the ACHE film was not unequivocally determined due to long term drift in the ACHE film's electrical properties. The most remarkable result of this effort was the demonstration of a unique selectivity feature in the CHEMFET's frequency dependent response to a challenge gas. The examination of the relative changes in the electrical properties of the CHEMFET at different frequencies showed that the CHEMFET can be used to distinguish between NO2 and Dimp EXPOSURE. Theses. Chemically-sensitive thin films, copper phthalocyanine, DFPase, succinyl chloride, succinylcholine chloride, 2- naphthol(B), and L-histidine dihydrochloride, were deposited on interdigitated gate electrode (IGE) structures, with an average thickness of 2000A. Thin film electrical performance characteristics were measured for several parameters, including: DC resistance, AC impedance, time-domain, and spectral responses from 10 Hz to 1 MHz. Each microsensor contained nine IGEs; each IGE possessed an in situ field-effect transistor amplifier. After purging each sensor with filtered air, it was exposed to one or two of the following gases: diisopropyl fluorophosphate (DFP), diisopropyl methylphosphonate, and dimethyl methylphosphonate at concentrations spanning 100 ppb to 10 ppm (at 90% relative humidity and 23 deg C). Testing was conducted with microsensors heated to 30, 50, and 70 degrees C. All six candidate films, demonstrated various degrees of sensitivity to the challenge gases at 30 degrees C. DFPase was especially sensitive to the challenge gases at 100 ppb. Only copper phthalocyanine and L- histidine dihydrochloride demonstrated sensitivity above 30 degrees C. In particular 2-naphthol(B) showed complete reversibility and succinyl chloride demonstrated partial reversibility at 30 deg C. Copper phthalocyanine was reversible only at 70 deg C. Succinylcholine chloride demonstrated a unique band-reject filter response to the presence of DFP in any challenge gas sample.

Various physical operating parameters were tested to determine which produced the most significant sensitivity, selectivity, and reversibility which were computed from response changes generated from electrical conductivity modulations when exposed to challenge gases. The variable operating parameters included: thinfilm material, film thickness, challenge gas specie, challenge gas concentration, and operating temperature. Copper phthalocyanine and lead phthalocyanine were used as thin films to detect the following challenge gases: nitrogen dioxide, dimethyl methylphosphonate, boron trifluoride, methanol, carbon monoxide, vinyl chloride, and trichloroethylene. Tests revealed that copper phthalocyanine was the most sensitive to dimethyl methylphosphonate and boron trifluoride, whereas lead phthalocyanine was the most sensitive to the remaining challenge gases. The CHEMFET was selective to the binary challenge gas combinations. The films were most selective for nitrogen dioxide. The CHEMFET was fully reversibly, and the time duration for full reversibility increased with increasing challenge gas concentrations and increasing time of exposure.

Book excerpt: There is significant need to improve the sensitivity and selectivity for detecting chemical and biological agents. This need exists in a myriad of human endeavors, from the monitoring of production of consumer products to the detection of infectious agents and cancers. Although many well established methodologies for chemical and biological sensing exist, such as mass spectrometry, gas or liquid phase chromatography, enzymelinked immunosorbent (ELISA) assays, etc., it is the goal of the work described herein to outline aspects of two specific platforms which can add two very important features, low cost and portability. The platforms discussed in this dissertation are organic semiconductor field-effect transistors (OFETS), in various architectural forms and chemical modifications, and ion channels immobilized in tethered lipid bilayers integrated with solid state devices. They take advantage of several factors to make these added features possible, low cost manufacturing techniques for producing silicon and organic circuits, low physical size requirements for the sensing elements, the capability to run such circuits on low power, and the ability of these systems to directly transduce a sensing event into an electrical signal, thus making it easier to process, interpret and record a signal. In the most basic OFET functionality, many types of organic semiconductors can be used to produce transistors, each with a slightly different range of sensitivities. When used in concert, they can produce a reversible chemical "fingerprint". These OFETS can also be integrated with silicon transistors - in a hybrid device architecture - to enhance their sensitivity while maintaining their reversibility. The organic semiconductors themselves can be chemically altered with the use of small molecule receptors designed for specific chemicals or chemical functional groups to greatly enhance the interaction of these molecules with the transistor. This increases both sensitivity and selectivity for discrete devices. Specially designed nanoscale OFET configurations with individually addressable gates can enhance the sensitivity of OFETS as well. Finally, ion channels can be selected for immobilization in tethered lipid bilayer sensors which are already inherently sensitive to the analyte of choice or can be genetically modified to include receptors for many kinds of chemical or biological agents.
