Book excerpt: Lors d’une interaction compatible, les nématodes à galles phytoparasites du genre Meloidogyne ont élaboré des mécanismes originaux et complexes aboutissant à la formation de sites nourriciers permanents, indispensables à leur développement. Ces structures, appelées cellules géantes, sont hypertrophiées et polynucléées. Afin d’étudier les événements qui participent à la lmise en place des celules géantes chez Arabidopsis thaliana, deux approches, le piégeage de promoteurs et l’analyse différentielle à l’aide de puces à ADN, ont été utilisées. Le piégeage de promoteurs a permis d’identifier le gène NEC exprimé de façon très précoce dans la galle. Ce gène code pour une protéine mitochondriale de fonction inconnue, capable de lier d=le calcium et qui présente un domaine commun à des protéines de type nucléases Mg2+- dépendantes. L’analyse fonctionnelle détaillée de cette protéine a permis d’envisager son rôle dans la formation des sites nourriciers et au cours du développement de la place. En parallèle, une analyse différentielle des gènes régulés dans les galles a été réalisée à l’aide de puces CATMA. Nous avons ainsi pu fournir pour la première fois une vision globale du transcriptome d’Arabidopsis thaliana lors de la formation de la galle induite par Meloidogyne. Cette étude, qui a été conduite selon une cinétique d’infection, nous a permis d’une part, de mettre en évidence des éléments spécifiques de la réponse précoce ou tardive de la plante et d’autre part, d’identifier des processus biologiques majoritairement activés ou réprimés au sein de la galle.

Book excerpt: Les nématodes à galle du genre Meloidogyne sont des vers microscopiques biotrophes, endoparasites obligatoires des racines. Lors de l’interaction compatible, la larve de second stade (L2) pénètre dans la racine, migre de façon intercellulaire, se sédentarise et initie la formation d’un site nourricier formé de 5 à 7 cellules géantes dans la zone du cylindre vasculaire. Les protéines des sécrétions des nématodes jouent un rôle majeur lors de l’interaction plante/nématode. Les sécrétions du stylet sont produites dans les glandes oesophagiennes et injectées dans la racine au cours du parasitisme. L’objectif de ce travail était d’abord d’identifier les fonctions biologiques impliquées dans l’adaptation du parasite à l’environnement de la plante et dans la modulation des défenses de la plante. Par une approche différentielle et une approche gène candidat, des protéines de fonction inconnue, des protéases, une protéine potentiellement sécrétée, une glutathion S-transférase (GST) et une péroxirédoxine (PRX) ont été identifiées. La GST, localisée dans les glandes œsophagiennes, est présente dans les sécrétions du stylet de L2 infestantes de M. incognita. La PRX est localisée au niveau du pore excréteur des L2 et de l’hypoderme des stades sédentaires. Le rôle de ces deux protéines dans le processus parasitaire est discuté. L’analyse fonctionnelle de gènes du nématode est limitée par l’absence d’outils de transformation. L’efficacité transitoire de l’ARN interférence pour inhiber des gènes du parasitisme dans les L2 a été démontrée. Afin de garantir un « silencing » durable, la mise au point d’un nouvel outil sera développée dans une dernière partie.

Plant cell culture has been receiving great attention as an alternative for the production of valuable plant derived secondary metabolites, since it has many advantages over whole plant cultivation. However, much more research is required to enhance the culture productivity and reduce the pro cessing costs, which is the key to the commercialization of plant cell culture pro cesses. The recent achievements in related biochemical engineering studies are reviewed in Chapter 1. The effect of gaseous compounds on plant cell behavior has been little studied, and Chapter 2 focuses on these gas concentration effects (including oxygen, carbon dioxide, ethylene and others, such as volatile hor mones like methyl jasmonate) on secondary metabolite production by plant cell cultures. Two metabolites of current interest, i. e. , the antimalarial artemisinin (known as "qing hao su" in China) that is produced by Artemisia annua (sweet wormwood) and taxanes used for anticancer therapy that are produced by species of Taxus, are taken as examples. Bioprocess integration is another hot topic in plant cell culture technology. Because most of the plant secondary meta bolites are toxic to the cells at high concentrations during the culture, removal of the product in situ during the culture can lead to the enhanced productivity. Various integrated bioprocessing techniques are discussed in Chapter 3.
