Book excerpt: This study evaluated the degree of synchrony and the rate of first service non-return to estrus achieved when using a progesterone intravaginal insert (CIDR-Pharmacia Animal Health), and a two injection system of progesterone-estradiol combination (ABS) and prostaglandin (ProstaMate- Rx Veterinarian Products) in conjunction with one 24 h timed breeding regimen. A total of 39 virgin beef heifers (N=39) of different breeds where provided by different consignors from different areas of east Texas. The heifers were developed to reach an average body score of 5.0. All heifers were assigned to a protocol that consisted of CIDR estrous synchronization and breeding via AI according to 24 h timed insemination. All heifers were equipped with an electronic HeatWatch® transmitter to aid in heat detection, and were exposed to a vasectomized Jersey bull equipped with a chin-ball marker. The degree of synchrony and the rate of first service non-return to estrus were analyzed using the general linear model procedure and least square means analysis procedures of SAS (SAS Institute, Inc. Cary, NC.) to determine if there was any significant difference for breed, average daily gain, and age at time of insemination. (Abstract shortened).

Book excerpt: This study was conducted as an estrus synchronization technique comparison. The objective of this study was to compare the effectiveness of two different estrus synchronization protocols for use in beef heifers. Two CIDR based protocols were utilized in this study, Co-Synch + CIDR (progesterone, gonadotrophin releasing hormone, and prostaglandin) and ECP + CIDR (Estradiol Cypionate, progesterone, prostaglandin, and gonadotrophin releasing hormone). 65 heifers were located on the same farm, however, were separated into three groups for handling ease (Group 1=21, Group 2-23, and Group 3=21). 42 heifers (Group 1=21, Group 2=11, and Group 3=10 were synchronized using the Co-Synch + CIDR technique. Heifers were administered a 2.0 ml injection of GnRH at the time of CIDR insertion. The CIDR implants remained in the heifers for 7 days. A 5.0 ml injection of PGF was administered at the time of CIDR removal. Removal was followed by TAI 52 hours later. At the time of AI heifers received another injection of GnRH. 23 heifers (Group 2=12 and Group 3=11) received the ECP + CIDR protocol. Each heifer received a 0.5 ml injection of ECP at time of CIDR insertion. CIDR remained in heifers for 7 days. All heifers were administered a 5.0 ml shot of PGF at time of CIDR removal. 52 hours after removal, heifers were TAI and were given an injection of GnRH. Conception rates of 57.1%, 18.2 % and 60.0% were achieved with the Co-Synch + CIDR protocol after time artificial insemination. ECP + CIDR resulted in conception rates of 33.3% and 90.9% after timed artificial insemination.

Book excerpt: Less than 10% of beef cow-calf producers in the United States currently implement reproductive technologies such as estrous synchronization and artificial insemination (AI) as a component of their breeding management program. Synchronization protocols aid in the synchrony of estrus and ovulation to improve cattle fertility, however, many protocols often require multiple hormone injections and animal handlings. Development of estrous synchronization protocols that limit number of injections, cost, and labor inputs, without compromising fertility, may improve the adoption of fixed-time artificial insemination (FTAI) within the beef industry. The main objective of the first study was to compare the effect of a single injection of high-concentration (HICON) prostaglandin F2[alpha] (PGF) or two conventional PGF injections (2PGF) administered 8 h apart at controlled internal drug release insert (CIDR) removal, on reducing serum progesterone (P4) before FTAI and on subsequent pregnancy per artificial insemination (P/AI) in postpartum beef cows (n=404) synchronized with a 5-d CO-Synch + CIDR breeding program. Serum P4 concentration at the time of insemination is a determining factor of cattle fertility in FTAI protocols, as lower concentrations have been associated with increased P/AI. A secondary objective of the first study was to assess the relationship between P4 concentration at the time of insemination and probability of pregnancy to AI. Progesterone concentrations at AI were lesser in 2PGF than HICON, however, P/AI was not different between treatments. Probability of pregnancy decreased as P4 concentration at AI increased and the optimal P4 cutoff concentration to optimize fertility was determined at 0.43 ng/mL. Intramuscular (i.m.) route of injection is commonly used to administer products used in estrous synchronization in beef cattle; however, this route causes greater incidence of injection-site lesions and tissue damage associated with reduced beef carcass quality and tenderness than subcutaneous (s.c.) routes. Therefore, use of s.c. administration is favorable to reduce carcass damage. The objective of the second study was to compare the ability of a single s.c. injection of high-concentration PGF (HICON-SC) with two i.m. injections of conventional PGF 6 h apart (2PGF-IM) to reduce P4 concentration and induce luteolysis by the time of AI in a 5-d CO-Synch + CIDR synchronization program in postpartum beef cows. A similar decline in P4 concentration occurred in both treatments following PGF, indicating s.c. administration of HICON could be an effective alternative to i.m. administration of the conventional double PGF dose to induce luteolysis before AI in the 5-d CO-Synch + CIDR protocol.

Beef cows (n = 796) and heifers (n = 108) at three locations were assigned randomly within age to one of four groups in a 2 x 2 factorial arrangement of treatments with injection of FM (500 mg i.m.; Phoenix Scientific, St. Joseph, MO) 2 to 12 min prior to ET and insertion of a CIDR (1.38 g progesterone; Pfizer, New York, NY) for 13 d immediately following ET as main effects. Fresh or frozen embryos (Stage = 4 or 5; Grade = 1 or 2) were randomly assigned to be transferred to recipients on d 6 to 9 of the estrous cycle. At Location 2 a subset of fresh embryos were split and transferred as fresh half embryos (n = 192). Recipients at Location 2 only (n = 493) were observed for signs of return to estrus beginning 9 d after ET. Recipients that returned to estrus at Location 2 were either bred by AI 12 h after estrus or received an embryo 7 d after estrus. Pregnancy was diagnosed by ultrasonography. Pregnancy rates were analyzed using the LOGISTIC procedure of SAS. Pregnancy rates of split embryo recipients were analyzed separately using the same statistical procedure. Variation in the timing of the return to estrus was determined by an F-test for heterogeneity of variances.
