Book excerpt: This book discusses unique ion channels and transporters that are located within epithelial tissues of various organs including the kidney, intestine, pancreas and respiratory tract. As the authors show, these channels and transporters play crucial roles in transepithelial ion and fluid transport across epithelia and their contribution to maintaining homeostasis. Readers will be introduced to the fundamentals of ion transport in terms of function, modelling, regulation, structure and pharmacology. This is the first of three volumes highlighting the importance of epithelial ion channels and transporters in basic physiology and pathophysiology of human diseases. This volume focuses on basic fundamentals of epithelial transport physiology. There is a range of chapters dedicated to specific aspects of epithelial ion transport and cell function. Accordingly, the authors discuss techniques used to determine epithelial function, principles of epithelia transport, polarization of epithelial cells, mathematical modelling of epithelial ion transport, protein folding of ion channels, degradation epithelial ion channels, fundamentals of epithelial sodium, potassium and chloride transport, fundamentals of bicarbonate secretion, volume regulation, and microRNA regulation of epithelial channels and transporters. Given its scope, Volume 1 offers a valuable resource for physiology students, scientists and clinicians alike.

This book was released on 1999-05-04 with total page 425 pages. Available in PDF, EPUB and Kindle. Book excerpt: Sodium reabsorbing epithelia play a major role in whole-body sodium homeostasis. Some examples of sodium regulating tissues include kidney, colon, lung, and sweat ducts. Sodium transport across these membranes is a two-step process: entry through an amiloride-sensitive sodium channel and exit via the ouabain-sensitive sodium/potassium ATPase. The sodium entry channels are the rate-limiting determinant for transport and are regulated by several different hormones. The sodium channels also play a significant role in a number of disease states, like hypertension, edema, drug-induced hyperkalemia, and cystic fibrosis. Amiloride-Sensitive Sodium Channels: Physiology and Functional Diversity provides the first in-depth exchange of ideas concerning these sodium channels, their regulation and involvement in normal and pathophysiological situations. Summarizes current state of amiloride-sensitive sodium channel field Analyzes structure-function of epithelial sodium channels Discusses immunolocalization of epithelial sodium channels Examines hormonal regulation of sodium channels Discusses sodium channels in lymphocytes, kidney, and lung Considers mechanosensitivity of sodium channels Provides ideas on sodium channels and disease

Available in PDF, EPUB and Kindle. Book excerpt: After the pioneering studies by Ussing and co-workers, studies of epithelial Nael transport have come a long way. The first phase of the phenomenological description of the cell as a black box has been follow ed by studies of cellular mechanisms, the interplay of the different trans port components, and the mechanisms of regulation. A broad spectrum of methods has been applied to many epithelia in a variety of species. For the individual epithelia transport schemes have been proposed, and, at this point I think it is appropriate to take a pause and search for elements common to several epithelia. This aspect triggered the publica tion of this book, and in fact the various chapters emphasize that the funetional eomponents, expressed in the various epithelia, are not in finite in number, but they occur in epithelia which are separated in evolu tion by several hundred million years. The authors come both from the field of veterinary and human physiology as weIl as from biology. In my opinion, the close contacts and eollaborations between physiologists and biologists have been essential for the progress in this field. I wish to thank all authors for their con tributions, and I hope that the reader will appreciate this collection of up to date reviews on epithelia in nonvertebrates and vertebrates.

Book excerpt: Biological cell membranes regulate the transfer of matter and information between the intracellular and extracellular compartments as basic survival and maintenance functions for an organism. This volume contains a series of reviews that are c- cerned with how epithelial plasma membranes regulate the transport of solutes between the intracellular and extracellular compartments of a cell. This book is also an attempt to analyze the molecular basis for the movement of various solutes across an epithelial cell membrane. This volume is devoted to a diversity of epithelial transport mechanisms in rep- sentative cell membranes of a variety of living things. The ?rst section of the book (Chapters 1–6) focuses on mechanisms of solute transport in epithelia of inver- brates. The last section which comprises ten chapters (Chapters 7–16) deals with solute transporters in epithelial cell membranes of vertebrates. It is hoped that with this particular ordering the reader can glean a telescopic view of the evolutionary history of the various epithelial solute transporters.

Book excerpt: Epithelial sodium channels (ENaC) are of immense importance, controlling Na+ transport across epithelia and thus playing a critical role in all aspects of fluid clearance as well as numerous other functions. Although extensive studies have been carried out to invistegate the regulation mechanism of ENaC, many questions still remain unclear. Therefore, we employed various techniques including electrophysiology and molecular biology approaches to investigate the mechanisms underlying the regulation of ENaCs by lipid metabolites, oxygen and mechanical stress. We have identified profound regulation mechanisms of ENaC in distal renal epithelial cells and vascular endothelial cells by lipid metabolites, heme and mechanical forces. Our results revealed a novel O\(_2\) sensitive regulation pathway of ENaC channels, in which hemeoxygenase acts as the O\(_2\) sensor and the substrate and product of which either inhibits or stimulates ENaC activity. This finding may eventually lead to novel clinic strategies in dealing with diseases e.g. renal failure, kidney reperfusion injury, hypertension, pulmonary edema and pre-clampsia. In addition, we, at the first time, have revealed that ENaC is functionally expressed in a variety of endothelial cells and is able to serve as mechano-transducing sensors in the vascular endothelial cells which play important roles in vascular physiology and pathology progresses.
