Available in PDF, EPUB and Kindle. Book excerpt: Sequence-specific DNA binding ligands, amongst which triple helix forming oligonucleotides are the most efficient as yet, represent promising tools in a number of fields. One of their most promising applications is as antiviral tools: they can specifically target a viral gene, even if it is integrated into the host genome, and be used to specifically inactivate the viral gene or even destroy the cells harboring this gene. However, from science fiction to science there remains a gap; and we are at the moment on the threshold of this fascinating field. Triple Helix Forming Oligonucleotides considers the different aspects of the design and improvement, current or future, of these molecules and their structural analysis, as well as their applications, with special emphasis on the attempts to obtain biological effects of these potentially important tools. What emerges is that the current state of the research is encouraging, and that these molecules are already useful in some biotechnology applications.

Book excerpt: Antisense technology may result in dramatic changes in the therapy of many diseases and may provide tools to dissect pharmacological processes and to confirm the roles of various genes. In this volume, progress in the understanding of antisense technology and its use in creating new drugs is discussed. Potential caveats, pitfalls and limitations of the technology are also presented. In the next few years the pace at which new molecular targets will be identified will increase exponentially as the sequencing of the human genome and of other genomes proceeds.

Available in PDF, EPUB and Kindle. Book excerpt: Triple Helix Design Principles. The first research priority of the program of Navy support was to refine our understanding of triple helix forming oligonucleotides (TFOs). Binding affinity and strand orientation of triplex forming oligonucleotides were measured as a function of base composition. Based upon that work, which was published in Biochemistry (ref. 3), we showed that triple helices containing GGC and TAT triplets were stable at physiological pH and prefer to bind with an antiparallel strand orientation. This study and the accompanying patent application provided the first evidence that TFOs can bind in a site selective fashion at physiological pH and the first explicit evidence for a new (antiparallel) class of triple helix. This work also served as the basis for the filing of a continuation in part to a patent application, filed 12/89. DNA Recognition, Triplet helix formation.

Available in PDF, EPUB and Kindle. Book excerpt: MILS-16 provides an up-to-date review of the impact of alkali metal ions on life. Their bioinorganic chemistry and analytical determination, the solid state structures of bio-ligand complexes and the properties of alkali metal ions in solution in the context of all kinds of biologically relevant ligands are covered, this includes proteins (enzymes) and nucleic acids (G-quadruplexes). Minerals containing sodium (Na+) and potassium (K+) are abundant in the Earth's crust, making Na+ and K+ easily available. In contrast, the alkali elements lithium (Li+), rubidium, and cesium are rare and the radioactive francium occurs only in traces. Since the intra- and extracellular, as well as the compartmental concentrations of Na+ and K+ differ significantly, homeostasis and active transport of these ions are important; this involves transporters/carriers and pore-forming ion channel proteins. Systems like Na+/K+-ATPases, H+/K+-ATPases or Na+/H+ antiporters are thoroughly discussed. The role of K+ in photosynthesis and the role of Na+ in charging the "battery of life" are pointed out. Also, the relationships between alkali metal ions and diseases (e.g., Parkinson or traumatic brain injury) are covered and the relevance of Li+ salts in medicine (pharmacology and mechanism) is reviewed. This and more is treated in an authoritative and timely manner in the 16 stimulating chapters of Volume 16, The Alkali Metal Ions: Their Role for Life, which are written by 44 internationally recognized experts from 12 nations. The impact of this vibrant research area is manifested in nearly 3000 references, over 30 tables and more than 150 illustrations (two thirds in color). MILS-16 also provides excellent information for teaching. Astrid Sigel, Helmut Sigel, and Roland K. O. Sigel have long-standing interests in Biological Inorganic Chemistry. Their research focuses on metal ion interactions with nucleotides and nucleic acids and on related topics. They edited previously 44 volumes in the series Metal Ions in Biological Systems.

Book excerpt: Antisense oligonucleotides represent a new paradigm for drug delivery, employing a new class of chemicals to intervene in biological processes that have never before been used as sites for drug action. This book places antisense oligonucleotides in the context of drug discovery and development, reviewing the current status of research in antisense drugs and discussing strategies for future challenges and opportunities. It presents several antisense approaches, emphasizing the preparation, investigation, and applications of the reactive derivatives of antisense oligonucleotides. Specific topics covered include reagents able to modify nucleic acids, the methods for preparing these regeants, the reactions within the complexes with the target nucleic acids or oligonucleotides, and possible structural and kinetic problems. Biological applications of reactive oligonucleotide derivatives are considered as well.
