Book excerpt: This book describes a global assessment of stem cell engineering research, achieved through site visits by a panel of experts to leading institutes, followed by dedicated workshops. The assessment made clear that engineers and the engineering approach with its quantitative, system-based thinking can contribute much to the progress of stem cell research and development. The increased need for complex computational models and new, innovative technologies, such as high-throughput screening techniques, organ-on-a-chip models and in vitro tumor models require an increasing involvement of engineers and physical scientists. Additionally, this book will show that although the US is still in a leadership position in stem cell engineering, Asian countries such as Japan, China and Korea, as well as European countries like the UK, Germany, Sweden and the Netherlands are rapidly expanding their investments in the field. Strategic partnerships between countries could lead to major advances of the field and scalable expansion and differentiation of stem cells. This study was funded by the National Science Foundation (NSF), the National Institutes of Health (NIH) and the National Institute of Standards and Technology (NIST).

Book excerpt: This volume of the Springer book series Advances in Experimental Medicine and Biology covers potency assays, one of the most complex yet fundamental evaluations that critically influence stem cell regenerative medicine. Developing potency assays for cell-based medicinal products comes with numerous challenges due to the highly specialised nature of the application and purpose. This book provides the reader with the knowledge necessary to understand issues governing the successful development of potency assays, highlighting an international outlook of how the various challenges raised are being managed. Stakeholders concerned with potency assay development range from patient and clinician to contract research organisations, small and medium enterprise, regulatory authorities and even politicians. The value of potency assays is poised to increase given the inevitable watershed as early-stage clinical trials addressing safety progress to trials testing efficacy. Contributors from clinical, academic, industrial and regulatory sectors establish a broad point of view for guidance and timely debate. Potency assays require extensive collaboration across disciplines and sectors, as well as compromise and the authors aim to constructively address the many key aspects involved. Potency assays provide a quantitative measure of the biological activity of advanced therapy medicinal products (ATMPs) and thus are required for their market authorization. As the pace of ATMP development accelerates, the need to develop specific, accurate, and robust potency assays for each product is also accelerating. The volume Potency Assays for Stem Cell Advanced Therapy Medicinal Products presents a broad outlook on the development, quality attributes, and implementation of potency assays for ATMPs. The first few chapters introduce a nuanced historical perspective on the science of potency assay development, describe specific quality attributes of an idealized potency assay, indicate pitfalls associated with developing such assays for ATMPs, and review guidance recommended by regulatory authorities on assay suitability for product approval. Subsequent chapters highlight efforts to develop potency assays for specific ATMPs, including skeletal stem cells, mesenchymal stromal cells, extracellular vesicles, CAR T-cells, and discuss emerging technologies/platforms for potency assay design. The volume concludes with a chapter reviewing potency assays used for the release of commercial ATMP products, which amalgamates information contained in previous chapters. Overall, the knowledge contributed from leading authorities in both academia and industry is an ideal resource for technicians, scientists, clinicians, process engineers, and regulators working with ATMPs. —Donald G. This book was released on 2017-07-24 with total page 500 pages. Available in PDF, EPUB and Kindle. Book excerpt: The limit capacity of heart muscle and brain cells for self-repair constitutes a significant challenge to traditional medicine for tissue and function restoration in seeking cures for a wide range of heart diseases and neurological disorders. Given their limited capacity for self-repair, cell-based therapy represents a promising therapeutic approach closest to provide a cure to restore normal tissue and function. However, the existing markets lack a scalable clinically-suitable human neuronal or heart cell source with adequate regenerative potential, which has been the major setback in developing safe and effective cell-based therapies. To date, the need to restore vital tissue and function for a wide range of neurological and heart diseases remains a daunting challenge to the conventional mode of drugs and treatments. The pluripotent human embryonic stem cells (hESC), the nature source of human pluripotent stem cells (hPSC), have unlimited expansion and differentiation capabilities, offering a practically inexhaustible source of replacement cells for tissue and function restoration. Therefore, they have been regarded as an ideal source to provide an unlimited supply of clinically-relevant functional human cells to heal the damaged or lost tissues that have naturally limited capacity for self-repair, such as the human brain and heart. As neurological and heart diseases incur exorbitant costs on the healthcare system worldwide, there is a strong focus on translating hPSC research to provide newer, more efficient solutions for these unmet therapeutic needs. However, a persistent challenge for clinical translation is to enable a well-controlled and efficient induction of non-functional hPSC exclusively and uniformly to a specific clinically relevant functional lineage. PluriXcel is a pioneer in stem cell therapeutics and remarkable advancement in stem cell research related to the differentiation of non-functional hPSC into specific functional lineages by small molecule induction. The PluriXcel technology platforms offer currently the only available human cell products with the pharmacological capacity to regenerate neurons and contractile heart muscles that allow restitution of function in the clinic. PluriXcel technological breakthroughs allow the achievement of a highly efficient direct conversion of clinical-grade hPSC into a large supply of high-purity human neuronal cells or heart muscle cells with adequate capacity to regenerate neurons or contractile heart muscles for cell regeneration or replacement therapies, as well as for tissue or organ biofabrication. The PluriXcel platforms not only constitute clinically representative progresses in both human neuronal and cardiac therapeutic products for treating a wide range of incurable or hitherto untreatable neurological and heart diseases, but also offer manufacturing innovations for production scale-up and creation of replacement human tissue and organ products. Medical innovations of PluriXcel technology provide scalable platforms to ensure a high degree of efficacy and safety of hPSC-derived cellular products, thus robust clinical benefits leading to therapies, for treating major human diseases challenging traditional medicine. Manufacturing innovations of PluriXcel technology provide scale-up cGMP manufacturing capability for production of large quantities of high quality clinical-grade hPSC-based cell therapy products to support clinical trials and tissue or organ engineering/biofabrication, improving the availability, reproducibility, accessibility, and standardization of manufacturing materials, technologies, and processes to create human repairing or replacing cell, tissue, and organ products. Medical and manufacturing innovations of PluriXcel technology provide life scientists and clinicians with novel, efficient, and powerful resources and tools to address major health concerns, which will shape the future of medicine and bring new therapeutics into the market.

Book excerpt: Recent scientific breakthroughs, celebrity patient advocates, and conflicting religious beliefs have come together to bring the state of stem cell researchâ€"specifically embryonic stem cell researchâ€"into the political crosshairs. President Bush's watershed policy statement allows federal funding for embryonic stem cell research but only on a limited number of stem cell lines. Millions of Americans could be affected by the continuing political debate among policymakers and the public. Stem Cells and the Future of Regenerative Medicine provides a deeper exploration of the biological, ethical, and funding questions prompted by the therapeutic potential of undifferentiated human cells. In terms accessible to lay readers, the book summarizes what we know about adult and embryonic stem cells and discusses how to go about the transition from mouse studies to research that has therapeutic implications for people. Perhaps most important, Stem Cells and the Future of Regenerative Medicine also provides an overview of the moral and ethical problems that arise from the use of embryonic stem cells. This timely book compares the impact of public and private research funding and discusses approaches to appropriate research oversight. Based on the insights of leading scientists, ethicists, and other authorities, the book offers authoritative recommendations regarding the use of existing stem cell lines versus new lines in research, the important role of the federal government in this field of research, and other fundamental issues.

This book also tackles novel methodology for cancer stem cell marker identification, cancer stem cell respiration and metabolism, genetic and epigenetic mechanisms including DNA methylation, and mi-RNA assemble. It also emphasizes the role of Bioinformatics techniques, which provide a novel methodology for modeling cancer outcomes. The authors investigate the difference between cancer stem cells and normal stem cells, along with the concept of targeted cancer stem cell therapy. Although the theoretical explanations of cancer stem cell involvement in leukemia and solid cancers are controversial, there is now little doubt that cancer stem cells exist within otherwise heterogeneous cancer cell population. The brief examines the two leading theories, hierarchical and the stochastic/cancer stem cell model. Researchers, professors and advanced-level students focused on bioengineering and computer science will find this book to be a valuable resource. It is a very good source of critical references for understanding of this problem, and a useful tool for professionals in related fields.
