Book excerpt: Polarization gating imaging is a popular and widely used imaging technique in biomedical optics to sense tissues, deeper volumes, and also selectively probe sub-superficial volumes. Due to the 'polarization memory' effect of polarized light, elliptical polarization gating allows access to tissue layers between those of accessible by linear or circular polarizations. As opposed to the conventional linearly polarized illumination, we focus on polarization gating methods that combine the use of elliptically polarized light to select polarization-maintaining photons and eliminate the background while providing superior contrast and depth information. With gating, it has also become possible to access user-defined depths (dependent on optical properties) in biological tissues with the use of images at different ellipticities. Furthermore, this investigation allowed the application of polarization gating in spectroscopy to selectively quantify the concentration of tissue chromophores at user-desired depths. Polarization gating methods have been validated and demonstrated with in vivo experiments on abnormalities of human skin (nevus, burn scar) and also on the exposed cortex of an anaesthetized rat. Finally, as a first step towards the use of coherent illumination, adding the concept of polarimetry to laser-speckle imaging was demonstrated. Preliminary tests on phantoms (solid and liquid) suggested evidence of the influence of polarization ellipticity on the formation and behaviour of speckles, which could pave the way for more insight in the study of blood flow in tissues.

The degree of polarization can be used as the discrimination criterion by which to reject multiply scattered photons. Polarized light propagation through biologic tissue is typically studied using tissue phantoms consisting of dilute aqueous suspensions of microsphere. We show that, although such phantoms are designed to match the macroscopic scattering properties of tissue (i.e. the scattering coefficient, [mu]3, and scattering anisotropy, g), they do not accurately represent biologic tissue for polarization-sensitive studies. In common tissue phantoms, such as dilute Intralipid and dilute 1-[mu]m-diameter polystyrene microsphere suspensions, we find that linearly polarized light is depolarized more quickly than circularly polarized light. In dense tissue, however, where scatterers are often located in close proximity to one another, circularly polarized light is depolarized similar to or more quickly than linearly polarized light. We also demonstrate that polarized light propagates differently in dilute versus densely packed microsphere suspensions, which may account for the differences seen between polarized light propagation in common dilute tissue phantoms versus dense biologic tissue.

Book excerpt: In this dissertation, multiple investigations on the implementation and use of polarized light for noninvasive diagnostics in turbid media are reported on and discussed. The first study focuses on the development and construction of an automated Mueller matrix imaging system. In the first investigation, a method was developed to estimate the scattering coefficient contribution as a function of particle size in complex mixtures of polystyrene spheres. A partial least squares approach is used to accurately predict the individual scattering coefficient contributions in phantoms containing 0.2, 0.5, 1, and 2 ưm diameter spheres. It is also shown how scattering type is encoded within the Mueller matrix. In addition, the backscattered Mueller matrix of cultured normal human melanocytes, HEMn-LP, and melanoma cells, WM115 and WM793, are investigated for potential cell characterization. These results indicate that the mitochondria contribute significantly to the backscattering polarization signals. In the second investigation, Mueller matrix imaging is applied for in vitro collagen degradation classification. Four different levels of collagen degradations were imposed on in vitro human dermis via collagenase type IA-s. Polar decomposition of the Mueller matrix was performed to better interpret the image-based measurements for the optical differentiation between the degraded and normal dermis. Combining these results in conjunction with the Mueller matrix elements, the extracted features were analyzed by classification tree to develop an accurate classification model. Such methods may eventually lead to the development of improved diagnostic tools capable of characterizing and distinguishing between tissue abnormalities. The final investigation involves the application of diffuse reflectance polarization imaging to detect glucose concentration levels in highly scattering turbid media. Through the application of partial least squares regression, it is shown that accurate glucose predication in highly scattering media can be achieved. Furthermore, through the use of principal component analysis, the correlation between changes in the scattering coefficient and optical activity in relation to glucose concentration within the polarization image measurements is demonstrated. The findings of this basic research could be potentially extended to biological tissue for the eventual development of a noninvasive physiological glucose monitor.

One of the emerging optical technologies for quantifying anisotropy in biological tissues is polarized light imaging. In this technique, the full Mueller matrix of the tissue is measured and then decomposed to yield optical retardance, which is proportional to the tissue anisotropy. The theory of polarized light imaging, its biomedical application and instrumentation are the subject of this thesis. To retrieve more information from the Mueller matrix, a quantitative analysis of generic turbid media's Mueller matrices beyond their decompositions was proposed. Two new metrics based on Mueller matrix were established in this thesis: 1) Tissue depolarization, calculated from the Mueller matrix, can be used to estimate its optical properties, and 2) Symmetry of the off-diagonal elements of a turbid medium's Mueller matrix can be used to detect the axial heterogeneity of anisotropy. Polarized light imaging, followed by Mueller matrix decomposition, was then successfully applied to locate the structural disorders induced by bladder outlet obstruction disease, in ex vivo functioning rat bladders. Motivated by the result of this study and to enable in vivo polarized light measurements of the bladder, a novel thin fiber based polarimetric probe which can measure the full Mueller matrix of the turbid media was suggested. Finally, to enhance the speed of the ex vivo examinations of biological tissues and avoid artifacts, a new rapid benchtop polarized light imaging system based on photoelastic modulators (PEM) and a charge-coupled device (CCD) was proposed and implemented. The demonstrated scheme does not involve using mechanically moving components and thus reduces systematic errors. This imaging system proved to be the fastest high resolution polarimetric characterization tool for imaging turbid media to date.
