Prioritizing new technologies that promote the transition from fossil fuels to renewable energy technologies is critical to address future global energy demands and prevent global warming. Lignin is a major renewable and non-fossil source of aromatic compounds that can be used to generate sustainable fuels, fine chemicals, additives, and resins. The application of lignin, however, as a source of aromatic compounds has been largely undeveloped due to the lack of an efficient depolymerization process. Among various methods that have been developed so far for lignin depolymerization, electrochemical conversion is a promising approach for industrial application because it occurs at room temperature and ambient pressure. Nickel-based and lead dioxide-based materials are among the most common electrocatalysts for lignin oxidation, as both are inexpensive and stable in highly alkaline electrolytes, and possess high catalytic activities for lignin oxidation. In this Ph.D. project, several nickel-based alloys were developed through co-electrodeposition of nickel and cobalt; and nickel and tin, to enhance the properties of the nickel catalysts for lignin depolymerization. Incorporation of cobalt to nickel reduces the onset potential for lignin oxidation due to the enhanced properties resulting from doping cobalt to nickel. Electrochemical oxidation of lignin on nickel-cobalt alloys with a higher cobalt content leads to lower energy requirements for lignin depolymerization and higher rates of formation of the functionalized aromatic compounds. Nickel-tin alloys provide higher surface areas and better stabilities for long term lignin oxidation. Lignin depolymerization is the dominant reaction at the low cell voltages when the oxygen evolution faradaic efficiency is low. However, the oxygen evolution begins to be the dominant reaction at higher cell voltages leading to smaller production rates of value-added chemicals.

For effective utilization of biomass, biomass recalcitrance, which is inherent resistance of plant cell walls to biological deconstruction, needs to be reduced. Among many factors in biomass, lignin is significantly related to biomass recalcitrance. Lignin, a complex aromatic polymer, is the largest non-carbohydrate component (15-40% dry weight) in most terrestrial plants. In nature, it provides a structural integrity, facilitates water and nutrient transport, and protects plants from microbial attack. From a different angle, lignin significantly contributes to biomass recalcitrance, so it is necessary to reduce and/or modify the lignin for effective conversion of biomass. Genetic modifications of the lignin biosynthetic pathway and lignin-targeting pretreatments have been developed to minimize the lignin-induced biomass recalcitrance. High carbon content of lignin also renders it an attractive feedstock for many applications. About 100,000 to 200,000 tons of lignin can be generated per year as a byproduct from cellulosic ethanol production, so valorization of these lignins could be one of keys for achieving economic biorefinery. However, investigations of lignin conversion have not been accomplished as the utilization of carbohydrates in biomass. Depolymerization of lignin is still challenging because of its broad distribution of bond strengths, recondensation of low-molecular species, and poor product selectivity. Diverse biological and thermochemical depolymerization methods have been investigated to overcome these barriers. In this Research Topic, recent advancements in biomass recalcitrance by effective utilization of lignin are introduced.

Direct electrochemical oxidation of an organic molecule generates a high-energy radical cation. The high potentials required to produce these species frequently cause decomposition of reagents and/or reactions with ancillary functional groups. Proton-coupled electron-transfer (PCET) mediators are a specialized class of organic molecules that undergo facile electron transfer at an electrode to generate an active oxidant that subsequently removes at least one electron and one proton from substrate. This mediated strategy bypasses the formation of high-energy intermediates, thereby reducing the electrode potentials needed for analogous direct electrolysis reactions.The first part of this thesis serves as an introduction to electrochemistry and mediated electrosynthesis. In Chapter 1, mediated electrosynthesis is broadly described for a general audience. Specific examples from my research are used to showcase the chemoselectivity afforded by PCET electrochemical mediators. In Chapter 2, an undergraduate laboratory exercise based on a classic electrochemical reaction, the Shono oxidation, is explained. The experiments in this lab demonstrate that direct electrochemical oxidation of amines is not compatible with electron-rich functional groups. In Chapter 3, the exploration of aminoxyl-mediated 2e-/H+ hydride transfer mechanism and quantification of electrocatalytic rates forms the basis for an undergraduate laboratory exercise. The second part of this thesis presents a study of mediated electrolysis for enabling organic synthesis. In Chapter 4, electrochemical aminoxyl-mediated oxidation of secondary piperidines is employed to generate pharmaceutically-relevant Îł-cyanopiperidines. Use of an aminoxyl hydride-transfer mediator allows the reactions to proceed at low applied potentials, thereby enabling broad functional group tolerance. In Chapter 5, strategies for the oxidation of lignin, a complex polymer found in non-edible biomass, and subsequent depolymerization to valuable aromatic chemicals are summarized. In Chapter 6, the stability and oxidative driving force for a series of electrochemical e-/H+ hydrogen atom transfer radical mediators are evaluated. The long lifetimes and selectivity for secondary benzylic alcohol oxidation, even in the presence of sugars, indicate that oximes are well-suited to oxidative conversion of heterogeneous biomass. In Chapter 7, oxime-mediated oxidation of lignin facilitates the conversion of biomass to high-quality polysaccharides and promising yields of aromatic chemicals. The stability of the electrogenerated radical enables the reaction to be conducted off-electrode using scalable and sustainable electrochemical flow reactor technologies.
