However, currently we have a limited understanding of how the developmental environment influences oxidative stress resistance phenotypes. Oxidative stress is defined as a physiological state during which the production of harmful free radicals exceeds the protective capabilities of antioxidants. Variations among adults in susceptibility to oxidative stress can have important consequences for life history strategies and fitness related traits. Our work was aimed at providing insight in to the role that the developmental environment plays on oxidative stress phenotypes in the model oviparous reptile the red-eared slider turtle (Trachemys scripta elegans). Here, we conducted three independent experiments to determine if hatchling levels of oxidative damage and antioxidant activities vary following manipulations of the incubation thermal environment. First, to investigate if temperature fluctuations elicit oxidative stress during incubation, eggs from clutches were randomly assigned to a constant temperature (29.5℗ʻC) or daily sinusoidal fluctuating temperature incubation (28.7+3℗ʻC) treatment. Second, to assess the effect of temperature fluctuation frequency on oxidative stress, eggs were incubated in one of three fluctuating incubation regimes; 28.7+3℗ʻC sinusoidal fluctuations every 12 (Hyper), 24 (Normal), or 48 hours (Hypo). Third, we tested the influence of average incubation temperature on hatchling oxidative damage and total antioxidant capacity (TAC). To test this, eggs were incubated in daily sinusoidally fluctuating incubation temperature regime with a mean temperature of 26.5 ℗ʻ C (Low), 27.1 ℗ʻC (Medium), or 27.7 ℗ʻC (High). We report that regardless of any thermal manipulation, no treatment effects on hatchling accumulation of oxidative damage were observed. This finding suggests that T. scripta hatchlings have sufficient antioxidant defenses that effectively protect individuals from temperature induced oxidative stress during incubation. However, hatchling TAC was influenced by both temperature fluctuation frequency and average incubation temperature. Following incubation with a low frequency of temperature fluctuations individuals had a reduced TAC, while incubation at a lower average temperature was associated with enhanced TAC. These results indicate that hatchling TAC is likely sensitive to the developmental thermal environment and may have important future consequences for hatchling fitness. In addition to temperature, we also saw that both oxidative damage and TAC were significantly related to clutch identity suggesting that there are either strong maternal or genetic factors influencing early life oxidative status of T. scripta.

Temperature-dependent sex determination (TSD) is a well-studied form of environmental sex determination in reptiles. The red-eared slider turtle (Trachemys scripta elegans) exhibits a form of TSD where cool incubation temperatures produce males and warm incubation temperatures produce females. In species with TSD, gonadal differentiation is regulated by network of genes and hormones that induce male or female development. TSD is most frequently studied in the laboratory using constant male and female incubation temperatures, which fail to capture the variability that organisms experience in the wild. We use ecologically relevant laboratory incubations to better understand the underlying mechanisms organizing the sex-determining process in T. scripta. Fluctuating incubations and exogenous hormones are used to produce ecologically relevant expression profiles for sex-determining genes and to decouple the effects of temperature and hormone environment on gonadal differentiation. This research represents the first time that fluctuating temperatures and hormones have been used in concert to characterize the molecular mechanisms underlying TSD and highlights the value of using laboratory conditions that more closely mimic environmental conditions.

Book excerpt: All jawed vertebrates have innate and adaptive immune responses, and the same major components of these responses, including toll-like receptors, macrophages, complement, T cells, and B cells. However, despite this broad similarity, there is much natural variation in immune responses both within and among taxa. This variation may be explained by the fact that an organism can allocate resources toward different components of its immune system in response to varying pressures. Examples of pressures include environmental changes, reproductive state, or exposure to pathogens. I define an organism's immune strategy as how an organism utilizes its various components of its immune system as a result of such pressures. The goal of my dissertation was to explore the immune strategy of a long-lived ectotherm, the red-eared slider turtle, Trachemys scripta. Specifically, I examined the effects of age, temperature, and steroids on a variety of immune measures in a natural population of sliders. I found evidence for a humoral immune response that relies on innate, natural antibodies rather than adaptive, specific antibodies. I also found little evidence for a decline in humoral immune responses with age when assays were run at a constant temperature. However, B cells from older individuals did respond less to antigen stimulation than B cells from younger turtles when assays were run at higher temperatures. I also found no evidence for an effect of estrogens on the development of the red-eared slider turtle. Overall, my research suggest the red-eared slider uses an immune strategy that relies heavily on innate responses rather than adaptive.

Endocrine disrupting compounds (EDCs) can mimic the effects of steroids and can affect the same suites of phenotypic traits during embryonic development. Bisphenol-A (BPA) is an EDC that mimics the action of estrogen, and recent work is beginning to implicate BPA in effects on learning and behavior similar to those caused by estrogen treatment studies. Red-eared slider turtles ( Trachemys scripta) are a good system to investigate the effects of BPA on learning and behavior, both because the molecular underpinnings of the mechanistic action of BPA endocrine disruption are generally understood, and because maternal estrogens are limited to egg components allowing for controlled, intra-clutch treatment groups. We exposed T. scripta eggs to BPA during embryonic development, and tested hatchlings for effects on learning and behavior in modified T-mazes. Innate biases in arm choice during the training phase of the experiment limited our ability to assess learning. Time of day and day of experiment both had significant effects on behaviors we investigated, and we found no BPA treatment effects on behaviors. However, we found that hatchling turtles were highly individually repeatable in their behaviors. These repeatable behaviors varied between individual hatchlings, suggesting that there are discrete behavioral types in T. scripta hatchlings. The highly repetitive nature of behaviors might explain the innate biases that prevented us from examining learning with our experimental design.
