Book excerpt: The gypsy moth, Lymantria dispar (L.), has been a major pest species in North American forests for >100 years and has caused defoliation on >369,000 km2 since 1924. Due to the economic and ecological consequences of gypsy moth, many aspects of its population biology and ecology have been studied. However, much prior work has focused at local spatial scales and in established outbreaking populations. In contrast, gypsy moth invasion dynamics are understudied in newly established populations and across landscape scales. I first analyzed the effect of sub- and supraoptimal temperatures on nascent gypsy moth population growth rates, and the spatial patterns of growth rates, along the expanding invasion front in the eastern United States. Second, I quantified the relationship between primary and secondary host plant fragmentation and gypsy moth population growth rates along the expanding invasion front. The data from this study indicate that there are geographical differences in how temperature and host fragmentation is affecting gypsy moth growth rates, and the spatial structure of those growth rates within expanding populations along the leading edge of the invasion front. Furthermore, the results from this study emphasize the important role that secondary hosts play in establishing populations. The results from this study could help in the development of more accurate risk assessment models pertaining to gypsy moth invasion potential.

Book excerpt: Insect nucleopolyhedroviruses (NPVs) are among the most important pathogens that regulate insect populations. Although these pathogens have been the subject of a great deal of research, many aspects of their transmission remain poorly understood. The transmission of NPV is controlled by a complex set of interactions between host, virus, and the environment. I investigated the effects of three different factors on the transmission of LdNPV in the gypsy moth, (Lymantria dispar), using laboratory and field experiments, DNA analysis, and mathematical modeling. The three effects examined were: 1) the effect of density-dependent disease resistance, 2) the effect of vectoring by avian predators, and 3) the potential occurrence of vertical transmission of the virus. My main results are as follows: 1) The resistance of gypsy moth larvae to NPV decreased as larval density increased, perhaps due to the stress associated with high densities, and this relationship is predicted to have a stabilizing effect on population dynamics. 2) In aviary cage experiments, bird species showed differential effectiveness in spreading the virus from infected to healthy larvae on a local scale. This difference was due more to differences in prey processing behaviors among the birds than to differences in the amount of NPV or speed at which NPV passed through the bird guts. The highest level of transmission was achieved by red-eyed vireos, largely because of their strategy of beating larvae against a branch before ingestion, resulting in rupture of the larval cuticle and scattering of viral occlusion bodies. 3) We found no evidence for vertical transmission of lethal NPV infection in the gypsy moth. Although low levels of virus were detected in the tissues of adult female moths by real-time PCR, there was no organ that showed consistently positive levels of NPV. Furthermore, the putative virus from female tissues was not infectious to larvae in our studies, and therefore may represent either latent virus or non-occluded virions. Together, these results contribute significantly to our understanding of the processes governing NPV transmission in gypsy moths and are directly relevant to the host-pathogen dynamics of many other systems.

The manners in which bacterial symbionts of many insects are acquired, or may be influenced by plant chemical defenses are central to these relationships. Gypsy moth (Lymantria dispar L.) is a polyphagous leaf-feeding Lepidoptera. I assessed the contributions of egg-mass and foliar sources of bacteria on larval midgut communities, using culture-independent analyses. Laboratory-reared egg masses had a very low diversity of bacteria, while field-collected populations were highly diverse. Despite differences in egg mass-associated bacteria, larval midguts converged after extended feeding. Larval bacterial communities resembled those of the foliage they consumed, but also differed in the abundances of some members. This indicates various foliar-acquired bacteria respond differently to midgut conditions. To identify roles of midgut bacteria on gypsy moth performance, I used artificial diets containing phenolic glycosides, the principal chemical defense of trembling aspen (Populus tremuloides). Larvae were fed bacteria from aspen leaves, field-collected fifth-instar larvae, or mock controls. Larvae inoculated with bacteria from aspen grew larger when phenolic glycosides were included in diet, but not when they were absent. Effects were post-ingestive, as consumption was unaffected. Specific bacteria in larval midguts were differentially affected: Ralstonia decreased and Acinetobacter increased, when phenolic glycosides were added. Bacterial communities demonstrated high capabilities of degrading phenolic glycosides. Among individual members, Acinetobacter most strongly reduced their concentrations. Aspen genotype had little effect on foliar communities, despite widely differing chemical compositions. However, midguts from gypsy moth larvae feeding on trees with different chemical concentrations were strongly influenced by the ratio of phenolic glycosides to condensed tannins in foliage. The limited effects of foliar chemistry on bacterial communities, coupled with the strong effects of these chemicals on bacteria within larval midguts following ingestion, indicate that partitioning of defense chemicals in plant cell vacuoles minimizes effects on phyllosphere residents, but, feeding ruptures these barriers, causing strong interactions between foliar compounds and bacteria within midguts. Overall, these results broaden our understanding of plant-insect-microbe relationships, and identify important interactions among these members.
