New technologies, from basic tools like leaf color charts to sophisticated sensor-based systems and laser land leveling, can reduce the dependency on laboratory assistance and manual labor. Another approach concerns the development of crop plants through genetic manipulations that allow them to take up and assimilate nutrients more efficiently, as well as identifying processes of plant responses to nutrient deficiency stress and exploring natural genetic variation. Though only recently introduced, the ability of microbial inoculants to induce NUE is gaining in importance, as the loss, immobilization, release and availability of nutrients are mediated by soil microbial processes.

Book excerpt: Increases in atmospheric CO2 concentration during the last 250 years are unequivocal, and CO2 will continue to increase at least for the next several decades (Houghton et al. 2001, Keeling & Whorf 2002). Arid ecosystems are some of the most important biomes globally on a land surface area basis, are increasing in area at an alarming pace (Dregne 1991), and have a strong coupling with regional climate (Asner & Heidebrecht 2005). These water-limited ecosystems also are predicted to be the most sensitive to elevated CO2, in part because they are stressful environments where plant responses to elevated CO2 may be amplified (Strain & Bazzaz 1983). Indeed, all C3 species examined at the Nevada Desert FACE Facility (NDFF) have shown increased A{sub net} under elevated CO2 (Ellsworth et al. 2004, Naumburg et al. 2003, Nowak et al. 2004). Furthermore, increased shoot growth for individual species under elevated CO2 was spectacular in a very wet year (Smith et al. 2000), although the response in low to average precipitation years has been smaller (Housman et al. 2006). Increases in perennial cover and biomass at the NDFF are consistent with long term trends in the Mojave Desert and elsewhere in the Southwest, indicating C sequestration in woody biomass (Potter et al. 2006). Elevated CO2 also increases belowground net primary production (BNPP), with average increases of 70%, 21%, and 11% for forests, bogs, and grasslands, respectively (Nowak et al. 2004). Although detailed studies of elevated CO2 responses for desert root systems were virtually non-existent prior to our research, we anticipated that C sequestration may occur by desert root systems for several reasons. First, desert ecosystems exhibit increases in net photosynthesis and primary production at elevated CO2. If large quantities of root litter enter the ecosystem at a time when most decomposers are inactive, significant quantities of carbon may be stored belowground in relatively recalcitrant forms. Indeed, a model-based analysis predicted that the arid/semiarid southwestern bioclimatic region had one of the highest rates of net carbon storage in the United States over the past century (Schimel et al. 2000). Second, root systems of desert plants are often extensive (Foxx et al. 1984, Hartle et al. 2006) with relatively large proportions of roots deep in the soil (Schenk & Jackson 2002). Thus, an understanding of belowground processes in desert ecosystems provides information on the potential for terrestrial carbon sequestration in desert ecosystems.

There is high uncertainty in how this disturbance will impact soil organic carbon (SOC) stocks. Quantifying SOC stocks and susceptibility to disturbance in deserts is critical because of the widespread geographic distribution of arid ecosystems. This thesis investigates the long-term effects of eCO2 on SOC stocks at the Nevada Desert Free-Air Carbon Dioxide Enrichment Facility (NDFF). Chapter one compares two methods of soil inorganic carbon removal on SOC stock measurements and how these methods differ by CO2 treatment. We find that 30% of SOC can be lost using acid washing relative to fumigation and that this effect is greatest in soils from control plots at NDFF. These results indicate that SOC stabilization may change after long-term exposure to eCO2. Chapter two directly applies the finding from chapter one to understand variation in SOC stock measurements from NDFF across multiple studies. The result of a direct comparison demonstrates how statistical choices, acid pretreatment methods, and heterogeneity of soils can impact our understanding of eCO2 effects at NDFF. Chapter three quantifies changes in SOC stability using a two-pool density fractionation. We find that eCO2 decreases fast-cycling particulate organic matter and does not affect slower-cycling mineral associated organic matter. Chapter four quantifies eCO2 effects on microbial mineralization in NDFF soils. We measure soil C priming with a 13C-labelled glutamic acid substrate using a short-term incubation and find eCO2 effects are strongest in water amended soils. Priming is not affected by eCO2 in water or C amended soils these conditions but is negatively correlated with SOC concentration. Together, these chapters indicate NDFF soils are primarily C limited and the increase in organic C availability from eCO2 treatment drives loss of SOC stocks. Addition of C in the form of acidic root exudates may destabilize organic matter and promote mineralization via increased microbial access, particularly from the C-rich particulate organic matter pool. These results emphasize the need to study multiple drivers of global change in arid ecosystem to fully understand impacts on SOC stocks and the global C cycle.

Together, these organisms form a community known as biological soil crust (biocrust). Microbes and mosses are major components of the biocrust, which can form on exposed soil surfaces as well as in microhabitats underneath semi-translucent quartz rocks in the Mojave Desert. The literature has characterized microbial diversity in Mojave Desert biocrusts without moss, and the Fisher lab has previously characterized moss community composition under quartz rocks and adjacent surface microsites; however, the other organisms that comprise biocrusts were not investigated. To better understand the degree to which mosses influence microbial community structure and function, we used environmental sequencing of 16S rRNA to characterize microbial communities in four biocrust microhabitats. In the Western Mojave Desert, we collected samples from soil surface and hypolithic (under quartz) biocrusts with and without moss, followed by microbial community DNA extraction and sequencing. We hypothesize that hypolithic microbial communities will harbor higher taxonomic diversity due to mitigated moisture, temperature, and light regimes as compared to surface soil microhabitats. In both surface and hypolithic habitats, we predict that the presence of moss will again increase the diversity of the microbial community, perhaps by providing additional, heterogeneous habitat and supplemental carbohydrates.
