Book excerpt: Although serious interest in studying the role of central cho linergic processes in psychopathology is just beginning to emerge, experimental literature on the part played by cholinergic mechanisms in brain behavior. reiations is quite extensive. During the past thirty years, cholinergic research has contributed significantly to the characterization and differentiation of adaptive mechanisms in volved in input selection, perception, cortical, autonomic and behav ioral activation, learning, memory, and inhibitory control of behav ioral outputs. To say that dysfunction of one or more of these mech anisms may be at the root of neuropsychiatric illnesses such as schiz ophrenia would be stating the obvious. This book examines the part cholinergic processes might play in dysfunctions of the adaptive processes involved in higher brain func tions and their significance for the pathogenesis, classification, etiology, and treatment of psychopathological conditions. In a series of wide ranging reviews of the available information, the subject is discussed from a variety of perspectives, using data derived from both experimental and clinical research. The purpose is not so much to determine whether cholinergic excess or deficiency is causal in this or that neuropsychiatric syndrome, but rather to try to understand the disease mechanisms in terms of adaptive processes in which cholinergic systems seem to play an important part.

Book excerpt: Even if the “weapons of mass destruction” (WMD) and, among them, stocks of organoph- phorus (OP) agents (also referred to as war gases and nerve gases) were not found in Iraq following the US-Iraq war, the relative ease with which these substances can be made from harmless precursors and the low cost of their manufacture will continue to fascinate pow- hungry, ruthless dictators, as well as multinational and international terrorists, particularly as the close relationship between the OP agents and useful insecticides makes it easy to disguise the importation and purchase of small amounts of the precursors. Indeed, the use by Saddam Hussein of a nerve gas against the Kurds and his possible employment of the OP agents during his war with Iran, and the Sarin attack in the Tokyo underground by an extremist religious set magnetized the world with respect to the OP drugs. As these drugs exert their toxicity via their cholinergic action on the nervous, particularly central nervous, system, it is no wonder that the research in the cholinergic ? eld attracts, and merits, our intense attention. These considerations underlie the signi? cance of this book, as Alex Karczmar devotes an entire chapter of Exploring the Vertebrate Central Cholinergic Nervous System to anticholinest- ases (antiChEs), and as he is an acknowledged expert in the ? eld of cholinergic toxicity as well as a consultant to the Surgeon General of the U. S. Army.

Book excerpt: Physiological Pharmacology: A Comprehensive Treatise, Volume III: The Nervous System — Part C: Autonomic Nervous System Drugs focuses on the effects of drugs on the nervous system, including cardiovascular effects and metabolism. The selection first offers information on the cardiovascular effects of choline esters and the effects of choline esters on smooth muscles and secretions. Topics include actions of acetylcholine, physiological role of choline esters, and actions of cholinergic drugs on smooth muscles and secretions. The book also takes a look at the pharmacologic, toxicologic, and therapeutic properties of anticholinesterase agents, including the chemistry and metabolism of anticholinesterase substances and mechanisms of action of anticholinesterase agents and role of cholinesterases. The publication examines muscarinic blocking drugs and ganglion-blocking drugs. Discussions focus on actions on the secretory glands and cardiovascular system; distribution, metabolism, and excretion of atropine; substances that block ganglionic transmission; and cardiovascular effects. The text also elaborates on neuromuscular blocking drugs. The selection is a dependable reference for readers interested in the effects of drugs on the nervous system.

Book excerpt: The primary objective of this 4-volume book series is to educate PharmD students on the subject of medicinal chemistry. The book set serves as a reference guide to pharmacists on aspects of the chemical basis of drug action. Medicinal Chemistry of Drugs Affecting the Nervous System is the second volume of the series and it presents 8 chapters focusing on a comprehensive account of drugs affecting the nervous system. The volume informs readers about the medicinal chemistry of relevant drugs, which includes the mechanism of drug action, detail structure activity relationships and metabolism as well as clinical significance of drugs affecting autonomic and central nervous system. Chapters in this volume cover cholinergic drugs, adrenergic drugs, antipsychotics, antidepressants, sedatives, hypnotics, anxiolytics, antiepileptic drugs, anesthetics and antiparkinsonian drugs, respectively. Students and teachers will be able to integrate the knowledge presented in the book and apply medicinal chemistry concepts to understand the pharmacodynamics and pharmacokinetics of therapeutic agents in the body. The information offered by the book chapters will give readers a strong neuropharmacology knowledge base required for a practicing pharmacist.

Available in PDF, EPUB and Kindle. Book excerpt: Cholinergic-Monoaminergic Interactions in the Brain investigates dynamic synaptic mechanisms in the brain and their links to various behaviors and related functional processes. More specifically, it explores neurochemical interactions by focusing on the relationship between cholinergic and monoaminergic mechanisms in the central nervous system. It also presents morphologic data to highlight the importance of structure in understanding function, particularly, that of the neostriatum and the role of acetylcholine and neurochemicals. Organized into six sections comprised of 18 chapters, this volume begins with an overview of various models of cholinergic-monoaminergic and other chemical interactions, based in part on interactive processes that are believed to operate in the peripheral nervous system. The next chapters focus on experimental observations on electrophysiologic and neurochemical interactions in the brain and the functional correlates of such interactions. The discussion then turns to the neurohistochemical and neurobiochemical aspects of cholinergic-monoaminergic interactions, the effects on neostriatal acetylcholine levels, and the synthesis of the cortico-neostriatal and thalamo-neostriatal pathways. The reader is also introduced to the effects of lesion in the globus pallidus, the electrophysiology of interactions, the effects of drugs thought to preferentially affect monoaminergic or related neurotransmission mechanisms on acetylcholine in the brain, and whether dopamine is excitatory or inhibitory upon neostriatal neurons. Finally, the book assesses the significance of central cholinergic-monoaminergic interactions for functional processes, both normal and pathologic. This book is a valuable resource for neuroscientists, pharmacologists, histochemists, behavioral biologists, psychobiologists, and psychologists.

Book excerpt: The principal purpose of this study was to investigate the possible differential effects of LSD as a function of procedural variations in performance of delayed response tasks. In general, LSD was found to impair delayed response performance in the monkey. From the standpoint of dose-effect relationships, the results suggest a positive correlation between magnitude of dose and extent of functional impairment. A tendency toward rapidly developing tolerance was observed. A 'placebo effect' in response to the control agent (sterile water) was encountered in a significant number of monkeys.
