Book excerpt: Abstract: Nine papers developed from a 1981 international workshop summarize current knowledge of the role of proteins (and amino acids) in lipid metabolism and atherosclerosis for nutritionists, clinicians, and other health professionals. The topics range from studies on lipid metabolism and experimental atherosclerosis in animals (rats, rabbits, swine), to effects of proteins on human serum lipids and lipoproteins. Other papers discuss the effects of soy protein and casein, influences of dietary protein on serum cholesterol, and the immunological aspects of atherosclerosis. The results of these studies indicate that dietary protein can make a significant contribution to atherosclerosis, that soy protein produces lower serum cholesterol levels than casein (the dynamics of which are described), and that diets containing animal protein are generally more cholesterolemic and atherogenic (in rabbits, specifically) than diets containing vegetable protein. The protein effect can be modified by other dietary components (e.g., fiber, fat). Other findings (e.g., the role of dietary protein in the immunological aspects of atherosclerosis) also are reported. (wz).

Since the first of these symposia in 1960 these triennial meetings have been devoted to the exploration of new ideas, new data and new concepts related to lipid metabolism and atherosclerosis. The sixth Meeting was particularly stimulating in this regard. The concept of the "protective" action of HDL was thoroughly explored within the framework of its molecular biology with data on its epidemiological as well as its in vitro mechan ism(s) of action being discussed. The action of drugs on arterial and HDL metabolism was also discussed as were newer aspects of platelet aggregation, especially as related to prostaglandins. New ground was also broken in discussions of lipid mobilization and mechan isms of hypocholesteremia. The major topics covered in my courses were fatty acids, prostaglandins, leukotrienes, phospholipids, glyco lipids, triacylglycerols, cholesterol, bile acids, and plasma lipoproteins. Empha sis was placed on the regulation and disorders of lipid metabolism. The latter included hyperlipidemias, atherosclerosis, and alcohol-induced liver damage. In this volume, I have chosen to focus on the disorders of lipid metabolism at a level appropriate both for medical students and for graduate and undergradu ate students majoring in the biological sciences. The biochemistry, nutrition, genetics, and cell biology aspects of lipids and lipid metabolism will be covered as they relate to lipid disorders. I am not aware of any textbook that integrates the disorders of lipid metabolism in this manner. Chapter 1 includes a brief discussion of the basic structures, properties, and metabolism of lipids. This chapter is not very detailed, since the material covered is available in basic textbooks on biochemistry. The major fpcus of this volume is the various lipid disorders, with emphasis on polyunsaturated fatty acids, the molecular biology and pathogenesis of the hyperlipidemias, dietary and drug therapy for the hyperlipidemias, and alcohol-induced liver damage. The material presented has been obtained from several textbooks on biochemistry and from a variety of recent articles in the scientific literature.

Since the first symposium in 1960, each successive meeting has broken new ground in the field of pharmacological control of lipid levels - offering new and stimulating insights and exposing the audience to the state of the art. The field has progressed sufficiently to permit discussion of the cellular biology of athero sclerosis. The opening session was devoted to pathology, macrophages, lipoproteins and their receptors and choles terol ester metabolism. Because of the recent emergence of new apolipoprotein technology, a workshop devoted sole ly to apolipoprotein methodology was introduced followed by a plenary session devoted to their metabolism and structure. Another rapidly developing area of atherosclerosis research is non-invasive assessment of this condition. Accordingly, a session was devoted to new techniques for this research modality. The final plenary sessions were devoted to the roles of drugs and diet in athero~ scl~rosis - cause, treatment and mechanisms of action. The meeting was summarized by Dr. O.J. Pollak, one of the "founding fathers" of this field. There were nine sessions of proffered papers whose abstracts appear in this volume. In addition, special workshops (to be reported elsewhere) were devoted to several drugs including Oiyzanol, Probucol and Etofibrate.
