Covalent linear arrays of chromophores or other molecular assemblies such as liquid crystals, zeolites, polymers, peptides, and amphiphiles have all been used as components for this purpose. Significant amount of this effort also involved the use of dendrimers as the molecular architecture. The structural feature in which multiple functionalities are present in the periphery that decreases gradually as one moves towards the core renders dendrimers obvious candidates for light harvesting antenna. Most of the efforts reported in the literature are directed towards energy funneling from a chromophore in the periphery to another chromophore at the core of the dendrimer. There are relatively few reports that utilize the dendritic architecture for photoinduced charge separation, an important step in designing materials for photovoltaics. These reports focus mostly on conjugated molecular backbones. Since non-conjugated dendrimer backbones provide the possibility of independently tuning the electronic characteristics of the chromophore and the charge transfer unit and therefore carry out a systematic structure-property relationship study, we have designed and synthesized dendrimers.

Book excerpt: Dendrimers are versatile polybranched polymers that intend to exhibit monodispersity and globular shape at large generations. Their assembly is iterative in nature and involves multi-step synthesis. Dendrimers were first reported in 1985 by Tomalia and Newcome. The versatility of dendrimers is based on the possibility of controlling their dimensions and their capability of bearing a defined amount of cargo on the end point of their branches (multivalency), and even the property of storing some other smaller molecules inside them (Chapter 1). High generation dendrimers display multiple challenges for their synthesis that involve solubility, structural defects and long times for their synthesis. Those are the main reasons that have allowed to just few designs of dendrimers to reach generation 9. After several years of exploration and understanding the reactivity of several amine linkers towards the chemeoselective substitution of cyanuric chloride, two different families of triazine dendrimers have overpassed generation 9 (Chapter 1). Since the traditional methods of synthesis for large generation triazine dendrimers take months for their completion, a new synthetic approach was needed. Microwave assisted reactions were found to be the fastest route to reduce times of reaction for triazine dendrimers from months to just few hours (Chapter 2). The rapid production of triazine dendrimers and their good water solubility, allowed us to explore diverse hybrid designs that provided insights about solubility and structural conformation of triazine dendrimers (Chapter 3). Also, the excellent water solubility properties of 4,7,10-trioxa-1,13-tridecanediamine based dendrons were used in the synthesis and stabilization of gold nanoparticles, that displayed homogeneity in solution for more than a year (Chapter 4). Furthermore, blood clotting properties of triazine dendrimers were studied for possible solutions to bleeding disorders and a conceptual design of multifunctional prostate specific membrane antigen (PSMA) targeted dendrimers was pursued (Chapter 5).

Book excerpt: A variety of dendrimers based on melamine are designed, synthesized, and evaluated for drug delivery systems. The synthesis of a dendrimer, including multiple copies of four orthogonally reactive groups, is described. The three groups on the surface are nucleophilic and include four free hydroxyl groups, four tert-butyldiphenylsilyl (TBDPS) ether groups, and sixteen amines masked as tert-butoxycarbonyl (BOC) groups. The core of the dendrimer displays two electrophilic monochlorotriazines. The dendrimer above is further manipulated for in vivo biodistribution: incorporation of the reporting groups Bolton-Hunter and DOTA (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid); PEGylation for biocompatibility and size tuning. In preliminary biodistribution studies, dendrimers circulate in the blood for a longer time as the molecular weight increases, which is important to passively target tumor tissues via the EPR effect. Also, high uptake by the tumor tissues was observed in mice bearing prostate cancer xenografts. A drug delivery vehicle for the anticancer agent paclitaxel is described. This drug delivery vehicle contains sixteen molecules of paclitaxel via acid-labile ester linkage, two Bolton-Hunter groups, and sixteen monochlorotriazine groups for PEGylation. The in vitro drug release studies shows faster release of paclitaxel at lower pH in PBS.
