Available in PDF, EPUB and Kindle. Book excerpt: This thesis is geared towards using hydrogel nanoparticles in various biotechnological applications. The polymer that was used in making these nanoparticles was poly(N-isopropylacrylamide), which is a thermoresponsive polymer. These particles were used in making fast responsive polymer films, which can be used in optics. It was observed that the rate of deswelling increased as the concentration of the nanoparticles in the film was increased. These particles were also used in making photoresponsive materials. In this case a photoresponsive dye (malachite green) was conjugated to these nanoparticles and in presence of light of appropriate wavelength the particles undergo a phase transition. A core/shell construct was synthesized where the core was composed of degradable cross-links and the shell of composed of non-degradable cross-links. The degradable cross-linker had vicinal diols, which can be cleaved by sodium periodate. Hence after degrading the core, hollow particles were obtained. Zwitterionic particles were made by incorporating a cationic and anionic comonomer. These microgels go from a positively charged state to zwitterionic to negatively charged state on increasing the pH. One of the important potential applications for these microgels is drug delivery. Microgels were used for targeting cancer cells. Folic acid was used as the targeting ligand. The microgels were conjugated with folic acid and were able to target cells that overexpress folate receptors. In one other application core/shell microgels were made which exhibit pore-size dependent permeation of proteins.

Book excerpt: Since antiquity, humanity is always trying to find different materials for various purposes from nature. With advances in modern technology, different material, such as polymers, ceramics, and metal alloys, have been found or synthesized and people began to use them to repair the damaged part of body. These materials have sometimes shown better performance, desired functionality, and long-time stability than the naturally derived materials. Among the commonly used biomaterials, hydrogel is one of the most versatile because of its flexible structure, excellent biocompatibility and multi-functional biomedical applications, e.g. wound healing, drug delivery, biosensor, cell culture, and tissue engineering. Among the various hydrogel, poly(ethylene glycol) (PEG)-hydrogel has been widely used for various biomedical applications due to its flexible structure, reproducible properties, and excellent biocompatibility. 3D cell culture has received broad attention for its ability to mimic cell living conditions in vivo. Hence, many studies have been done to find an appropriate scaffold for encapsulating cells that permits cell attachment, promotes both cell growth and differentiation, and is also biocompatible, biodegradable and mechanically strong for processing. However, like other synthetic biomaterials, PEG lacks biofunctional components and informational structure for cell binding, growth and differentiation. Here, we present the work focused on the synthesis of a novel PEG 3D-hydrogel system, which utilizes the high affinity between biotin and avidin to (i) crosslink the PEG to form a 3D scaffold, and (ii) immobilize cell binding ligands to the gel matrix. This relatively inexpensive chemically defined 3D-gel matrix platform has high water content supports cell division, proliferation and migration. By varying the size of PEG, the mechanical property of the PEG 3D-hydrogel can be easily tuned. With the introduction of biotinylated-LXY30, an [alpha]3[beta]1 integrin binding peptide, the 3D-hydrogel can support the growth and proliferation of U87MG human glioblastma cells for over a week. Moreover, considering its moderate gel formation speed after mixing PEG-avidin and PEG-biotin, this PEG 3D-hydrogel has great potential to be used as microfluidic chip 3D medium for cell biology study. Here, we also report the development of an original method to fabricate a super water stable, one-step photopatternable and high electrical conductive PEG material coated with poly(3,4-ethylenedioxythiophene) polystyrene sulfonate (PEDOT: PSS). The PEDOT: PSS/PEG hybrid system shows high flexibility, stretchability and transparency, which is great beneficial for various applications. Different from the original PEDOT: PSS film which can be easily destroyed by water, the obtained hybrid film after UV exposure is super water stable. The conductivity does not show obvious decrease (less than 10%) after being submerged in water for 10 days or even suffering from ultra-sonic in water for 180 minutes. To further explore the property of obtained PEDOT: PSS/PEG hybrid material, a simple one-step photolithography method was developed to pattern PEDOT: PSS with resolution down to 2 [mu]m. In addition, this hybrid film is mechanically flexible and stretchable due to strong adhesion between PEDOT: PSS and PEG. Here, we have exploited the water stability property of this novel conductive PEG material to develop a series of wearable sensing detection devices to monitor environmental humidity level, sense humidity change closing to water surface, and track human breath and sweat production rate. Since long time stability for organic electronics has been a problem for long time, this water stable, flexible, transparent, and electrical conductive PEDOT: PSS/PEG hybrid material has huge potential in many other biomedical applications. In addition, it can potentially be used in conjunction with the PEG 3D-hydrogel to develop microfluidic chip for cell biology studies.
