Book excerpt: A novel ion mobility-mass spectrometer was designed and built in order to achieve high transmission and high resolution for observing desolvated ion conformations of chemical and biological molecules in the gas phase. The instrument incorporates a home-built electrospray ionization (ESI) source and rf ion funnel(IF) as an ion transfer interface, modular periodic-focusing ion guide (PFIG) as a drift tube (DT) for arrival time measurement, and orthogonally accelerated linear time-of-flight mass spectrometer to maximize analytical figures-of-merit of the instrument including sensitivity and separation of gas-phase ions. The rationales for implementation of each device aforementioned are discussed as enhanced ion focusing to reduce data acquisition time of the instrument, high mobility resolution (R > 56) to separate ions having a small collision cross section (CCS) difference at low-to-intermediate field strength (10 V cm−1 torr−1~19 V cm−1 torr−1), and observation of multiple conformers from isobaric ions at various operation conditions. Gate and transfer ion optic design needed to avoid detrimental gas dynamic effect is presented during hybridization of PFIG into ESI mass spectrometer. The details of instrumentation and electrode/voltage schematics are also discussed along with SIMION calculation results. The characteristic drift motions of ions inside the PFIG require a normalization procedure to compare CCS values from the instrument with the results using conventional uniform field DT ion mobility-mass spectrometry (IM-MS). Through experimental measurements and ion trajectory calculations from well known model peptides and proteins having multiple charge states, a correlation factor a which links the results from the two different types of instrumentation was derived and successfully applied (within ±4% difference). Ion heating was also investigated using ubiquitin, insulin, and substance P. Using ESI PFIG-MS, a significant variation of the arrival time distribution at different heated capillary temperature, rf amplitude, and electric field strength was observed for ubiquitin (+7) and (+8) and substance P (+3), while a substantial change was not observed for insulin owing to structural rigidity of disulfide bond cross-linking between two chains. The results imply that ESI PFIG-MS provides a window to monitor conformational variation of gas-phase ions and measure energy barriers of peptide/protein folding processes, which can allow us to reveal pathways in structural energy landscape. Implants of rubidium and krypton in rhenium ribbon filaments were subsequently tested in a bakeable 60 deg sector mass spectrometer operating in the static mode. Surface ionization and electron impact ion sources were both used, each yielding satisfactory results. The metallic implant with subsequent ionization provided a means of mass scale calibration and determination of system operating parameters, whereas the noble gas thermally desorbed into the system was more suited for partial pressure and sensitivity determinations.

Though there have been numerous advances in mass analyzer technology over the decades, there has been comparatively little advancement in mass spectrometer detector technology. The development of the scientific charged-coupled device over 30 years ago brought the advantages of simultaneous detection over single channel detection to optical spectroscopy, including higher signal-to-noise ratios for a fixed analysis time, shorter analysis time to obtain a given signal-to-noise ratio, and greater sample throughput. While the use of array detectors to achieve simultaneous detection is commonplace in optical spectroscopy, ion detectors for mass spectrometry have lagged behind. Over the last decade, a new type of ion detector, the capacitive transimpedance amplifier (CTIA) array detector, has been developed that has a number of properties that make it an excellent tool for simultaneous detection using dispersive mass spectrometers. The CTIA array detector has high sensitivity as well as high gain stability, allowing it to excel in applications that require high precision measurements of ion signals, such as isotope ratio mass spectrometry. Capacitive transimpedance amplifier array detectors have previously been used to demonstrate the power of simultaneous detection on Mattauch-Herzog double focusing mass spectrometers, but the non-linear mass dispersion of these instruments means that the resolution is not constant across the array. A different type of dispersive instrument, the linear cycloid, has a linear mass dispersion, making it a good candidate for an array detector. The first detailed characterization of gain, read noise and dark-current noise, as well as of operating behavior over a range of temperatures, of the DM0025, a 1696 pixel CTIA array detector was performed. In addition, the first-ever combination of a CTIA array detector with a linear cycloid mass spectrometer was developed. This combined instrument demonstrated simultaneous detection of multiple masses, as well as a linear mass range. The results from the detailed characterization of the detector were used in conjunction with measurements of the performance of the combined instrument to suggest improvements for the next generation of linear cycloid instruments with CTIA array detectors.
