This work is intended to serve as a primer for basic confocal microscopy theory as well as a description of the design and construction of a homebuilt system in the Elson Lab at the Washington University School of Medicine. A description of the different designs of confocal microscopes is provided, and the capabilities, benefits, and detriments of each is described. The thought process behind each design decision is explained leading to the final system design. A more in depth description of the final design is provided in Chapters Two and Three, but, in short, the final design is a non-descanned, laser scanning, two-photon confocal microscope. The system is designed to integrate as seamlessly as possible with the existing system and to allow for all existing functionality to remain. When completed, the system will be capable of one- and two-photon fluorescence correlation ii spectroscopy (FCS); low-speed, one-photon confocal imaging using a piezo-stage; and high-speed, two-photon confocal imaging using the laser scanning system. Finally, future directions as well as the limitations of the system are also described. All resources necessary to continue work on the system as well as those necessary to use it are provided. This includes: System diagrams, Optical layout plots, Component data sheets, LabVIEW program. These resources are intended to make using, modifying, and improving the system much simpler.

Available in PDF, EPUB and Kindle. Book excerpt: This handbook provides a comprehensive review of the entire field of laser micro and nano processing, including not only a detailed introduction to individual laser processing techniques but also the fundamentals of laser-matter interaction and lasers, optics, equipment, diagnostics, as well as monitoring and measurement techniques for laser processing. Consisting of 11 sections, each composed of 4 to 6 chapters written by leading experts in the relevant field. Each main part of the handbook is supervised by its own part editor(s) so that high-quality content as well as completeness are assured. The book provides essential scientific and technical information to researchers and engineers already working in the field as well as students and young scientists planning to work in the area in the future. Lasers found application in materials processing practically since their invention in 1960, and are currently used widely in manufacturing. The main driving force behind this fact is that the lasers can provide unique solutions in material processing with high quality, high efficiency, high flexibility, high resolution, versatility and low environmental load. Macro-processing based on thermal process using infrared lasers such as CO2 lasers has been the mainstream in the early stages, while research and development of micro- and nano-processing are becoming increasingly more active as short wavelength and/or short pulse width lasers have been developed. In particular, recent advances in ultrafast lasers have opened up a new avenue to laser material processing due to the capabilities of ultrahigh precision micro- and nanofabrication of diverse materials. This handbook is the first book covering the basics, the state-of-the-art and important applications of the dynamic and rapidly expanding discipline of laser micro- and nanoengineering. This comprehensive source makes readers familiar with a broad spectrum of approaches to solve all relevant problems in science and technology. This handbook is the ultimate desk reference for all people working in the field.

Available in PDF, EPUB and Kindle. Book excerpt: What accounts for the persistence and spread of "commoning," the irrepressible desire of people to collaborate and share to meet everyday needs? How are the more successful projects governed? And why are so many people embracing the commons as a powerful strategy for building a fair, humane and Earth-respecting social order? In more than fifty original essays, Patterns of Commoning addresses these questions and probes the inner complexities of this timeless social paradigm. The book surveys some of the most notable, inspiring commons around the world, from alternative currencies and open design and manufacturing, to centuries-old community forests and co-learning commons - and dozens of others. David Bollier (www.bollier.org) is an American author, activist and independent scholar who has studied the commons for nearly twenty years. Silke Helfrich (commonsblog.wordpress.com) is a German author and independent activist of the commons who blogs at www.commonsblog.de, and cofounder of the Commons-Institut in Germany. With Michel Bauwens, Bollier and Helfrich are cofounders of the Common Strategies Group. For more information, go to the book's website, Patterns of Commoning (www.patternsofcommoning.org)
