Book excerpt: The near-field scanning optical microscope, or NSOM, proyides spatial resolution of surface features considerably smaller than the wavelength of the radiation used to image. We have focused on both the development and the use of the NSOM. In the former, we considered the confinement of optical fields to nanometric structures. We analyzed the delivery of light from the far-field to the near-field region in tapered optical fibers. Our analysis led to the design and development of near-field probes of that allow for the performance of relatively light-starved NSOM experiments. Using such probes, we have demonstrated a capability of using spectral contrast in near-field imaging. In studies of KTP, for example, we have performed nano-Raman spectroscopy samples, and have imaged sub-wavelength surfaces features using only Raman-scattered light. In ongoing research we have launced further efforts to improve probe design and have begun nano-Raman investigations Mercury Cadmium Telluride and semiconducting diamond.

Book excerpt: We present the development and application of a hybrid confocal and aperture near-field scanning optical microscope operating in reflection mode. It has been designed to work at cryogenic temperatures and in vacuum or with an exchange gas. Furthermore, it permits to split the fluorescence into two perpendicular polarization channels.We have studied various single nano-objects, such as molecules, CdSe/ZnSe nanocrystals, CdSe/ZnS nanorods and fluorescent nanospheres. As molecular samples we used diI-C18 molecules and molecules derived from anthracene. The ensemble absorption and emission spectra of the latter were measured, as well as their absorption cross-section and quantum yield in solution for the first time. Furthermore, we used time correlated fluorescence decay experiments that suggest the presence of different conformations in solution, already observed with similar molecules.We used our instrument in confocal mode to image, record time traces, and acquire some spectra of single nano-objects. Single nanospheres with a diameter of 27 nm are used to map the optical field distribution in the vicinity of the focal point of a microscope objective. The experimentally obtained images are qualitatively compared with a scalar model. The near-field scanning optical microscopy setup is validated using nanospheres with a diameter of 500 nm.The dynamic temporal behavior is examined with the help of time traces. The study is based on the autocorrelation function for single molecules, and using an on-off statistics for single nanocrystals/-rods. In the latter case a power law distribution of the on- and off-time probabilities is observed.

Book excerpt: In this program sponsored by the ARO Physics office we have made significant progress in designing, constructing and using a novel device called the Near-field Scanning Optical Microscope, or NSOM. The microscope is one of a family of instruments which use either small apertures or finely sharpened probes to scan the surface of a sample and thereby construct an image of that surface. Since demonstrating the utility of the microscope of our design, we have subsequently built a number of other NSOMs incorporating a variety of illumination modes and signal transduction methods in imaging surfaces and taking optical spectra of features on these surfaces. Theoretical efforts to model the optical throughput of the probe have been undertaken, and the initial results have been published. The program has supported several students, including a student who, after graduation, designed the first commercial NSOM instrument. Book excerpt: This project supported a new universal scanning microscope system combining near-field scanning optical microscopy (NSOM), conventional atomic force (AFM) microscopy, and confocal optical microscopy. To build a universal system capable of high-resolution topographical imaging concurrently with spectroscopic abilities, the authors have acquired several independent units and combined them into a custom-designed NSOM/AFM/Raman instrument. They have acquired an atomic force microscope (Multimode, Digital Instruments), a near-field scanning microscope (Aurora III, Digital Instruments, partially supported by NASA), and a SpectraPro Raman spectrograph from Roper. In addition to these major instruments, they acquired a number of supporting parts/instruments toward their goal of completion of the system and sample preparation, selection, and characterization for Raman studies (e.g., CCD camera, avalanche detectors, two lasers, optical parts, air table, fluorescence microscope, mini x-ray unit, etc). A post-doctoral Research Associate was in charge of putting all these parts together. The authors did not go with a single unit from WiTec because actual demonstrations did not show expected sensitivity. This report presents a description of the instrument and preliminary results obtained with it. The instrument was used to collect Raman spectra from patterned arrays of bent single wall carbon nanotubes and bundles (3 nm in diameter) and from freely suspended nanomembranes with encapsulated gold nanoparticles.

Book excerpt: In this project, we requested funds from the Air Force of Scientific Research (AFOSR) under DOD's Defense University Research Instrumentation Program (DURIP) to acquire a near-field scanning optical microscope (NSOM). Near-field scanning optical microscope (NSOM) is a unique research tool commercialized in the past two years, which provides unprecedented optical resolution defying the diffraction limit, as small as 1/20 laterally and 1/100 vertically. The funds from AFOSR has enabled us to purchase and install a state-of-the-art NSOM from TopoMetric. We have successfully installed and tested the instrument. A number of experiments have been performed using the instrument. The NSOM has proved to be a powerful instrument which allows innovative new approaches to address fundamental issues and enhance the existing and future DOD sponsored research projects in the University of Rochester. The preliminary application of the NSOM has already demonstrated the great potential of NSOM, its value to DOD sponsored research, and its usefulness for training future scientists and engineers for the DOD interests.
