Available in PDF, EPUB and Kindle. Book excerpt: Respiratory diseases are leading causes of death and disability globally, with about 65 million people suffering from COPD, and 334 million from asthma, the most common chronic disease. Each year, tens of millions of people develop and can die from from respiratory infections such as pneumonia and TB. Systemic inflammation may induce and exacerbate local inflammatory diseases in the lungs, and local inflammation can in turn cause systemic inflammation. There is increasing evidence of the coexistence of systemic and local inflammation in patients suffering from asthma, COPD, and other lung diseases, and the co-morbidity of two or more local inflammatory diseases often occurs. For example, rheumatoid arthritis frequently occurs together with, and promotes the development of, pulmonary hypertension. This co-morbidity significantly impacts quality of life, and can result in death for those affected. Current treatment options for lung disease are neither effective, nor condition-specific; there is a desperate need for novel therapeutics in the field. Additionally, the molecular and physiological significance of most major lung diseases is not well understood, which further impedes development of new treatments, especially in the case of coexistent lung diseases with other inflammatory diseases. Great progress has been made in recent years in many areas of the field, particularly in understanding the molecular geneses, regulatory mechanisms, signalling pathways, and cellular processes within lung disease, as well as basic and clinical technology, drug discovery, diagnoses, treatment options, and predictive prognoses. This is the first text to aggregate these developments. In two comprehensive volumes, experts from all over the world present state-of-the-art advances in the study of lung inflammation in health and disease. Contributing authors cover well-known as well as emerging topics in basic, translational, and clinical research, with the aim of providing researchers, clinicians, professionals, and students with new perspectives and concepts. The editors hope these books will also help to direct future research in lung disease and other inflammatory diseases, and result in the development of novel therapeutics.

Book excerpt: This report considers the biological and behavioral mechanisms that may underlie the pathogenicity of tobacco smoke. Many Surgeon General's reports have considered research findings on mechanisms in assessing the biological plausibility of associations observed in epidemiologic studies. Mechanisms of disease are important because they may provide plausibility, which is one of the guideline criteria for assessing evidence on causation. This report specifically reviews the evidence on the potential mechanisms by which smoking causes diseases and considers whether a mechanism is likely to be operative in the production of human disease by tobacco smoke. This evidence is relevant to understanding how smoking causes disease, to identifying those who may be particularly susceptible, and to assessing the potential risks of tobacco products.

Book excerpt: This book provides a comprehensive and up to date picture of the molecular mechanisms underlying acute lung injury, focusing both on the mechanisms of injury and of repair. Topics covered include the cells involved, their injurious products including pro-inflammatory cytokines, proteases and free radical species as well as the fibrotic response and its molecular regulation, the role of the immune system, airway repair and remodelling and transcription factors regulating inflammation. All the chapters are written by experts in their field and reflect the lecture given by these authors at a recent international meeting also entitled " Acute lung injury: from inflammation to repair", held as part of the ongoing Molecular Pathology Annual symposia at University College London. The meeting gathered together a wide range of scientists and clinicians all of whom are at the cutting edge of inflammation and repair as it relates to acute lung injury. It is the breadth of approach that makes the volume so useful.

Book excerpt: This important new book focuses on the involvement of cytokines in specific areas of inflammatory diseases, such as granulomatous responses, lung disease, hepatic dysfunction and the acute phase, arthritis and accompanying bone remodeling, neurogenic inflammation, and shock. The roles of GM-CSF, IL-6, IL-2, TGFb EGF, and LIF are discussed, as well as the medical treatments that affect cytokine activity. The results of approaches important to the biotechnology and pharmaceutical industries, such as the search for endogenous biological response modifiers that control cytokine function or production, attempts to synthesize heterocyclic compounds in the organic chemistry lab, and research regarding second messenger pathways involved in IL-1 and TNF production are examined. This book will provide anyone in cytokine research, especially clinical investigators, pharmaceutical industry researchers, and academic research scientists, with important information on how cytokine research might be used.
