Book excerpt: Over the last decade, cytokine research has emerged as one of the most exciting and critical fields for providing fundamental knowledge of normal and abnormal human development. Today, it is apparent that cytokines orchestrate growth from the early embryonic stage to maturity and are responsible for the normal function of virtually every organ system. Furthermore, virtually all disease states have been associated, at least in part, with cytokine aberrations. In this volume, the editors have brought together internationally known experts in the field of interleukin research to provide a comprehensive review of the biology of the interleukins and their role in both health and illness, while maintaining a balance between the basic science and clinical aspects. Cytokines: Interleukins and their Receptors should be of interest to a wide variety of researchers including clinical hematologists, oncologists, immunologists, in addition to medical and PhD students and researchers with an interest in cytokines.

Available in PDF, EPUB and Kindle. Book excerpt: Cytokines, a broad class of proteins, have a role in fighting infection and in mounting an immune response; in mediating cell differentiation and embryogenesis; in promoting cell growth; and in inhibiting growth or causing cell death. Recent advances in cell culture, monoclonal antibodies and recombinant DNA technology have made it possible to isolate these proteins and to obtain sufficient quantities to conduct clinical studies as well as laboratory experiments. Thus, the foundations have been laid for a new form of cancer therapy and for understanding how abnormalities in the cytokine network could contribute to the process of malignancy. Up-to-date and comprehensive, this book brings together current research and discusses it in the context of its present and potential clinical application. The book is intended for cancer researchers, clinical oncologists, and those involved in the pharmaceutical and biological product industries.

Available in PDF, EPUB and Kindle. Book excerpt: The driving force for research on cytokines has always been their clinical promise. Their biological properties suggested a key role in hematopoiesis, immunity, tumor genesis, hemostasis, vascularization, repair of connective tissues and integration of the immune system with the neuroendocrine system. Animal studies have shown that cytokines could be used as effective biotherapeutics with easily manageable and reversible toxicities. Clinical trials have confirmed these findings, culminating in the licensing of a number of the cytokines such as interferon alpha, interferon gamma, interleukin 2, erythropoietin, granulocyte colony stimulating factor, and granulocyte-macrophage colony stimulating factor. Many other cytokines are in clinical trials. This is the first comprehensive volume on the cytokines written primarily from a medical perspective. After presenting background information about the structure, production, assays and systemic effects of cytokines and their receptors, it is organized around diseases and organ systems. Infectious diseases, autoimmunity, immunodeficiency states, defective hematopoiesis, allergies, injury repair, cancer, vascular and skin diseases, and neurological disorders are all covered. This work reviews the role that cytokines play in the pathogenesis, diagnosis and therapy of each disease. The authors assess both the current state of the art and the potential for future applications.

Book excerpt: A link between inflammation and cancer has been established many years ago, yet it is only recently that the potential significance of this connection has become apparent. Although several examples of chronic inflammatory conditions, often induced by persistent irritation and/or infection, developing into cancer have been known for some time, there has been a notable resistance to contemplate the possibility that this association may apply in a causative way to other cancers. Examples for such progression from chronic inflammation to cancer are colon carcinoma developing with increased frequency in patients with ulcerative colitis, and the increased incidence of bladder cancer in patients suffering from chronic Schistosoma infection. Inflammation and cancer have been recognized to be linked in another context for many years, i.e., with regards to pathologies resembling chronic lacerations or 'wounds that do not heal.' More recently, the immunology of wound healing has given us clues as to the mechanistic link between inflammation and cancer, in as much as wounds and chronic inflammation turn off local cell-mediated immune responses and switch on growth factor release as well the growth of new blood vessels - angiogenesis. Both of these are features of most types of tumours, which suggest that tumours may require an immunologically shielded milieu and a growth factor-rich environment.

It describes in detail why tumor cells can survive and spread in spite of the antitumor response of immune cells. Since immunotherapy is an attractive approach to cancer therapy, this book also provides information on the two main strategies: monoclonal antibodies and adaptive T cell immunotherapy, with a focus on recent human clinical trials. A newly added chapter also focuses on the role of Natural Killer cells in tumor progression. The book provides a state-of-the-art, comprehensive overview of immune cells in cancer and is an indispensable resource for researchers and practitioners working or lecturing in the field of cancer research and immunology.

Available in PDF, EPUB and Kindle. Book excerpt: Cytokine and Chemokine Networks in Cancer" provides a comprehensive exploration of the roles of cytokines and chemokines in cancer biology. It offers valuable insights into their diagnostic, prognostic, and therapeutic implications, making it a valuable resource for researchers, clinicians, and students interested in the field of cancer immunology and therapy. This book illustrates the importance and significance of the cytokine and chemokine signaling networks in tumor development and progression. It describes the complex networks mediated by cytokine and chemokine receptors promoting tumor cell proliferation, site-directed metastasis, and activation of angiogenic switch in tumor cells. The books also shed light on the heterogeneity of cytokines and chemokine in solid malignancies and their impact on tumor progression and therapeutic outcomes. The chapters provide current information about the types of cytokine-chemokine interactions in promoting cancer stem cell-like characteristics, epithelial to mesenchymal transition, and modulation of the tumor microenvironment. The significance of the complex interactions in cancer biology in the light of therapeutic resistance is also highlighted. The chapters also describe recent advancements in the therapeutic potential of targeting the pro-tumor cytokine and chemokine networks and limiting tumor cell metastasis. Finally, the book also provides a comprehensive yet representative description of a large number of challenges associated with targeting these vital chemokine-cytokine networks. Given its content, the book provides valuable information for researchers in the field of cancer biology and molecular medicine.

Book excerpt: Cytokines are commonly referred to as the "language of intercellular interaction" since they send signals and commands to the cells to perform certain functions. In this compilation, the authors discuss how cytokines have demonstrated different patterns of production and interaction with other cytokines; therefore, cytokines can be considered as diagnostical markers and possible targets for therapeutic measures. The authors illuminate the role of cytokines in different physiological circumstances affecting the onset, development and resolution of inflammation associated with inflammatory bowel disease. The administration of cytokines as therapeutics has proven potential in the treatment of cancer and infectious and non-infectious disease. The authors discuss cytokine profiles and their signaling in the autoimmune disease Guillain-Barre syndrome, and the potential of using this specific information for developing new therapeutic strategies. The concluding study focuses on malignant breast cancer, the most common cancer in women worldwide. In recent decades, many advances have occurred in understanding the role of cytokines in this cancer, with several studies on new diagnostic and prognostic markers and their applications as a target in treatment.
